The present invention relates to a method for culturing bacterial cells belonging to the genus Brevibacterium. According to the method of the present invention, it is possible to increase the content of aspartase produced within microbial cells belonging to the genus Brevibacterium, and the relative specific activity of aspartase is significantly increased. It is well known that aspartase is an enzymatic catalyst for the reaction that produces L-aspartic acid from fumaric acid and ammonia. Also, this L-
Aspartic acid is widely known as one of the important amino acids and is used as a medical product and food additive. Conventionally, L- is produced from fumaric acid, ammonia, etc. by fermentation methods using various bacterial cells or enzymatic methods using aspartase produced by these bacteria.
Many attempts have been proposed to produce aspartic acid. In this case, it is important to newly collect bacterial cells with high aspartase activity from nature. However, it is extremely difficult to obtain such bacterial cells, so instead, for example, the proposal of the present inventors (Japanese Patent Application Laid-Open No. 56-26196)
α-
A method for conferring amino-n-butyric acid tolerance and increasing the ability to synthesize L-aspartic acid from fumaric acid and ammonia, also by I. Chibata et.al., Appl.
Microbiol., 27, 878 (1974) proposed Escherichia coli as an aspartase-producing bacterium. After culturing, the bacterium was placed in an aqueous solution containing fumaric acid at 37°C for several tens of hours. It is known that immersion can increase aspartase activity by about 10 times. It has been revealed that the increase in activity in this method is due to the destruction of the bacterial cell membrane by autolysis, which improves the permeation efficiency of substrates and products, resulting in an apparent increase in activity.
Furthermore, JP-A-57-138383 proposes a method of increasing the yield of L-aspartic acid by treating microorganisms with aspartase activity with acid to selectively inactivate only fumarase activity. . However, when the purpose is to industrially produce L-aspartic acid, the yield of L-aspartic acid can be improved by increasing the aspartase activity by treating the aspartase-containing bacterial cells after culturing as described above. It is also important to increase the amount of aspartase produced by the bacterial cells. In view of the above, the present inventors have intensively investigated a method for culturing bacteria belonging to the genus Brevibacterium in order to obtain aspartase in a short culture time and in high yield. The present invention was completed based on the discovery that when cultured aerobically in a medium containing the following, the content of aspartase produced within the microbial cells increases significantly in a short culture time. That is, the present invention provides a method for culturing microbial cells belonging to the genus Brevibacterium, in which the microbial cells having aspartase-producing ability are cultured with fumaric acid in an amount of 0.5 to 2.8%.
The present invention provides a method for culturing bacteria belonging to the genus Brevibacterium, which is characterized by culturing aerobically in a medium containing the present invention. The aspartase-producing bacterial cells used in the present invention belong to the genus Brevibacterium. For example, Brevibacterium flavum MJ233 (FERM3068), a strain derived from Brevibacterium flavum MJ233 as an α-amino-n-butyric acid resistant strain: Brevibacterium flavum MJ233-AB-41 (FERM3812)
(Refer to Japanese Patent Application Laid-Open No. 56-26196). The composition of the medium used in the present invention, including the carbon source, nitrogen source, and inorganic salts, is not particularly limited. Examples of carbon sources include ethanol, n
- Paraffin, molasses, etc. can be used, and suitable nitrogen sources include ammonium salts such as ammonium sulfate, ammonium chloride and ammonium phosphate, nitrates such as sodium nitrate and potassium nitrate, or ammonia, urea and the like. As the inorganic salt, potassium monohydrogen phosphate, potassium dihydrogen phosphate, agnesium sulfate, etc. are used. In addition, nutrients such as veptone, meat extract, yeast extract, cornstarch liquor, casamino acids, and various vitamins can be added to the medium if necessary for the growth of the bacteria. Cultivation is carried out under aerobic conditions with aeration, stirring, shaking, etc. The culture temperature is 20-40°C, preferably 25-35°C. The pH during the cultivation is 5 to 10, preferably about 7 to about 8. In addition, the culture solution
A known method of adding acid or alkali is used to adjust the pH. In the method of the present invention, the bacterial cells belonging to the genus Brevibacterium are cultured in the above medium, and at this time, fumaric acid is added in an amount of 0.5 to 2.8% by weight, preferably 1 to 2.8% by weight.
It is necessary to add it within the range of 2.6% by weight. When the amount of fumaric acid added is outside this range, the improvement in the relative aspartase activity of the culture is small or almost non-existent. Ethanol concentration at the start of culture is 1~
5% by volume, preferably 2-3% by volume is suitable. The culture period is usually 15 to 24 hours. Since the bacterial cells cultured by the method of the present invention contain highly active aspartase at a high concentration, culture fluid containing these bacterial cells, bacterial cells isolated from the culture fluid, destroyed or ground bacterial cells , Fumaric acid or a fumarate salt such as its sodium salt or calcium salt, and ammonia or an ammonium salt such as ammonium chloride or ammonium carbonate, by a known method using an autolytic solution of bacterial cells or immobilized bacterial cells. L-aspartic acid can be efficiently biosynthesized from In the method for producing L-aspartic acid, for example, bacterial cells cultured by the method of the present invention or a treated product such as an immobilized product thereof can be used as an enzyme source for producing L-aspartic acid, and more preferably. The bacterial cells or their immobilized products were heated in advance at 40°C or higher and at 60°C in the alkaline pH range in the presence of L-aspartic acid and ammonium ions.
A heat-treated product can also be used below. When using fumaric acid or a salt thereof and ammonia or an inorganic ammonium salt, the molar ratio of these two components is suitably between 1:1 and 5.
The enzymatic reaction can be carried out at a temperature range of 0 to 60°C, but considering the stability of aspartase,
Preferably it is carried out at 20-50°C. Example 1 50 ml of the culture medium shown in Table 1 was dispensed into a 500 ml Erlenmeyer flask and sterilized at 120°C for 15 minutes.
After adding 2% by volume of ethanol, Brevibacterium flavum, an aspartase producing bacterium
Inoculated with MJ233-AB-41 (FERM3812) and 30℃
Culture was carried out for 24 hours. 20ml of this culture solution,
Inoculate medium 1 with the composition shown in Table 2 in the jar fermenter No. 2 at 33°C, pH 7.6 (adjusted with 28% ammonia water), aeration volume 1vvm, and stirring speed.
Culture was performed at 800 rpm for 20 hours. At this time, the temperature of the fumaric acid added is 0 to 0 as shown in Table 4.
The content was changed to 2.5% by weight. In all experiments, ethanol was added intermittently so that the ethanol concentration in the medium was maintained at 1 to 1.5% by volume. After culturing, centrifuge the culture solution (4000 rpm, 15
minutes), then suspend the collected bacteria in distilled water and OD
A bacterial cell suspension with a (optical density, absorbance at a wavelength of 610 mm) value of 50 was prepared, and this bacterial cell suspension was used as a test solution. Aspartase activity was measured by reacting 1.0 ml of the reaction solution shown in Table 3 at 46°C for 1 hour.
Centrifuge the reaction completed solution (4000 rpm, 15 minutes)
After that, the amount of aspartic acid produced in the supernatant was determined by a microbial quantitative method using Leuconostocmesenteroides ATCC8042.
Aspartase activity was expressed as relative specific activity, with the oxygen activity of bacterial cells cultured without fumaric acid added as 100. The results obtained are shown in Table 4.
