Description
本発明は有機酸生産性微生物の培養方法に関
し、さらに詳しくは、有機酸生産性微生物を特定
の波長領域の光線の照射下に培養することによつ
て該有機酸の生産性を向上させる有機酸生産性微
生物の培養方法に関する。
本発明者は各種有用植物の成育、有用植物に対
する病害糸状菌類の繁殖の防除、藻類植物の培養
等において、光質条件が如何に影響するかを研究
している過程において、全く偶然的に、有機酸生
産性微生物を少くとも280nmおよびそれ以上の
光線を実質的に含有する特定の光線の積極的照射
下に培養すると、有機酸の生産性が大いに向上す
ることを見い出し本発明を完成するに至つた。
かくして、本発明に従えば、有機酸生産性微生
物を光線を照射しつつ、培養し、有機酸を生産す
る方法において、
前記微生物が、有機酸を産する能力を有する
細菌類、放線菌類及び菌類から選ばれた微生物
であり、
前記光線の照射開始時期が前記培養系内の前
記微生物の対数増殖期以降であり、かつ、
前記光線が少なくとも280nm及びそれ以上
の波長域の光線を実質的に含有する光線であ
る、
ことを特徴とする有機酸生産性微生物の培養方
法、が提供される。
本発明において「有機酸」とは乳酸、クエン
酸、α−ケトグルタール酸、コハク酸、フマール
酸、l−リンゴ酸、イタコン酸、酢酸、プロピオ
ン酸、酪酸、グルコール酸、イソ吉草酸、イソク
エン酸、D−アラボン酸、D−キシロン酸、D−
グルコン酸、酒石酸、2−ケトグルコン酸、5−
ケトグルコン酸、2,5−ジケトグルコン酸、D
−アラボアスコルビン酸、コージ酸、D−マンノ
ン酸、D−ガラクトン酸、2−ケトグロン酸、ク
ミン酸、トルイル酸、2,3−ジヒドロキシトル
イル酸、桂皮酸、テトラデカン−1,14−ジカル
ボン酸、アロイソクエン酸、及びこうじ酸等が包
含され、中でも、乳酸、クエン酸、α−ケトグル
タール酸、コハク酸、l−リンゴ酸、フマール
酸、イタコン酸、2,5−ジケトグルコン酸、酒
石酸、ピルビン酸、2−ケトグロン酸、酢酸、D
−グルコン酸、2−ケトグルコン酸、5−ケトグ
ルコン酸、及びこうじ酸が好ましく、特に乳酸、
クエン酸、α−ケトグルタール酸、コハク酸、l
−リンゴ酸、フマール酸、イタコン酸、2,5−
ジケトグルコン酸、酒石酸、ピルビン酸、2−ケ
トグロン酸及び5−ケトグルコン酸が好適であ
る。
本明細書において「有機酸生産性微生物」と
は、有機酸を微生物菌体外に代謝生産物として生
産する能力を有する微生物をいい、かかる微生物
には、菌類、放線菌類、細菌類及び酵母類が包含
される。
本発明の方法は、本発明者らの経験及び後述す
る実施例の結果から明らかなように、一般的に言
つて、どのような種類の微生物に対しても適用す
ることができ、それによつて大なり小なり有機酸
の生産性の向上効果を期待することができるが、
中でも菌類、細菌類、放線菌類及び酵母類の微生
物に対してその効果が著しく特に菌類及び細菌類
に適し、最も細菌類に好適である。
本発明の方法を適用することができる代表的な
微生物を例示すれば次のとおりである。なお下記
の微生物の例示においては、微生物の名称を和名
で属(Genus)及び種(Species)を示し、その
次に原名を( )内に示す。

従来、有機酸の発酵による工業的生産は、通
常、終始暗黒のタンク内で行われており、光線の
照射を実質的に回避した条件下に行なわれてい
る。本発明は、かかる従来の有機酸の発酵的生産
法とは対照的に、上記特定の光線を含まない光線
を積極的に照射しながら微生物の培養を行なうも
のであり、この点、本発明の方法は従来の発酵法
とは本質的に相違するものである。
上記特定光線の照射は有機酸生産工程でいう、
前培養および、本培養いづれの場合も適用される
が、本培養に行うことによりその効果が大とな
る。
照射しうる光線は、少なくとも280nmおよび
それ以上の波長域の光線を実質的に含有する限
り、特に制限はなく、280〜800nmの波長域光を
実質的に含有する光線であれば、人工光線のみな
らず、自然光線も使用することができる。
しかして、人工光線及び/又は自然光線を用い
る場合には、必要に応じ光フイルターを用い、少
なくとも280nmおよびそれ以上の波長域光を実
質的に含有する光線の照射光量が100000μw/cm2
以下、好ましくは10000μw/cm2以下、更に好まし
くは1000μw/cm2に抑制された人工光線の照射下
に培養することが好ましい。
本発明の方法に従い、微生物の培養系に照射さ
れる前記光線の強度は、厳密に制限されるもので
はなく、培養すべき微生物の種類やその他の培養
条件等により異なり、個々の場合における最適の
照射条件は当業者であれば小規模の実験を行なう
ことにより容易に決定しうるが、一般には、
400nm〜800nmの範囲の波長の可視光線領域光
の光量が100000μw/cm2以下、好ましくは10000〜
1μw/cm2、さらに好ましくは1000〜50μw/cm2の
範囲内に調節された光線を照射するのが有利であ
る。また、280nm〜400nmの波長の紫外線領域
の光線はあまり強くない方が好ましく、通常該波
長範囲の紫外線の強度は一般に80000μw/cm2以
下、好ましくは10000μw/cm2以下、さらに好まし
くは1000〜20μw/cm2とするのが望ましい。
また、本発明の方法に従い、微生物の培養系に
対して前記特定の光線を積極的に照射する具体的
方法としては、例えば、実質的に外光線から密閉
された系内(タンク内)において、少なくとも
280nmおよびそれ以上の波長域の光線を実質的
に含有する光線、好ましくは、280〜800nmの波
長域光を実質的に含有する人工光線(この場合、
人工光線源それ自体がかかる光質特性の光を発す
るものであつてもよく、或いは人工光線源を適当
なフイルターで覆うことにより照射される光が上
記のような光質特性をもつようにしてもよい)を
照射する方法;太陽又は自然光線の照射下に、少
なくとも280nmおよびそれ以上の波長域の光線
を実質的に含有する光線、好ましくは280〜800n
mの波長域光を実質的に含有する光を透過する、
透明な無色乃至有色の有機質又は無機質の被覆材
(例えば、紫外線吸収剤を配合した合成樹脂フイ
ルム)により被覆した条件下に培養を行なう方
法;並びに上記両方法の組合わせ等が考えられ
る。
本発明の方法に従がい上記特定波長域光を実質
的に含有する光線の有機酸生産性微生物に対する
照射開始時期は、該微生物培養系内に於いて、該
微生物の対数増殖期が好ましい。該微生物は、培
養系内に植菌されると、直ちに急激なる増殖は行
なわず、誘導期を経過した後に、急速に増殖を行
う対数増殖期となる。対数増殖期以降の、該微生
物は、系内の栄養源の枯渇に伴ない定常期を経
て、減数、死滅する。本発明によれば、上記特定
波長域光を実質的に含有する光線の照射開始は、
対数増殖期、好ましくは、対数増殖期の前期さら
に好ましくは、中期である。また、上記特定の波
長域光を実質的に含有する光線を照射する期間と
しては、前期の照射開始時点より発酵が終了する
時点まで、あるいは発酵がある過程に達する時点
まで等あげられるが、好ましくは発酵が終了する
時点までである。
なお、照射形式としては、続けて照射を行う連
続照射法、照射と暗黒とを交互にくり返す間欠照
射法、およびこれらの組み合せ法等があり、適宜
選択することが出来る。
本発明でいう[Xnmおそびそれ以上の波長域
の光線を実質的に含有する」とは照射する全光線
量のうち、Xnm未満の波長域の紫外線が完全に
存在しないことのみならず、該紫外線が本発明の
培養に悪影響を及ぼさない程度の範囲で少量含有
していても支障はないことを意味する。
また、本発明でいう「Y〜Znmの波長域光を
実質的に含有する」とは、照射する全光線量のう
ち、Y〜Znmの波長域光が100%の場合のみなら
ず、60%以上、好ましくは、80%以上更に好まし
くは90%以上である。
本発明の方法に従う有機酸生産性微生物の培養
は、上記特定の光線の照射下に行なうという条件
を除けば、従来から行なわれている条件と全く同
様の条件下に行なうことができる。例えば、有機
酸生産性微生物を適当な栄養培地中で液体培養又
は固体培養することにより行うことができる。そ
の際の培地の栄養源、窒素源及び無機塩類等は、
使用する微生物や培養手段に応じて適宜変更選択
されるが、微生物の培養に敵常用いられるものが
広く使用される。炭素源としては、同化可能な炭
素化合物であればよく、例えばブドウ糖、シヨ
糖、乳糖、麦芽糖、澱粉、デキストリン、糖密、
グリセリン、炭化水素、エチルアルコール、メチ
ルアルコール、酢酸などが使用される。また、窒
素源としては、使用可能な窒素化合物であればよ
く、例えばコーン・スチーブ・リカー、大豆粉、
綿実油、小麦グルテン、ペプトン、肉エキス、酵
母エキス、酵母、カゼイン加水分解物、大豆タン
パク加水分解物、アンモニウム塩、硝酸塩、など
が使用される。その他無機塩としては例えば、リ
ン酸塩、マグネシウム、カルシウム、カリウム、
ナトリウム、亜鉛、鉄、マンガンなどの塩類が、
さらに必要に応じてアミノ酸類、ビタミン類、有
機酸類、核酸関連物質、発酵原料となる前駆体な
どが使用される。
培養温度および培養時間は、使用する微生物に
よつても多少異なるものであつて、その微生物が
充分発育し得る範囲内で適宜変更することができ
るが一般に、例えば細菌の場合は約25〜37℃程
度、糸状菌、酵母菌、担子菌の場合は約20〜26℃
程度、放線菌類の場合は約26〜32℃程度で培養す
ることがよい。
更に具体的な培養条件は、個々の微生物によつ
て異なるが、例えば、山口辰良、山口和夫著、最
新応用微生物学入門、(昭和52年7月1日、1版

いて行うことができる。
更に詳しくは、クエン酸の生産では、例えば特
許公告第50−16879号、同50−16436号、同51−
45673号、同51−21073号、同51−6751号、同51−
34474号、同51−44195号、同53−7514号、同53−
8797号、同53−19676号等の明細書、及びL−リ
ンゴ酸は、例えば特許公報第53−24515号同53−
31955号及び同53−43590号等開示された培養条件
を用いて行うことができる。
本発明において、上記光質条件下で培養を行う
際に使用する資材としては、上記の光線透過特性
を有するものであれば、その材質等は特に制限さ
れるものではなく、どのようなタイプの被覆材で
も使用することができる。そしてかかる資材は通
常無機質又は有機質のフイルム、板、その他の成
形体から成ることができる。しかして、例えば無
機質フイルム又は板としては、典型的には染料又
は顔料(例:エメラルドグリーン)を配合したガ
ラス板、一般的な紫外線吸収剤を含有する合成樹
脂膜を塗布又は積層したガラス板およびガラスフ
イルター等が挙げられ、また、有機質フイルム又
は板としては、特に紫外線吸収剤を塗布又は含有
せしめた合成樹脂フイルム又は板が好適である。
この成形に使用しうる樹脂としては、後述する
熱可塑性樹脂の他、例えば、メラミン樹脂、フエ
ノール樹脂、エポキシ樹脂、シリコーン樹脂、尿
素樹脂、アルキツド樹脂、アリルフタレート樹脂
等の熱硬化性樹脂もまた用いることができる。
本発明に使用し得る透明フイルム又は板は、例
えば通常のフイルム形成性熱可塑性樹脂に適当な
紫外線吸収剤を配合し、フイルム又は板に成形す
ることにより製造することができる。
使用し得るフイルム成形性熱可塑性合成樹脂と
しては、例えばポリ塩化ビニル、ポリ塩化ビニリ
デン、ポリエチレン、ポリプロピレン、ポリスチ
レン、ポリエステル、ポリアミド、ポリカーボネ
ート、ポリメチルメタクリレート、ポリアクリレ
ート、ポリ酢酸ビニル、ポリビニルアルコール、
含フツ素樹脂、セルロース系樹脂、ABS樹脂等、
又はこれら重合体を主体(好ましくは50重量%以
上)とする共重合体もしくはブレンド物が包含さ
れ、特に耐光性、強度、光線透過性の理由からポ
リ塩化ビニル、ポリエチレン、ポリプロピレン、
ポリエステル、含フツソ系樹脂、セルロース系樹
脂及びポリカーボネートが好適である。
本発明に使用し得る人工光源として、上記した
特定波長域の光線を放射する光源であれば、いづ
れものでも使用できる。そして、かかる光源とし
ては例えば、螢光灯、水銀灯、陽光ランプ、一般
電球、投光用電球、特殊電球(東芝ランプカタロ
グより、東芝電材株式会社)等がある。更に具体
的には、螢光灯〔( )内は、商品名を示す。〕と
して、日照灯(FL40SW−E/M、東芝)、プラ
ントルクス(FL40B/NL、東芝)、フイツシユ
ルクス(FL40SBRF/NL、東芝)、青色
(FL40B/NL、東芝)、青白色(FL40BW/NL、
東芝)、白色(FL40SD/NL、東芝)、昼光色
(FL40SD/NL、東芝)、デラツクス(FL40SW

以上述べた本発明の方法に従えば、醗酵法によ
る有機酸の生産において、特定の光質条件下に微
生物を培養することにより、有機酸の生産が大い
に促進され、医薬、食品等の分野に資する所極め
て甚大である。
次に実施例を挙げて、本発明をさらに説明す
る。
実施例1、比較例及び17
マルト・エキストラクト・ブロス(Difco社
製)溶液100mlを500ml三角フラスコに分注し、加
圧減菌後、あらかじめマルツ・エキストラクト・
アガー(Difco社製)スラントに植継いでおいた
L−リンゴ酸生産性微生物キヤンデイダ・プルン
プテイー(Candida brumptii IFO0731)を1白
金耳植菌し、培養温度30℃、振とう回転数200
回/分、暗黒条件下で36時間前培養した。
予め、表−1から成る水溶液を調製しておき、
その水溶液20mlをハリオガラス製の500ml坂口フ
ラスコに分注し、110℃、10分間減菌後、上記前
培養菌体浮遊液2mlを植菌し、培養温度30℃、振
とう回転数200回/分、暗黒条件下で、本培養を
行つた。

実施例15、比較例15
乳酸生産性微生物ラクトバチルス・デルブルキ
(Lactobacillus delbiu¨ckii IFO3202)を少量の
CaCO3を添加したグルコース1.5%、酵母エキス
0.8%、寒天1.5%からなる培地(PH6.8)に37℃で
2日間穿刺培養し、その1白金耳を表−22の液体
培地100mlに接種し、37℃で18時間前培養した。
無菌的に遠心分離(4000rpm、15分)により集
菌し、滅菌生理食塩水で洗浄したのち、100mlの
滅菌食塩水に浮遊した。予め、表−23から成る水
溶液を調製しておき、その水溶液100mlをパイレ
ツクスガラス製の500ml三角フラスコに分注し、
滅菌・冷却後、上記前培養菌体浮遊液5mlを植菌
し、別殺菌しておいてCaCO35gを添加し、7日
間静置培養した。但し、光照射区では、本培養開
始より18時間目に、表−2、図−1、及び図−2
に示した280〜360nm及び300〜800nmの波長域光
を放射する螢光灯下に移し、培養を継続した。な
お培養中は時々フラスコを振とうした。その培養

からなる斜面寒天培地に培養した5−ケトグルコ
ン酸生産性微生物グルコノバクター・オキシダン
ス・亜種サブオキシダンス(アセトバクター・サ
ブオキシダンス)〔Gluconobacter oxydans
subsp.suboxydans(Acetobacter suboxydans)
ATCC23773〕をソルビトール5%、酵母エキス
1%の水溶液5mlに一白金耳植菌し、27℃で48時
間前培養した。
これを更に表−25からなる水溶液50mlに無菌的
に加えて、30℃で48時間振とう培養した。
予め、表−26から成る水溶液を調製しておき、
その水溶液50mlをハリオガラス製の500ml坂口フ
ラスコに分注し、滅菌後、上記前培養菌体浮遊液
1mlを植菌し、実施例1、比較例1と同様の方法
で光照射による培養を行なつた。但し、本培養期
間は40時間であり、光照射開始は菌体の増殖が対
数増殖期の中期に達した時期、すなわち本培養開
始後14時間目に行なつた。

養し、有機酸を生産する方法において、 前記微生物が、有機酸を産する能力を有する
細菌類、放線菌類及び菌類から選ばれた微生物
であり、 前記光線の照射開始時期が前記培養系内の前
記微生物の対数増殖期以降であり、かつ、 前記光線が少なくとも280nmおよびそれ以
上の波長域の光線を実質的に含有する光線であ
る、 ことを特徴とする有機酸生産性微生物の培養方
法。 2 該培養を少なくとも280〜800nmの波長域光
を実質的に含有する光線の照射下に行なう特許請
求の範囲第1項記載の方法。 3 該光線の照射強度が100000〜1μW/cm2の範
囲である特許請求の範囲第1〜2項いずれか記載
の方法。 4 該光線が、人工光線及び/又は自然光線であ
る特許請求の範囲第1〜3項いずれか記載の方
法。 5 該有機酸が、乳酸、クエン酸、α−ケトグル
タール酸、コハク酸、l−リンゴ酸、フマール
酸、イタコン酸、2,5−ジケトグルコン酸、酒
石酸、ピルビン酸、2−ケトグロン酸、及び5−
ケトグルコン酸である特許請求の範囲第1〜4項
いずれか記載の方法。 6 該微生物が、アセトバクター
