Abstract

PURPOSE:To discriminate bovine male and female simply and in high sensitivity by using bovine male specific DNA having a specific base sequence or a bovine female specific DNA hybridisable with the DNA as a primer and a probe. CONSTITUTION:Peripheral blood is collected from mature Holstein male and female, centrifuged separately, leukocyte fractions are treated with proteinase, RNase and phenol to extract DNAs, which are mixed with primers shown by formula I and formula II and thermostable polymerase, reacted at 60 deg.C for 2 minutes, at 80 deg.C for 2.5 minutes, further at 93 deg.C for 1.5 minute, amplified by repeating PCR operation 30 times, subjected to electrophoresis on agarose gel, a bond existing in male DNA but not in female DNA is selected and the base sequence is determined to give a bovine male specific DNA containing front part of IO-1 sequence shown by formula III or part thereof. The DNA is used as a probe to accurately discriminate bovine male and female.

【発明の詳細な説明】 【0001】 【産業上の利用分野】本発明は、ウシの雄特異的DNA
および該DNAをプローブとして用いるウシ雌雄の判別
方法に関する。 【0002】 【従来の技術】近年、胎児または胚の性別判定は、重要
な課題となっている。特に、胎児移植などの生殖生物学
分野で重要となりつつある。例えば、乳牛及び食肉業の
分野だけでも、1985年にはおよそ50,000例の
胎児移植が報告されている。胎児を母体に移植する前に
簡単にその性別の判定ができれば、雌の子孫を増すこと
が可能となり、乳牛業において極めて有利となる。また
、胎児の性別が簡単に判定できれば、乳を多く出すこと
ができるとか子供を多く生むことができるなどの望まし
い特質を持った子孫を選択することが可能となる。 【0003】性別(雄もしくは雌)が性染色体によって
決定されるということは従来より広く知られている。哺
乳動物において、Y染色体上に存在する遺伝子が雄とし
ての特徴形成の原因となるので、個々の哺乳動物の性別
は、ある特定のDNA配列をそのゲノムが含んでいるか
否かに依存することになる。従って、DNA中のY染色
体特異的遺伝子を分析することによって雌雄の判別をす
ることができる。あるいは、遺伝子的に関連した配列、
具体的にはY染色体と関連した配列を分析することによ
っても判定することができる。 【0004】従来より、DNAによる雌雄判別の方法と
しては種々の報告がある。例えばヒトの男性DNAと優
先的にハイブリダイズするDNA配列は多くの研究者に
よって同定されている(Kunkel et al, 
Science191, 1189−1190 (19
76), Bishop etal, Nature 
303, 831 (1983))。しかし、これらの
DNA配列がヒト以外の他の種、例えばウシのDNA配
列とハイブリダイズするか否かについては不明である。 一方特開昭63−500214号公報には、雄性DNA
と優先的にハイブリダイズするウシDNAプローブが記
載されている。このDNAは、胎児および胚の性別を判
定するためのハイブリダイゼーションプローブとして有
用であるといわれている。 また、ウシの亜科に属し、ことにボス属の反芻動物の胚
もしくは胎児の性別を決定するのに有用な雄特異的DN
Aが特開昭62─273000号公報に、牛、ヒツジ、
ヤギおよび他の反芻動物のY染色体特異的DNA配列に
のみハイブリダイズすることのできるDNAが特表平1
−500720号公報に開示されている。 【0005】 【発明が解決しようとする課題】しかしながら、ウシの
雌雄判別に関する前記の特開昭63−500214号公
報に記載のプローブは長すぎるという点が、また、特開
昭62−273000号公報記載のDNAでは短かすぎ
るのでPCR等には使えないといった問題点が指摘され
る。また、特表平1−500720号公報では実用性の
例が示されてない。 【0006】このように当業界では前記のようなウシの
性別判定の重要性に鑑み、簡便にかつ高感度に雌雄判別
ができるような方法の開発が望まれているものの、未だ
充分な方法は見い出されていないのが実情である。しか
して、本発明の目的は、ウシ雌雄判別を簡便かつ高感度
に行うことのできるウシの雄特異的DNAおよびそれを
プローブとして用いるウシ雌雄の判別方法を提供するこ
とにある。 【0007】 【課題を解決するための手段】本発明者らは前記課題を
解決するために鋭意検討した。その結果、ウシの雄特異
的DNAを見い出し、さらに該DNAを用いることによ
りウシ雌雄の判別を簡易かつ高感度に行うことのできる
ことを見い出し本発明を完成するに到った。 【0008】即ち、本発明の要旨は、 (1)ウシの雄特異的DNAおよび (2)前記のDNAをプローブとして用いることを特徴
とするウシ雌雄の判別方法に関する。 【0009】本発明のウシの雄特異的DNAは、具体的

のウシの雄特異的DNAは、前記のようにIO−1配列
、IO−2配列IO−3配列およびIO−4配列のそれ
ぞれ全部からなるものであってもよく、またウシ雌雄の
判別方法にプローブとして使用することができるもので
あればその一部を含有するものであってもよい。さらに
、同様にウシ雌雄の判別方法にプローブとして使用する
ことができるものであれば、該DNAとハイブリダイズ
し得るものも本発明のウシの雄特異的DNAに含まれる
。ここで、該DNAとハイブリダイズし得るものとして
は、例えばIO−1配列のDNAの一部に塩基の置換、
欠失、挿入等の変異のあるものであっても該DNAとハ
イブリダイズし得るもの等を挙げることができる。 【0014】本発明のウシの雄特異的DNAを得るには
、例えば次のような方法が挙げられる。 (1)ゲノムDNAの抽出 ウシの白血球、精巣、卵巣、肝臓などから得られた新鮮
、凍結あるいはホルマリンなどで固定した検体をプロテ
イナーゼ等の酵素処理によりタンパク質を消化した後、
フェノール抽出およびエタノール沈澱処理など常法によ
りDNAを抽出する。 【0015】(2)PCRによるゲノムDNAの増幅E
S8プローブ(Ellis ら、Theriogeno
logy 29:242, 1988 )の1−20b
p(MI304)および541−560bp(MI30
5)に相同する配列を有するオリゴヌクレオチドをDN
A合成機(例えば、ABI社製)を用いて合成し、得ら
れたオリゴヌクレオチドをプライマーとして用い、常法
によりPCRを行うことによりウシゲノムDNAの増幅
を行う。これらのプライマーの塩基配列は次のとおりで
ある。 プライマーMI304(配列番号:5):  5’−C

Rに用いるゲノムDNA量は、通常1μg程度を標準と
し、プライマーの量は通常、100pmol程度を用い
る。PCRの条件は、ディナチュレーション工程は通常
93℃で1.5分、アニーリング工程が通常60℃で2
分、イクステンション工程が通常80℃で2.5分の条
件であり、これらの工程を1サイクルとしたPCRを通
常30サイクル行なう。 【0016】(3)クローニング (2)で得られた増幅されたDNA断片は、アガロース
ゲル電気泳動にかける。電気泳動を行ったゲルより、用
いたプライマーから推定される鎖長を有するバンドを切
りだし、目的の鎖長を有するDNA断片を得る。次いで
得られたDNA断片を市販のプラスミドに挿入し、さら
に該プラスミドを大腸菌などの適切な宿主に導入する。 形質転換体の有するプラスミドの挿入DNAの鎖長を調
べ、目的の挿入DNAを有するクローンを陽性とする。 【0017】(4)制限酵素による消化(3)で得られ
た陽性クローンのプラスミドDNAをEcoRIとPs
tIにより消化することにより本発明のIO−1配列、
IO−2配列IO−3配列およびIO−4配列を得るこ
とができる。 【0018】本発明のウシ雌雄の判別方法は、これらの
本発明の雄特異的DNAのうち少なくとも1つをプロー
ブとして用いることを特徴とするものである。具体的に
は、本発明のウシの雄特異的DNAをプローブとして用
い、検体中のDNAとハイブリダイゼーションが生じる
か否かにより雌雄を判別する。即ち、ハイブリダイゼー
ションを認めればその検体は雄性であり、認めなければ
雌性と判定される。 【0019】このハイブリダイゼーションの方法は、こ
のような目的を達成することができるものであれば特に
制限されるものではない。例えば、ドットブロットハイ
ブリダイゼーションにより行うことができる。この場合
、本発明の雄特異的DNAは該DNAを保持するプラス
ミドDNAをプローブとし、ニックトランスレーション
によって放射標識して用いることができる。具体的には
、まずドットブロットフォーマットを用い、アルカリ変
性および熱変性処理を行った後、検体のDNAをニトロ
セルロースフィルターに吸着させる。該フィルターをハ
イブリダイゼーション液(5×SSPE、5×デンハル
ト溶液、0.5%v/vSDS)に75℃で1時間浸漬
することによって、プレハイブリダイゼーションを行う
。次いで、該フィルターを前記プローブとともに1晩ハ
イブリダイゼーションに付した後、該フィルターを6×
SSC液で1時間洗浄し、X線フィルムに−80℃で一
夜露光させることにより行われる。 【0020】本発明のウシ雌雄の判別方法は、本発明の
雄特異的DNAのうち少なくとも1つをプローブとして
用いればよいが、複数の雄特異的DNAを同時にプロー
ブとして用いてドットブロットハイブリダイゼーション
を行ってもよい。 【0021】 【実施例】以下、実施例により本発明についてさらに詳
しく説明するが、本発明はこれらの実施例によりなんら
限定されるものではない。 実施例 (1)DNA試料 成熟ホルスタイン牛(雌雄1頭づつ)の末梢血5mlを
遠心分離にかけた。白血球画分をプロテイナーゼK、R
Nase およびフェノールで処理し、DNAを抽出し
た。 得られたDNAを定量した後、−20℃で保存した。 【0022】(2)PCRによるウシゲノムDNAの増
幅 増幅に用いたDNAの量は、雄ウシ細胞DNAの場合は
10−6〜1μgであり、雌ウシ細胞DNAの場合は1
μgであった。ES8プローブ(Ellis ら、Th
eriogenology 29:242,1988)
の1−20bp(MI304)および541−560b
p(MI305)に相同する配列を有するプライマーを
DNA合成機(ABI社製)を用いて合成した。得られ
たプライマーはそれぞれ100pmolをPCRに用い
た。これらのプライマーの塩基配列は次のとおりである

方法に従って調製したPCR用緩衝液に、最終容量が1
00μlになるよう前記のプライマーを加えた。2.5
ユニットの耐熱性DNAポリメラーゼ(Taqポリメラ
ーゼ、シータス社製)を各試料に加えた後、プログラム
式熱コントローラー(MJリサーチ社製)を用いてPC
R操作(60℃で2分間反応させ、80℃で2.5分間
反応させ、さらに93℃で1.5分間反応させる操作)
を30回繰り返した。PCRの終了後、反応生成物を4
%アガロースゲル(シーケムME、FMC社製)上で電
気泳動にかけ、臭化エチジウム染色後、紫外線照射下で
現像した。 【0023】このようにしてDNA断片を電気泳動で分
画したものを図1に示した。即ち、図1は反応生成物の
1/10を4%アガロースゲル上で電気泳動にかけ、臭
化エチジウム染色後、紫外線照射下で現像した結果であ
る。レーン1〜7はそれぞれ1、10−1、10−2、
10−3、10−4、10−5および10−6μgのウ
シ雄DNAから増幅された生成物の泳動結果を示したも
のである。レーン8はウシ雌DNA1μgから増幅され
た生成物についての泳動結果である。図中で矢印を付し
た4つの未知のバンドが雄DNA検体で見られた。 【0024】このバンドを分析したところ、約100b
p、130bp、180bpおよび230bpの位置に
存在していた。また他の位置にも残存するプライマーに
由来するバンドが出現した。これらのDNA断片は、1
0−6μgのDNA試料のPCRでも確認された。一方
、1μgの雌DNA試料をPCRで分析したが、このよ
うなDNA断片は見られなかった。 【0025】(3)PCR生成物のクローニングと配列
決定 PCR生成物をプラスミドDNA  pUC119のS
maIサイトに結合させた後、大腸菌MV1184にク
ローン化した(図2参照、aはPstIサイトを、bは
SmaIサイトを、cは挿入DNAを、そしてdはEc
oRIサイトを示す)。クローニング終了後、プラスミ
ドDNAをEcoRIとPstIにより制限酵素による
分析に付した後、サンガーらのジデオキシ法によって配
列を決定した。 【0026】その結果、制限酵素による分析によりPC
R生成物を4種に分けることができた(図3)。図3は
、4種のPCR生成物をもつプラスミドをEcoRIと
PstIで同時に消化し、その各種の一部を4%アガロ
ースゲル上で電気泳動にかけ、臭化エチジウム染色した
結果を示すものである。図中の*はマーカーである。 これらのDNA断片についてジデオキシ法で決定した塩
基配列を図4に示した。これら4種のDNA断片のサイ
ズは、それぞれ92bp(I0−1)、142bp(I
0−2)、170bp(I0−3)、および224bp
(I0−4)であった。これらのサイズは、電気泳動度
に基づく推定値とほぼ同じであった。 【0027】これら4種のDNA断片の塩基配列はES
5やES8の塩基配列と一致していなかった。ES8と
4種のDNA断片の塩基配列の一致度を調べたところ、
I0−1断片では全配列の65%、I0−2断片では5
1%、I0−3断片では57%、I0−4断片では84
%のホモロジーがみられた。また、図4の塩基配列から
も明らかなようにこれら4種のDNA断片はいずれも反
復CA配列を有していた。 【0028】(4)DNA断片の雄特異性のドットブロ
ット分析 前記のようにして配列の決定された4種のDNA断片を
保持するプラスミドDNAをプローブとし、ニックトラ
ンスレーションによって放射標識した後、以下のハイブ
リダイゼーションによる分析を行った。即ち、まずドッ
トブロットフォーマットを用い、アルカリ変性および熱
変性処理を行った後、ウシゲノムDNAをニトロセルロ
ースフィルターに吸着させた。該フィルターをハイブリ
ダイゼーション液(5×SSPE、5×デンハルト溶液
、0.5%v/vSDS)に75℃で1時間浸漬するこ
とによって、プレハイブリダイゼーションを行った。 次いで、該フィルターを前記プローブとともに1晩ハイ
ブリダイゼーションに付した。続いて、該フィルターを
6×SSC液で1時間洗浄した後、X線フィルム(コニ
カ社製)に−80℃で一夜露光させた。 【0029】図5はこのようにして得られたドットブロ
ットハイブリダイゼーション分析の結果を示す図である
。即ち、ウシ雄およびウシ雌DNA1μgを4種のニト
ロセルロースフィルターにアプライし、32Pで標識し
た4種のDNA断片をそれぞれハイブリダイズさせた。 図中、1はI0−1断片を、2はI0−2断片を、3は
I0−3断片を、4はI0−4断片をそれぞれハイブリ
ダイズさせた結果である。図5から明らかなようにこれ
ら4種のDNA断片を雌雄ウシゲノムDNAのそれぞれ
とハイブリダイズさせたところ、4種の断片はいずれも
雄ゲノムDNAのみに対して陽性反応を示し、これらの
DNA断片は雄特異性を有することが明らかとなった。 【0030】 【発明の効果】本発明において、ES8から得たプライ
マーを用いたPCRでは、予想されたES8の増幅は起
こらず、4種の本発明のDNA断片(92bp、142
bp、170bp、224bp)が得られた。これらの
増幅DNA断片の塩基配列は、ES8の塩基配列と部分
的に一致が見られたものの、ES5あるいはES8とも
完全に同一のものではなかった。ES8はウシY染色体
上に存在する約600コピーにのぼる反復配列の1単位
を成していることが知られているので(Bondiol
iら、Theriogenology 31:95−1
04, 1989)、多少異なる部分もあるが類似して
いる塩基配列を有する一群に含まれるものと思われる。 したがって、プライマーMI304およびMI305で
増幅させた本発明の4種のDNA断片は前記の類似する
塩基配列に由来するものである可能性が高いと思われる
。 【0031】また、4種のDNA断片の塩基配列はいず
れも反復CA配列を含んでいた。この反復配列はES8
の塩基配列のかなりの部分を占めていた。この点につい
て、Kashiら(Genomics. 7:31−3
6 1990)は、(TG)10塩基配列を有するプロ
ーブを制限酵素(HaeIIIまたはMboI)で消化
した後で雌雄ウシゲノムDNAにハイブリダイズさせる
と、数kb〜数10kbの位置に雄特異性バンドが検出
されたと報告している。これらの知見から、反復CA配
列はウシY染色体に特有の塩基配列であることが推察さ
れる。このようにこれらの4種のDNA断片は雄特異性
であり、これらをプライマー及びプローブとして用いる
ことにより簡易にかつ高感度にウシ雌雄を判別すること
