【発明の詳細な説明】[Detailed description of the invention]

産業上の利用分野 本発明は、好気的培養条件下に、酸素を通気す
ることにより、高収量でクエン酸を製造する方法
に関するものである。 従来技術 ノルマルパラフイン(炭素数10ないし20のパラ
フイン類)を発酵原料としてキヤンデイダ属
(candida)の酵母によりクエン酸を製造する方
法は従来から知られている。 また炭素源としてオレフイン類を含む発酵培地
を用い、キヤンデイダ・トロピカリス
(Candida・tropicalis)に属する微生物を好気的
に培養してクエン酸を製造する方法については本
出願人と同一出願人によつて特許出願されている
(特許第1096815号等)。炭素源としてパラフイン
類およびオレフイン類を含む発酵培地からのクエ
ン酸発酵においては培養条件によつて異なるが、
クエン酸の他に常にかなりの量のイソクエン酸の
生成をともなう。その量はクエン酸と同量または
それより多量である場合もある。 発明の解決しようとする問題点 従来、石油発酵および油脂発酵によるクエン酸
の製造においては副生してくるイソクエン酸の生
産を抑制してクエン酸の生産を支配的にするため
に、もつぱら変異株の使用による方法が採用され
てきた。 しかしながら、この方法では変異株の探索およ
びこれに適する培養条件の決定など多くの労力と
経費とを必要とする。また、取得した変異株を使
用する場合でも、回避不可能に近い鉄イオンの影
響により、イソクエン酸の生成をみることがあ
り、結果として、クエン酸の収量低下をきたすこ
とがある。 これは、菌体中に保持されている、クエン酸を
イソクエン酸に変換する酵素アコニターゼが鉄イ
オンの存在により再び賦活されるからである。 従つて、一端、イソクエン酸生成の少ない変異
株が得られたとしても、その保守管理に細心の注
意を要するものである。 本発明方法によれば変異株を使用することなく
イソクエン酸の生成を抑制することができるので
本発明の工業的意義はきわめて大きいものであ
る。 問題点を解決するための手段 本発明者らは、クエン酸の収率向上を目的とし
てクエン酸を製造するにあたり、好気的条件下で
培養する際に酸素を通気することにより、培地中
の酸素濃度を増大させることによりイソクエン酸
の生成を抑制し、クエン酸の収率向上が図れるこ
とを発見して本発明に到達したものである。 本発明は、炭素源として油脂類、脂肪酸類、こ
れらのエステル類、直鎖状パラフイン類および糖
類を含む培地にキヤンデイダ属より選ばれた酵母
を好気的に培養し、該培地中にクエン酸を蓄積さ
せ、これを採取するクエン酸の製法において、好
気的条件下における培養の際に、酸素を通気する
ことにより、イソクエン酸の生成を抑制し、高収
量でクエン酸を得ることを特徴とするクエン酸の
製法に関するものである。 溶存酸素量は少なくとも10ppm以上、好ましく
は20ppm以上、さらに好ましくは30ppm以上であ
る。 溶存酸素量が10ppm以上であればイソクエン酸
生成の抑制効果が得られる。 本発明で使用する微生物は、各種炭素源を資化
してクエン酸を培地中に蓄積する能力を有するキ
ヤンデイダ属より選ばれた酵母である。更に、こ
れらの酵母の栄養要求変異株および変種も含むも
のである。 本発明において炭素源として使用する油脂類、
それら油脂類から得られる脂肪酸類、および天然
または合成脂肪酸類、それらのエステル類には特
に制限がなく、単独または混合物として適宜使用
することができる。 また、ノルマルパラフイン類としては炭素数10
ないし20のものが使用されるが、通常はn−C12
〜n−C13がよく使われる。 さらに、各種糖類も良好な炭素源となる。 発酵原料の培地への添加量は1ないし20%(重
量)で、好ましくは5ないし15%(重量)であ
る。 窒素源としては無機アンモニウム塩、硝酸塩お
よびアンモニアも使用できる。上記の各種窒素源
は単独で使用してもよくまた2種以上を混合して
使用してもよい。 無機塩としては、第1燐酸塩、第2燐酸塩、硫
酸塩、塩酸塩、カリウム塩、ナトリウム塩、マグ
ネシウム塩、マンガン塩、銅塩、亜鉛塩などの通
常の無機塩類が使用できる。 更にPHを調整するため炭酸カルシウム、アンモ
ニア、水酸化ナトリウム等が使用される。 有機微量栄養素としては、ピオチンおよびチア
ミン単独またはこれらの混合物およびこれらを含
む酵母エキス、コンステイーブリカーなどの天然
物を使用してもよい。 チアミンおよびピオチンをそれぞれ単独で使用
する場合には1000μg/以下であれば十分で、
それ以上使用してもクエン酸の生成量には変化が
ない。好ましくは50ないし100μg/を使用する
場合が適当である。チアミンとビオチンとを併用
する場合は合計で1000μg/以下を使用すれば
十分である。 本発明方法で使用する発酵培地において、発酵
原料の培地への分散性を良くするため阻害効果の
ない界面活性剤を使用することは勿論差支えない
が、発酵原料を培養条件下で液状に保持できるの
で界面活性剤を使用する必要がない。従つて界面
活性剤による酵母への阻害作用を考慮する必要が
ない。 発酵条件は、酸素通気による好気的条件がよ
い。培養温度は25℃ないし40℃の範囲で、好まし
くは25℃から38℃である。培養中のPHは3ないし
10の範囲で、好ましくは4ないし6の範囲が適当
である。培養中のPHの調整は酸、アルカリ、炭酸
カルシウム、炭酸ナトリウムまたはアンモニアを
用いて行なうことができる。 培養は通常50ないし150時間、好ましくは60な
いし100時間行なわれる。 発酵培地中に蓄積されたクエン酸は、通常微生
物の培養によつて生産された有機酸塩類をその培
養物から分離するのに用いられる手段、たとえば
過、遠心分離などの方法で菌体を分離した後、
カラムクロマト処理、イオン交換樹脂処理、ある
いは濃縮処理などによりクエン酸またはその塩と
して分離採取される。 クエン酸とイソクエン酸の分析は高速液体クロ
マトグラム法で行なつた。 実施例 以下に実施例により本発明を説明するが、本発
明はこれに限定されるものではない。 実施例 1 炭素源としてパームオレインを使用した。 使用菌株はキヤンデイダ・トロピカリスIFO−
0589(Candia tropicalis)であつた。 本発明方法で使用する培地の基本組成は次の如
くである。 種母用 パームオレイン (Palm Olein) 5(wt/vol%) NH4Cl 0.50(wt/vol%) KH2PO4 0.05(wt/vol%) MgSO4・7H2O 0.05(wt/vol%) FlSO4・7H2O 1(mg/) MnSO4・7H2O 0.1(mg/) ZnSO4・nH2O 0.1(mg/) CuSO4・5H2O 5(μg/) ビオチン 100(μg/) チアミン塩酸塩 100(μg/) PH 5.0 生産用 パームオレイン (Palm Olein) 10wt% NH4Cl 0.3(wt/vol%) KH2PO4 0.05(wt/vol%) MgSO4・7H2O 0.03(wt/vol%) FlSO4・7H2O 5(mg/) MnSO4・7H2O 0.06(mg/) ZnSO4・nH2O 0.05(mg/) CuSO4・5H2O 5(μg/) ビオチン 50(μg/) チアミン塩酸塩 100(μg/) PH 5.0 種母培養はフラスコ振とう培養機を用い、つぎ
の条件で行つた。 種培養としてスラント培養後の上記組成の寒天
斜面培地から4白金耳量の菌株を、上記組成の培
地30mlを容量500mlの肩付振とうフラスコに入れ
た液体培地に接種した後、振とう数120往復/分、
振巾7cmの往復振とう機で30℃、3日間培養し
た。 なお、培地のPHが5.0以下に低下したとき、別
に殺菌したCaCO35wt/vol%を添加し、PHを調
整した。ジヤーフアメンターを使用して種母培養
する場合には酸素通気することができる。 この種母を用い生産培養を行つた。内容3の
ジヤーフアーメンターに上記培養液1258ml、パー
ムオレイン166ml、種母75mlを入れ、温度34℃、
回転数1200r.p.m、酸素通気量は培養液容量に対
して1/2V/V/min.(溶存酸素量10ppm以上)
である。 培養終了後、この培養物を塩酸でPH2.5にした
後珪藻土を過助剤として吸引過し、酵母を主
とする固型物はこれを少量の水で洗滌した。次に
液と洗滌液とを合せた後、苛性ソーダで中和後
加熱して沈澱を析出させ、これを冷却後吸引過
してクエン酸カルシウムを得た。 得られたクエン酸カルシウムは約10倍量の水に
懸濁し、これにその液が塩化バリウム溶液によ
つて硫酸根の存在をわずかに示すに至るまで撹拌
しつゝ、50%硫酸水溶液を滴下した後沸とう水中
で約30分間加熱する。 必要によつてはこれに脱色操作を施した後熱時
過して液を50〜80℃にて減圧濃縮し、途中石
膏の沈澱が析出した場合、これを別し、更にこ
れを別し、更に濃縮を続け、やゝうすいシラツ
プ状にする。このシラツプ状濃縮物を氷点下に放
置してクエン酸を析出させた。 同様の方法で、酸素にかえた空気通気による培
養も行つた。 その結果を第1表に示す。

This is because the enzyme aconitase, which converts citric acid to isocitrate, which is retained in the bacterial cells, is reactivated by the presence of iron ions. Therefore, even if a mutant strain that produces less isocitrate is obtained, careful attention must be paid to its maintenance and management. According to the method of the present invention, the production of isocitrate can be suppressed without using mutant strains, so the industrial significance of the present invention is extremely large. The present invention involves culturing yeast selected from the genus Candeida aerobically in a medium containing fats and oils, fatty acids, their esters, linear paraffins, and sugars as carbon sources, and adding citric acid to the medium.
