本発明は分離剤、特に多糖のヘテロ芳香族誘導
体を有効成分とする分離剤に関する。 〔産業上の利用分野〕 本発明の分離剤はあらゆる化学物質の分離、特
に光学分割に用いることができる。 よく知られているように、化学的には同じ化合
物であつてもその光学異性体は通常生体に対する
作用を異にする。従つて医、農薬、生化学関連産
業等の分野において、単位当りの薬効の向上や、
副作用、薬害の防止等の目的のために、光学的に
純粋な化合物を調製することが極めて重要な課題
となつている。光学異性の混合物を分離、即ち光
学分割するためには従来優先晶出法やジアステレ
オマー法が用いられているが、これらの方法では
光学分割される化合物の種類は限られており、ま
た長い時間と多大な労力を要する場合が多い。従
つてクロマトグラフイー法によつて簡便に光学分
割を行なうための技術が強く望まれている。 〔従来技術〕 クロマトグラフイー法による光学分割の研究は
以前から行なわれている。しかし従来開発された
分離剤は、分離効率が良くないこと、分割の対象
とする化合物が特殊な官能基を必要とすること、
あるいは分離剤の安定性が良くないことなど、い
ろいろな問題があり、すべての化合物に対して満
足すべき光学分割を行なうことは難かしかつた。 〔発明の目的〕 従つて既存の分離剤とは異なつた化学構造を持
ち、そのことによつてそれらとは異なつた分離特
性を有し、あるいはより高度な光学異性体識別能
力を有する分離剤を提供することが本発明の目的
である。特にヘテロ芳香族環の有するヘテロ原子
(窒素、酸素、イオウ等)の有する物理的、化学
的特性を新規な分離特性に結びつけようとするも
のである。 〔発明の構成〕 本発明はセルロースのヘテロ芳香族誘導体を有
効成分とする分離剤によつて上記目的を達成する
ものである。 本発明の分離剤は好ましくは何らかの化合物の
光学異性体に対して異なつた吸着力を示すもので
ある。 (多糖) 本発明における多糖とは、合成多糖、天然多
糖、天然物変成多糖のいずれかを問わず、光学活
性であればいかなるものでも良いが、好ましくは
結合様式の規則性の高いものである。例示すれば
β−1,4−グルカン(セルロース)、α−1,
4−グルカン(アミロース、アミロペクチン)、
α−1,6−グルカン(デキストラン)、β−1,
6−グルカン(プスツラン)、β−1,3−グル
カン(例えばカードラン、シゾフイラン等)、α
−1,3−グルカン、β−1,2−グルカン

いものであればいかなるものでも良いが、例えば
ピリジンまたはキノリン等である。しばしば4−
(N,N−ジメチルアミノ)ピリジンのような触

ラフイー法などのクロマトグラフイー法を用いる
のが一般的であるが、膜分離を行なうこともでき
る。 本発明の分離剤を液体クロマトグラフイー法に
応用するには、粉体としてカラムに充填する方
法、キヤピラリーカラムにコーテイングする方
法、該分離剤によつてキヤピラリーを形成し、そ
の内壁を利用する方法、紡糸し、これを束ねてカ
ラムとする方法などの方法がとられるが、粉体と
することが一般的である。 該分離剤を粉体とするにはこれを破砕するかビ
ーズ状にすることが好ましい。粉子の大きさは使
用するカラムやプレートの大きさによつて異なる
が、1μm〜10mmであり、好ましくは1μm〜300μm
で、粒子は多孔質であることが好ましい。 更に分離剤の耐圧能力の向上、溶媒置換による
膨潤、収縮の防止、理論段数の向上のために、該
分離剤を担体に保持させることが好ましい。適当
な担体の大きさは使用するカラムやプレートの大
きさにより変るが、一般に1μm〜10mmであり、好
ましくは1μm〜300μmである。担体は多孔質であ
ることが好ましく、平均孔径は10Å〜100μmであ

離剤を結晶化する場合には熱処理などの処理を行
なうことができる。また、少量の溶剤を加えて該
分離剤を一旦膨潤あるいは溶解せしめ、再び溶剤
を留去することにより、その保持状態、ひいては
分離能を変化せしめることが可能である。 担体としては多孔質有機担体又は多孔質無機担
体があり、好ましくは多孔質無機担体である。多
孔質有機担体として適当なものは、ポリスチレ
ン、ポリアクリルアミド、ポリアクリレート等か
ら成る高分子物質が挙げられる。多孔質無機担体
として適当なものはシリカ、アルミナ、マグネシ
ア、酸化チタン、ガラス、ケイ酸塩、カオリンの
如き合成若しくは天然の物質が挙げられ、該分離
剤との親和性を良くするために表面処理を行なつ
ても良い。表面処理の方法としては有機シラン化
合物を用いたシラン化処理やプラズマ重合による
表面処理法等がある。 液体クロマトグラフイーあるいは薄層クロマト
グラフイーを行なう場合の展開溶媒としては、該
分離剤を溶解またはこれと反応する液体を除いて
特に制約はない。該分離剤を化学的方法で担体に
結合したり、架橋により不溶化した場合には反応
性液体を除いては制約はない。いうまでもなく、
展開溶媒によつて化学物または光学異性体の分離
特性は変化するので、各種の展開溶媒を検討する
ことが望ましい。 一方薄層クロマトグラフイーを行なう場合には
0.1μm〜0.1mm程度の粒子から成る該分離剤と、必
要であれば少量の結合剤より成る厚さ0.1〜100mm
の層を支持板上に形成すればよい。 膜分離を行なう場合には中空系あるいはフイル
ムとして用いる。 (発明の効果) 本発明の多糖のヘテロ芳香族誘導体を有効成分
とする分離剤は各種化合物の分離に有効であり、
特に従来分離が非常に困難であつた光学異性体の
分離に極めて有効である。分離の対象となる光学
異性体は本分離剤によつてそのどちらか一方がよ
り強く吸着されるものである。 以下本発明を実施例によつて詳述するが、本発
明はこれらの実施例に限定されるものではない。 合成例 1 シリカビーズ(Merck社製LiChrospher
SI1000)10gを200ml枝付丸底フラスコに入れ、
オイルバスで120℃、3時間真空乾燥した後N2を
入れた。CaH2を入れて蒸留したトルエンをシリ
カビーズに100ml加えた。次にジフエニルジメト
キシシラン(信越化学KBM202)を3ml加えて
攪拌後、120℃で1時間反応させた。更に3〜5
mlのトルエンを留去後120℃で2時間反応させた。
グラスフイルターで過し、トルエン50mlで3
回、メタノール50mlで3回洗浄し、40℃で1時間
真空乾燥を行なつた。 次に該シリカビーズ約10gを200ml枝付丸底フ
ラスコに入れ、100℃で3時間真空乾燥した後、
常圧に戻し室温になつてからN2を入れた。蒸留
したトルエン100mlを乾燥したシリカビーズに加
えた。次にトリメチルシリル化剤N,O−Bis−
(トリメチルシリル)アセトアミド1mlを加えて
攪拌し、115℃で3時間反応させた。次にグラス
フイルターで過後、トルエンで洗浄をし、約4
時間真空乾燥した。 合成例 2 数平均重合度110、置換度2.97のセルロースト
リアセテート(ダイセル化学工業製)を1の酢
酸(関東化学製)に溶解し、5.2mlの水と5mlの
濃硫酸を加え、80℃、3時間反応させた。反応液
を冷却し、過剰の酢酸マグネシウム水溶液で硫酸
を中和した。該溶液を3の水中に入れて、低分
子量化したセルローストリアセテートを沈澱させ
た。グラスフイルター(G3)によつて別、更
に1の水に分散後別し、真空乾燥した。生成
物を塩化メチレンに溶解させ、2−プロパノール
に再沈澱する操作を2回繰り返して精製した後乾
燥した。生成物は、IRスペクトル及びNMRスペ
クトルよりセルローストリアセテートであり、蒸
気圧浸透圧法より求めた数平均分子量は7900で、
数平均重合度に換算すると27であつた。蒸気圧浸
透圧法は、ペーパープレツシヤ−オスモメーター
CORONA117を用いて溶媒にクロロホルム−1
%エタノールの混合溶媒を使用して測定した。 こうして得られたセルローストリアセテート60
gを2−プロパノール200mlに分散し、ゆるやか
に攪拌しながら60mlの100%ヒドラジンヒドラー
ド(半井化学製)をゆつくり滴下した。該懸濁液
を60℃に3時間保つた後、グラスフイルターで生
成したセルロースをロ別し、これをアセトンによ
り繰り返し洗滌した後60℃で真空乾燥した。生成
物のIRスペクトルには1720cm-1付近のカルボニ
ル基に基づく吸収は全く検出されず、セルロース
のそれと一致した。 合成例 3 (セルローストリスチオフエン−2−カルボキシ
レートの合成) 合成例2で得たセルロース1.5gに脱水したピ
リジン70ml、脱水したトリエチルアミン7.7ml、
4−ジメチルアミノピリジン50mgを加え、攪拌し
ながら、チオフエン−2−カルボニルクロリド
12.2gを添加し、100℃で5時間攪拌反応した。
冷却後エタノール400mlに生成物を攪拌しながら
加えて沈澱させ、グラスフイルターでろ過後エタ
ノールで良く洗浄した。真空乾燥した後、塩化メ
チレン30mlに溶解し、不溶物を除いた後400mlの
エタノールに再沈澱した。沈澱をろ過後、エタノ
ールで洗浄し、脱液、乾燥した。 3.9gのセルロースチオフエン−2−カルボキ
