【発明の詳細な説明】[Detailed description of the invention]

本発明は、重合性リン脂質の製造法に関する。
更に詳しくは、L−α−グリセロホスホリルコリ
ン(以下L−α−GPCと略す。)と一般式 R1CH=CHCH=CHCOOH (R1はC5〜C17のアルキル基) で表わされるアルカジエンカルボン酸(以下ジエ
ンカルボン酸を縮合剤存在下に直接縮合させ一般
式 (R1はC5〜C17のアルキル基) で表わされるL−1,2−ビス(2,4−アルカ
ジエノイル)−3−ホスホリルコリン(以下ジエ
ンレシチンと略す。)を製造する方法である。 このジエンレシチンは、例えばコンタクトレン
ズ表面に塗布した後、重合することにより、湿潤
性のあるコンタクトレンズの作成が可能であり、
又人工臓器表面へ、塗布重合することにより、生
体適合性のよい人工臓器を製造できるなどに利用
される。 また、このジエンレシチンからリポソームを調
製し、そのリポソームを重合することにより、保
存安定性の優れたリポソームの調製ができる。さ
らにこのリポソームに薬剤を封入することにより
保存安定性に優れた薬剤徐放システム担体として
利用できる等、このジエンレシチンは、生体適合
性材料として非常に有用な化合物である。 このジエンレシチンの合成法としては、
「Makromol.Chem.,182,247(1981)」で既に知
られている。しかしこの方法では、グリセリンよ
り出発し、8工程の反応を行わなければならず、
ジエンレシチンの工業的製造法としては問題があ
る。 レシチンの合成法としては、グリセリンを出発
原料とする純合成法と、天然レシチンを加水分解
し容易に得られるGPCとカルボン酸誘導体から
合成する半合成法が知られている。しかし前述の
如く純合成法は、反応工程が長く工業的製法とし
ては好ましいものではない。 一方半合成法としては(1)ピリジン類の存在下
GPCと酸塩化物を反応させる方法(2)GPCと酸無
水物をその酸のアンモニウム塩存在下で反応させ
る方法および(3)ピリジン誘導体存在下にGPCと
酸無水物を反応する方法等が知られている。 しかし、これらの半合成方法でジエンレシチン
合成を行う場合は長鎖ジエンカルボン酸を酸塩化
物、酸無水物等に誘導しなければならず、また一
方、これらの長鎖ジエンカルボン酸およびその誘
導体は、光あるいは熱で容易に重合が進行するた
め、反応収率も悪く、その精製も非常に難しい等
の問題がある。 本発明者は、長鎖ジエンカルボン酸とGPCを
直接縮合させる方法について鋭意研究を行つた結
果、天然レシチン、例えば卵黄レシチンを加水分
解して得られるL−α−GPCあるいはその塩化
カドミウム錯体と長鎖ジエンカルボン酸をクロロ
ホルム中N,N−ジアルキルピリジン触媒存在下
ジシクロヘキシルカーボジイミドと反応を行う
と、一段階でジエンレシチンを製造できることを
見い出し本発明に到つた。 また「Angew.Chem.Int.Ed.,17,522(1978)}
には、カルボン酸のエステル化について記載され
ているが、本発明とは目的物も異なり、また、触
媒量も異なる。事実本発明法のGPCとジエンカ
ルボン酸の反応においては比較例にも示したが、
この方法では目的のジエンレシチンは全く単離で
きなかつた。この原因は明らかではないが、一般
に共役不飽和カルボン酸のエステル化に対する反
応性は非常に低いことが知られている。従つて、
共役不飽和カルボン酸とジシクロヘキシルカーボ
ジイミドから生成した反応中間体は非常に活性化
されており触媒のN,N−ジアルキル−4−アミ
ノピリジンが少ないと容易にN−アシル尿素体等
に変化することが考えられ、それ故GPCとジエ
ンカルボン酸とのエステル化が進行しなかつたも
のと考えられる。 本発明に用いるGPCとしては例えば卵黄レシ
チンを加水分解して容易に得られるL−α−
GPCあるいはその塩化カドミウム錯体を用いる
ことができる。 長鎖ジエンカルボン酸としては、一般式 R1CH=CHCH=CHCOOH (R1はC5〜C17のアルキル基) で表わされるα,β,γ,δ一不飽和カルボン酸
を用いることができる。このようなジエンカルボ
ン酸として、2,4−デカジエン酸、2,4−ウ
ンデカジエン酸、2,4−トリデカジエン酸、
2,4−テトラデカジエン酸、2,4−ペンタデ
カジエン酸、2,4−ヘキサデカジエン酸、2,
4−ヘプタデカジエン酸、2,4−オクタデカジ
エン酸、2,4−ノナデカジエン酸、2,4−エ
イコサジエン酸、2,4−ヘンエイコサジエン
酸、および2,4−ドコサジエン酸の全ての幾何
異性体を挙げることができる。 ジエンカルボン酸の量は、化学量論量以上、す
なわち、GPC1モルに対しジエンカルボン酸2モ
ル以上用いればよい。 溶媒としては、乾燥したクロロホルム、好まし
くは、エタノールを含有しない乾燥クロロホルム
である。 反応系にエタノールが存在するとエタノールと
ジエンカルボン酸の反応が進行し、目的のジエン
レシチンの収率が低下する原因となり、好ましく
ない。 溶媒量は、ジエンカルボン酸が完全に溶解し、
かつ、GPCあるいはGPCの塩化カドミウム錯体
および反応中に析出する尿素が存在しても円滑な
反応液の撹拌が可能な量であればよく、ジエンカ
ルボン酸1重量部に対し溶媒10容量部以上であれ
ばよい。 ジシクロヘキシルカーボジイミド量は、ジエン
カルボン酸1モルに対し、1モル以上、好ましく
は、1,2モル以上である。 N,N−ジアルキル−4−アミノピリジンとし
ては、N,N−ジメチル−4−アミノピリジン、
N−ピペラジノ4−ピリジン等を挙ることができ
る。 N,N−ジアルキル−4−アミノピリジン量
は、反応を円滑に行うために特に重要であり、ジ
シクロヘキシルカーボジイミド1モルに対しN,
N−ジアルキル−4−アミノピリジン0.2モル以
上好ましくは0.4モル〜1モルを用いなければな
らない。 反応は、室温下、30時間程度で終了するが、ク
ロロホルムの分解およびジエンレシチン類の重合
を抑へるため、窒素雰囲気下、かつ遮光下に行う
ことが好ましい。ジエンレシチンは、上記反応混
合物を既存の方法で処理することにより単離精製
が可能である。 本発明に従へば、生体適合性材料として有用な
ジエンレシチンを天然由来のGPCと長鎖ジエン
カルボン酸から1段階で容易に製造できる。 さらに本発明により、飽和カルボン酸およびカ
ルボニル基に対して非共役は不飽和結合を有する
カルボン酸残基を有するレシチン合成も容易に行
へる。 以下実施例により本発明をさらに詳しく説明す
る。 実施例 1 2E,4E−オクタジエン酸10g、L−α−グリ
セロホスホリルコリン塩化カドミウム錯体6.3g
およびN,N−ジメチル−4−アミノピリジン
2.2gの乾燥クロロホルム200ml溶液に窒素気流中
氷冷下にジシクロヘキシルカーボジイミド8.8g
の乾燥クロロホルム20ml溶液を滴下した。 滴下終了後、5分間氷冷下で反応を行つた後、
窒素雰囲気下、室温で30時間撹拌を行つた。 反応終了後、ジパルミトイルホスホリルコリン
(DPPCと略す)を対照物質としてシリカゲル薄
層クロマトによる分析(展開液クロロホルム:メ
タノール:水=65:25:4)を行なつたところ

同様な方法により反応を行つた。反応終了後、溶
媒を留去し、残渣を「トヨパールHW40」(東洋
曹達工業(株)製商品名、展開液;クロロホルム:メ
タノール=1:4)のゲルクロマトにかけC18−
ジエンレシチン4.5gを得た。 比較例 1 N,N−ジメチル−4−アミノピリジンを0.2
gとした以外は、実施例1と同様な方法により反
応を行つた。反応終了後実施例1と同様な薄層ク
ロマト分析を行つたところジエンレシチンに相当
するスポツトは全く認められなかつた。 比較例 2 N,N−ジメチル−4−アミノピリジンを0.6
gとした以外は実施例1と同様な方法に反応を行
つた。反応終了後、薄層クロマト分析を行つたと
ころジエンレシチンに相当するスポツトが若干認
められた。その後、実施例1と同様な後処理を行
つたが、ジエンレシチンを単離することができな
かつた。 実施例 3〜5 N,N−ジメチル−4−アミノピリジンの量を
変えた以外は、実施例1と全く同様にして、反応
を行つたその結果を表1に示す。
