The present invention is based on the following formula (), The present invention relates to an anti-inflammatory agent containing optically active dibenzothiepic acid represented by (where * indicates optical activity). Traditionally, the following expression () It is known that (±)-dibenzothiepic acid, represented by −53282). Furthermore, as a result of research on the dibenzothiepic acid, the present inventor found that the (+) form has superior anti-inflammatory effects compared to its (±) form, thereby completing the present invention. Therefore, the present invention provides an anti-inflammatory agent containing (+)-dibenzothiepic acid as an active ingredient, which has excellent anti-inflammatory, analgesic, and antipyretic effects, and substantially does not contain its (-)- form. It is. The optically active (+)-dibenzothiepic acid, which is the active ingredient of the anti-inflammatory agent of the present invention, can be obtained by reacting (±)-dibenzothiepic acid () with optically active 1-phenylethylamine, resulting in the following formula (): A diastereomeric salt represented by is formed, and then the two diastereomeric salts are separated using the difference in solubility in a solvent, and (+)-dibenzothiepic acid is liberated from either salt. Manufactured by In this method, either dextrorotatory or levorotatory 1-phenylethylamine can be used as the optically active 1-phenylethylamine. Also, optically active 1 as a resolving agent
- The molar ratio of phenylethylamine and (±)-dibenzothiepic acid is not particularly limited, but (±)-
Substantially resolving agent for dibenzothiepic acid
When 0.5 to 1 equivalent is used, (±)-dibenzothiepic acid can be efficiently resolved. At this time, if (+)-1-phenylethylamine is used, (-)-dibenzothiepic acid precipitates as a poorly soluble diastereomer salt, and the (+)-dibenzothiepic acid of the present invention is easily soluble. Remains in solution as diastereomeric salt.
Furthermore, when (-)-1-phenylethylamine is used, the (+)-dibenzothiepic acid of the present invention forms a poorly soluble diastereomeric salt. In this method, (±)-dibenzothiepic acid and 1-phenylethylamine are reacted in a solvent, and the solvents used at this time include methanol, ethanol, 1-propanol, 2-propanol,
-It is preferable to use butanol, 2-butanol, acetone, methyl ethyl ketone, etc. alone or a mixture thereof, or a hydrated product thereof. In particular, when methanol is used, highly purified (+)-dibenzothiepic acid of the present invention can be obtained. be able to. Next, a preferred embodiment will be described using an example in which methanol is used as a solvent. (±)-Dibenzothiepic acid was heated and dissolved in methanol, an equivalent amount of optically active 1-phenylethylamine was added to the solution while heating (under methanol reflux temperature), and the poorly soluble diastereomeric salt was cooled. Generate. Although the crystallization temperature depends on the boiling point of the solvent used, it is generally preferably in the range of 0°C to 50°C. The mixture of (+)- and (-)-dibenzothiepic acid, which is the substance to be separated, does not need to be a mixture of equal amounts of the (+) and (-) isomers, but may contain at least an equal amount of either one of them. It's okay to be there. The poorly soluble diastereomer salt precipitated by this operation is separated into solid and liquid, once recrystallized from methanol (or partially dissolved by heating), and then treated with a base such as sodium hydroxide, potassium hydroxide, or ammonia. Or (-) 1-phenylethylamine is recovered. Furthermore, the (+)-dibenzothiepic acid of the present invention is obtained by reacting with a mineral acid such as hydrochloric acid or sulfuric acid. Next, the pharmacological results of (+)-, (-)- and (±)-dibenzothiepic acids investigated using the carrageenan edema method on experimental animals will be described. In other words, each group consists of 6 animals weighing 120 to 130 g.
(+)-, (-)-, and (±)-dibenzothiepic acid were orally administered to male Wistar rats, and 1 hour later, 0.1 ml of 1% λ-carrageenan saline was administered subcutaneously to the hind footpad. Inject and increase footpad volume
Using a differential meter, the inhibition rate at the third hour was determined. The results are shown in Table 1.

[Table] As shown in Table 1, the anti-inflammatory effect of the (+)-dibenzothiepic acid of the present invention is 56 times more active than the (-) form and more than twice as active as the (±) form. Next, we used sheep seminal vesicle microsomes as the enzyme source and 14 C-arachidonic acid as the substrate.
The prostaglandin biosynthesis inhibitory activity of the test compounds was tested by the TLC method. The results are shown in Table 2.

[Table] As is clear from Table 2, the (+) dibenzothiepic acid of the present invention is 141 times more than the (-) form, (±)
Shows more than twice the prostaglandin biosynthesis inhibitory activity of the human body. When the (+)-dibenzothiepic acid of the present invention is used as a medicine, it can be used as such or in the form of a salt thereof. The salt is preferably a non-toxic salt such as sodium, potassium, calcium or aluminum. The (+)-dibenzothiepic acid of the present invention exhibits its effects both in oral and parenteral administration, and can be formulated into dosage forms for oral, injection, rectal, and topical administration. Solid dosage forms for oral administration include capsules, tablets, pills, powders, and granules. As additives for these dosage forms, in addition to excipients such as white sugar, lactose, and starch, a lubricant such as magnesium stearate can be used. It can also be made into enteric-coated and sustained-release dosage forms. Liquid dosage forms for oral administration include emulsifiers, solutions, suspensions, syrups, and elixirs. As additives for these dosage forms, in addition to solvents such as purified water and alcohols, auxiliary agents such as wetting agents, emulsifiers, and suspending agents can be added. Preparations for parenteral administration of (+)-dibenzothiepic acid of the present invention include injections, suppositories, and the like. In the case of injections, it can be a sterile aqueous or non-aqueous solution. Examples of solvents include propylene glycol, polyethylene glycol, vegetable oil,
Examples include organic acid esters. It may also be in the form of a powder filling, which can be dissolved in a solvent before use. In the case of suppositories, excipients such as cocoa, butter or suppository waxes are added. The dosage of (+)-dibenzothiepic acid of the present invention varies depending on the symptoms, administration route, administration period, etc., but in general, the daily dose for humans is 1 to 1.
500mg is preferred. Therefore, the (+)-dibenzothiepic acid of the present invention can exhibit the desired medicinal effect with a smaller amount of active ingredient than the (±) form, and can reduce side effects and make tablets etc. smaller. It is practically useful. Next, an example will be given and explained. Example 1 (±)-dibenzothiepic acid in 70 ml of methanol
