Description
本発明は、7−デアザプリン誘導体およびその
製造法に関する。
本発明の化合物と同一の骨格を有する天然の6
−オキソ修飾塩基類(例、Q塩基、PreQ1塩基)
は、特定のtRNA(tRNATyr、tRNAHis、tRNAAsp
およびtRNAAsn)の構成成分として、広く生物界
に分布しており、上記tRNAのアンチコドン第一
字目に位置し、直接mRNAからの遺伝情報を認
識し蛋白質合成に関与する重要な生物学的意義を
有している。
特に最近の生化学的基礎研究の進歩にともな
い、tRNAの構造や生命現象における役割の解明
が急速に進み、がん細胞と正常細胞との間におけ
るtRNAの差異も明らかになつてきた。その1つ
は、がん細胞ではQ塩基のtRNA前駆体への取り
込みが完全ではなく、常にQ欠陥tRNAが存在す
る点である。又、外部よりQ塩基を与えることに
より、Q欠損tRNAはQ塩基を所定の位置(アン
チコドンの第一字目)に取り込み正常なtRNAに
戻ること、さらにQ塩基の取り込みは一般に正常
細胞には認められずがん細胞に特異的であること
が観察されている〔西村 〓、代謝、Vol17、臨
時増刊号「癌80」p127〜136(1980)〕。
そこで、本発明者らは、これら6−オキソ修飾
塩基類の誘導体について種々検索したところ、該
塩基類の6位をチオ体にした化合物は新規化合物
であり、また優れた抗腫瘍作用および抗菌作用を
有することを見い出し、これに基づいてさらに研
究した結果、本発明を完成した。
本発明は、

シル、イソヘキシル、ヘプチル、オクチル、ノニ
ル、デシル基などがあげられ、なかでも炭素数1
ないし6程度のアルキル基が好都合に用いられ
る。α位がメチレン基であるアルケニル基として
は、炭素数3ないし13程度の、たとえばアリル
(2−プロペニル)、2−ブテニル、2−ペンテニ
ル、2−ヘキセニル、4−プロピル−2−ペンチ
ニル、シンナミル、2−ノニル−2−ブテニル基
などがあげられ、なかでも炭素数3ないし9程度
のアルケニル基が好都合に用いられる。これらの
アルキル基およびアルケニル基はα位以外の任意
の位置に1ないし3個の置換基を有していてもよ
く、かかる置換基としては、炭素数1ないし4程
度のアルキル基(例、メチル、エチル、プロピ

単離してもよいし単離せずに直接化合物()と
の置換反応に供してもよい。またR1がアシル基
の場合、必要ならば反応液をそのまま約70ないし
100℃で加熱しつづけてR1の脱アシル化反応を併
せて行なうか、または自体公知の方法により加水
分解(例、酸加水分解、アルカリ加水分解、アン
モニア分解)することにより脱アシル化すること
ができる。化合物()と化合物()との置換
反応における反応溶媒としては、たとえば水、メ
タロール、エタノール、プロパノール、ブタノー
ル、ペンタノール、テトラヒドロフラン、ジオキ
サン、アセトニトリル、ピリジン、ジメチルホル
ムアミド、ジメチルスルホキシド、スルホランま
たはそれらの適宜の混合溶媒が使用される。化合
物()または化合物()が塩の形で使用され
る場合には、塩基(例、水酸化ナトリウム、水酸
化カリウム、水酸化リチウム、水酸化バリウム、
トリエチルアミン、N−メチルモルホリン)或い
は塩(例、塩化ナトリウム、塩化カリウム、塩化
カルシウム、炭酸ナトリウム、炭酸カリウム、炭
酸水素ナトリウム)で至適PH(通常、PH約5ない
し13)に調整することにより、有利に目的物
()を製造することができる。
R4、R5で示される炭化水素残基が有するこれ
らの置換基のうち、例えば、カルボキシ基、水酸
基、アミノ基あるいはアルキルアミノ基のような
場合、必要とあれば、自体公知の方法〔J.F.W.
McOmie、Protective Groups in Organic
Chemistry Plenum Press、London and New
York(1973).〕により保護された、または、既に
保護されている置換基(例、アルカノイルオキシ
基、アルコキシカルボニル基、アルカノイルアミ
ド基)を有する原料化合物()を用いて、もう
一方の原料化合物()との置換反応を行い、化
合物()としたのちに保護基の脱離反応を行つ
てもよい。
該保護基の脱離反応は通常行なわれる保護基脱
離反応に付すことにより行なわれる。たとえば酸
(例、塩酸、臭酸、硫酸、硝酸、リン酸、酢酸、
トリフルオロ酢酸、三フツ化ホウ素、三臭化ホウ
素、臭化水素−酢酸)または塩基(例、水酸化リ
チウム、水酸化ナトリウム、水酸化カリウム、水
酸化バリウム、炭酸カリウム、炭酸ナトリウム、
アンモニア水、メチルアミン、ジメチルアミン、
エチルアミン、ジエチルアミン、トリメチルアミ
ン、トリエチルアミン)を添加して、適当な溶媒
(例、水、メタノール、エタノール、プロパノー
ル、ブタノール、ジオキサン、テトラヒドロフラ
ン、メチルエーテル、エチルエーテル、ジクロロ
メタン、クロロホルム、四塩化炭素、ベンゼン、
トルエン、キシレンまたはこれらの適宜な混合溶
媒)中、約−50ないし100℃、好ましくは−10な
いし50℃の温度で、約30分間ないし100時間、好
ましくは約1ないし20時間反応させるか、または
適当な接触還元の触媒(例、パラジウム、白金、
ロジウム、ルテニウム、ニツケル)を添加して、
適当な溶媒(例、水、メタノール、エタノール、
プロパノール、ブタノール、ジオキサン、テトラ
ヒドロフラン、メチルエーテル、エチルエーテ
ル、ベンゼン、トルエン、キシレン、酢酸エチ
ル、酢酸メチル、酢酸またはこれらの適宜な混合
溶媒)中、約10ないし50℃の温度で約1ないし
100時間接触還元することにより行なわれる。
本発明方法により製造された7−デアザプリン
誘導体()は、通常の分離精製手段、たとえ
ば、濃縮、溶媒抽出、クロマトグラフイー、再結
晶などにより、反応混合物から単離することが出
来る。また、化合物()が遊離形で得られたと
きは、常法により、薬学的に許容される塩の形に
変換してもよい。
化合物()の塩としては、たとえば塩酸、硫
酸、硝酸、リン酸、ホウ酸などの鉱酸塩、シユウ
酸、酒石酸、酢酸、トリフルオロ酢酸、メタンス
ルホン酸、ベンゼンスルホン酸、p−トルエンス
ルホン酸、カンフア−スルホン酸などとの有機酸
塩、臭化メチル、ヨウ化メチル、メタンスルホン
酸メチルエステル、ベンゼンスルホン酸メチルエ
ステル、p−トルエンスルホン酸エステルなどと
の四級塩があげられる。
化合物()が四級塩以外の塩の形で得られる
場合、所望によりこれを遊離形にすることもでき
る。この場合は、化合物()の塩を通常用いら
れる方法、たとえば、中和反応や陰イオン交換ク
ロマトグラフイーなどに付すことにより容易に実
施し得る。
このようにして得られた化合物()またはそ
の塩は、in vitroにおけるL5178Y培養細胞の増
殖およびin vivoにおけるMeth.A、Sarcoma180
などの増殖をそれぞれ抑制するので、抗腫瘍作用
を有する。また、化合物()またはその塩は、
マウスに200mg/Kgとなる量を腹腔内投与しても、
死亡例を認めない。したがつて、化合物()ま
たはその塩は、温血動物とりわけ哺乳動物(例、
マウス、ラツト、ネコ、犬、ウサギなど)の腫瘍
の治療を目的として、抗腫瘍剤として用いること
ができる。
抗腫瘍剤として用いる場合には、そのもの、あ
るいは通常用いられる方法により薬理的に許容さ
れうる担体、賦形剤、稀釈剤などを使用して、た
とえば、粉末、顆粒、錠剤、カプセル剤、坐剤、
注射剤などの形態で経口的または非経口的に投与
し得る。投与量は、対象動物、疾患、症状、化合
物の種類、投与経路などにより異なるが、経口投
与の場合は化合物()として1日当り約10ない
し200mg/Kg体重であり、非経口投与の場合は1
日当り約10ないし100mg/Kg体重である。
さらに、化合物()またはその塩は種々のウ
イルスおよび微生物に対して抗ウイルス作用およ
び抗菌作用を有し、上述のように毒性が低いの
で、温血動物とりわけ哺乳動物(例、マウス、ラ
ツト、ネコ、犬、ウサギ、人)のウイルスおよび
細菌感染症の予防、治療を目的とする抗ウイルス
剤、抗菌剤、消毒剤として用いることができる。
化合物()またはその塩を殺菌剤、消毒剤と
して使用する場合には、たとえば化合物()ま
たはその塩を約0.5ないし500mg/mlの濃度で水、
等張のブドウ糖溶液、リンゲル液の様な水溶液又
は植物性(例、木綿種子、ピーナツツ、コーン、
ごま)脂肪油の様な非水溶液中に含有する液剤と
し、これを哺乳動物の手、足、耳などに塗布する
ことにより、投与部位の細菌、消毒に用いること
ができる。
また、化合物()またはその塩を約0.5ない
し500mgを乳糖、澱粉、タルク等の賦形剤を含む
錠剤として経口的に該哺乳動物のウイルス感染
症、細菌感染症の予防、治療に用いることができ
る。この場合の投与量は、化合物()として一
日約10ないし200mg/Kg体重となる量である。
本発明方法において原料として用いられる化合
物()は、たとえば以下に示す反応工程により
製造することができる。
工程Aにおいては、化合物()と塩素化剤
(例、チオニルクロリド、オキシ塩化リン、五塩
化リン、オキザリルクロリド、フタロイルクロリ
ドとを、化合物()/塩素化剤=1ないし1/
100(モル比)程度で、それら自体又は適当な反応
溶媒を用いて、約50ないし150℃、約30分ないし
10時間反応することにより反応物()へ導びく
ことができる。上記反応において用いられる溶媒
としては、たとえばベンゼン、トルエン、キシレ
ン、メチレンクロリド、クロロホルム、四塩化炭
素、1,2−ジクロロエタン、1,1,2−トリ
クロロエタン、1,1,2,2−テトラクロロエ
タン、アセトニトリルなどが挙げられる。上記反
応の際、ジメチルアニリン、ジエチルアニリン、
トリエチルアミン、ジメチルホルムアミド等を添
加すると、緩和な条件で反応を進行させることも
出来る。
工程Bにおける硫化反応は、化合物()と硫
化剤(例、硫化水素、ナトリウムスルフイド、チ
オ尿素)とを、化合物()/硫化剤=1ないし
1/20(モル比)程度で、反応に関与しない溶媒
を用い通常約20ないし150℃、約30分間ないし1
日間反応することにより行うことが出来る。反応
溶媒としては、水、アルコール類(例、メタノー
ル、エタノール、プロパノール、ブタノール、
sec−ブタノール、t−ブタノール、エチレング
リコール、メトキシエタノール、エトキシエタノ
ール)、エーテル類(ジメチルエーテル、ジエチ
ルエーテル、テトラヒドロフラン、ジオキサン、
モノグライム、ジグライム)、ハロゲン化炭化水
素(例、ジクロロメタン、クロロホルム、四塩化
炭素)、アセトニトリル、ジメチルホルムアミド、
又はそれ等の適宜の混合溶媒が使用される。
化合物()は、化合物()と化合物()
とをホルムアルデヒド類の存在下に縮合させるこ
とにより製造され得る(工程C)。
化合物()および化合物()は塩の形で使
用してもよく、かかる塩としては、前記した化合
物()の鉱酸塩または有機酸塩と同様のものが
あげられる。
ホルムアルデヒド類としては、マンニツヒ
(Mannich)反応時にホルムアルデヒドと等価の
作用を有する試薬、たとえばホルムアルデヒド、
パラホルムアルデヒド、ホルマリン、メチラー
ル、エチラール、ピペリジノメチルフタルイミ
ド、ヘキサメチレンテトラミンなどがあげられ
る。
本工程Cの反応においては、化合物()と化
合物()とを()/()=1ないし50(モル
比)程度でそれら自体又は適当な反応溶媒を用い
て約0℃からその反応溶媒の沸点、好ましくは約
20ないし100℃の範囲の反応温度で約10分間から
48時間程度反応させた後、酸で処理することによ
り、化合物()を得ることができる。なお、反
応溶媒としては、水、メタノール、エタノール、
プロパノール、ブタノール、ペンタノール、テト
ラヒドロフラン、ジオキサン、アセトニトリル、
ピリジン、ジメチルホルムアミド、ジメチルスル
ホキシド、スルホラン又はそれらの適宜の混合物
が使用される。反応溶液のPHを酸(例、塩酸、硫
酸、リン酸、ホウ酸、酢酸、シユウ酸、酒石酸、
トリフルオロ酢酸、メタンスルホン酸、ベンゼン
スルホン酸、p−トルエンスルホン酸、カンフア
−スルホン酸)、塩基(例、水酸化ナトリウム、
水酸化カリウム、水酸化リチウム、水酸化バリウ
ム、アンモニア、トリエチルアミン)或いは塩
(例、塩化ナトリウム、塩化カルシウム、炭酸ナ
トリウム、炭酸カリウム、炭酸水素ナトリウム、
塩化アンモニウム)で至適PH(通常、PH約2ない
し10)に調整することにより反応速度並びに収率
を向上させることができる。また最後の酸処理で
使用される酸としては、たとえば塩酸、硫酸、硝
酸、リン酸、ホウ酸、酢酸、シユウ酸、酒石酸、
トリフルオロ酢酸、メタンスルホン酸、ベンゼン
スルホン酸、p−トルエンスルホン酸などが挙げ
られる。上記方法によつて製造される化合物
()は、反応混合物から通常の分離精製手段、
たとえば濃縮、溶媒抽出、再結晶、クロマトグラ
フイーなどを適宜使用して単離する事ができる。
このようにして、化合物()が遊離形で得ら
れた場合に、常法により塩の形にして、本発明方
法の原料化合物として用いてもよい。
化合物()の塩としては、前記した化合物
()の塩と同様の塩が挙げられる。
本発明方法に用いられる化合物()は、塩の
形のものを用いてもよい。化合物()の塩とし
ては、前記した化合物()および化合物()
の塩と同様のものがあげられる。
以下に、実施例、参考例および実施例を挙げて
本発明をさらに具体的に説明する。
実験例 1
実施例1の化合物のtRNAへの取り込み実験:
〔3H〕グアニンラベルtRNA(※)
(8000cpm)、70mMのトリス塩酸塩、60mMの塩
化マグネシウム、0.5単位のラツト肝tRNA−グ
アニントランスグリコシレース及び0.02OD260の
後述の実施例1で得られた化合物を100μにな
る様に調整し、37℃で17時間反応した。反応液を
ワツトマン3MM紙(ワツトマン社製、米国)
につけて5%トリクロロ酢酸水溶液で3回洗滌
し、更にエタノール/エーテル(1:1)及びエ
ーテルのみで各1回洗滌し、乾燥後、トルエン系
シンチレーシヨン液にて測定すると81%のtRNA
に実施例1の化合物が取り込まれていた。
(※) tRNAのアンチコドンの第1字目〔3H〕
グアニンでラベルしたtRNA。
実験例 2
実験例1と同様にして、実施例2の化合物の
tRNAへの取り込み率を測定したところ57%であ
つた。
実験例 3
1×104個のL5178Yマウス腫瘍細胞を10%牛胎
児血清、20μmの2−メルカプトエタノールおよ
び100μg/mlのカナマイシンを含有する2ml培
養液RPMI−1640(日水製薬株式会社製)に懸濁
し、薬物投与前24時間37℃で培養した。ついで稀
釈液として上記の培養液を用い、後述の実施例1
で得られた化合物を5段階の1/4稀釈で化合物の
最大濃度が200μg/mlになる様に培地に入れ、
更に72時間培養し、細胞数をカウンターで測定
し、非処理対象群を100%としてIC50(50%増殖阻
止濃度)を算出したところ、22μg/mlであつ
た。
実験例 4
体重20gのBALB/cマウスの皮下に5×104
個のMethA腫瘍細胞を移植し、移植後、4日目
か1日1回、14日間連続して上記マウスに対する
投与量が100mg/Kgとなるよう後述の実施例1で
得られた化合物を蒸留水0.1mlに溶解した溶液を
マウスの腹腔内に注射投与した。移植後35日目の
腫瘍結節を摘出し、その重量を測定した。無投与
の対象群のそれと比較して腫瘍の成長抑制が認め
られた。腫瘍阻止率は71%を示した。
参考例 1
2−アミノ−4−クロロピロロ〔2,3−d〕
ピリミジンの製造:
2−アミノピロロ〔2,3−d〕ピリミジン−
4−オン(45g)をオキシ塩化リン(150ml)に
懸濁し、110℃で3時間撹拌反応した。過剰のオ
キシ塩化リンを減圧で留去し、残渣に氷水(600
ml)を加え全溶した後、冷却撹拌下、濃アンモニ
ア水でPH9に調整した。生じた沈澱物を取し、
水で洗滌後、熱メタノール(4)より再結晶す
ると目的物(34.6g)が得られた。

4−クロロ−2−n−オクタノイルアミノプロ
ロ〔2,3−d〕ピリミジンの製造:
参考例1で得られた2−アミノ−4−クロロピ
ロロ〔2,3−d〕ピリミジン(16.9g)を乾燥
ピリジン(200ml)に懸濁、溶解し、氷冷撹拌下
にn−オクタノイルクロリド(21.2g)を加えた
後、室温に戻して1時間反応した。反応混合物
に、9.9%エタノール性アンモニア(w/v)を
加え、更に室温で2時間撹拌放置した後、溶媒を
減圧で留去した。残渣に水(500ml)を加え、室
温で1時間撹拌し、生じた不溶物を取し、重曹
水、次いで水で洗滌し乾燥すると黄色の粉末状晶
(23.3g)となつた。このものを1,2−ジメト
キシエタンより再結晶すると目的物(20.9g)が
得られた。

エタノール(240ml)に懸濁、溶解し、100℃で
2.5時間撹拌反応した。溶媒を減圧で留去した後、
残渣に1.8%の重曹水(300ml)を加え、室温にて
よく撹拌した。生じた沈澱を取し、エタノール
により再結晶すると目的物(15.7g)が得られ
た。

(3.5g)とジベンジルアミン(9.5g)とを、
水/酢酸(1:4、120ml)に溶解し、35%ホル
マリン(4.2g)を加えて、60℃で14時間反応し
た。溶媒を減圧で留去し、水(30ml)を加えて再
び留去した後、残渣に2N−HCl(64ml)及びメタ
ノール(100ml)を加えて60℃で1.5時間撹拌し
た。大部分のメタノールを減圧留去し、水層をア
ンモニア性アルカリとなし、酸性亜硫酸ソーダ
(5.7g)を添加した後、クロロホルムで抽出し
た。クロロホルム層を濃縮乾固すると粗目的物
(3.95g)が得られた。このものはこれ以上精製
することなく直接次の反応に用いることが出来
る。

ン−4,5−ジヒドロキシシクロペント−1−エ
ン−3−イルアミン(1.55g)とをエタノール
(150ml)に溶解し、封管中75℃で20時間反応し
た。冷後、反応液にテトラヒドロフラン(75ml)
及び40%水酸化カリウム水溶液(11ml)を加え、
5℃で3日間放置した。このものに30%塩化アン
モニウム水溶液(11ml)を加え、濃縮乾固し、残
渣をシリカゲルカラムクロマトグラフイーで、7
%NH3含有エタノール/クロロホルム(1:4)
を流出溶媒として分離精製すると2−アミノ−5
−{(3S,4R,5S)−4,5−O−イソプロピリデ
ン−4,5−ジヒドロキシシクロペント−1−エ
ン−3−イルアミノメチル}ピロロ〔2,3−
d〕ピリミジン−4−チオンが得られた。このも
の全量をメタノール(100ml)及び2N−HCl(16
ml)に溶解し、室温にて一夜放置した後、溶媒を
濃縮乾固すると目的物の2塩酸塩(1.07g)が得
られた。

PreQ1塩基)の製造:
参考例4で得られた粗5−N,N−ジベンジル
アミノメチル−2−オクタノイルアミノピロロ
〔2,3−d〕ピリミジン−4−チオン(1.0g)
をエタノール(50ml)、テトヒドロフラン(30ml)
及び濃アンモニア水(20ml)の混液に溶解し、封
管中、80℃で1.5時間反応した。溶媒を減圧で留
去した後、残渣をエタノールで洗滌し、不溶分を
セルロースパウダー(アビセル、旭化成工業株式
会社社製)で、濃アンモニア飽和n−ブタノール
を流出溶媒とし分離精製すると目的物(215mg)
が得られた。理化学データはメタノール性塩酸で
塩酸塩に変換した後測定した。

よびエーテルで洗浄後乾燥すると目的物(0.89
g)が得られた。
