本発明は、光学活性アミノプロパンジオール類
の立体配置を反転する方法に関するものである。
更に詳しくは、一般式
(式中、*は不斉炭素を示す。R1はC1〜C4の
低級アルキル基を示す。)で表わされる光学活性
アミノプロパンジオール類を、一般式
(式中、*は不斉炭素を示す。R1はC1〜C4の
低級アルキル基を示す。R2はC1〜C4の低級アル
キル基、ベンジル基、または置換ベンジル基を示
す。)で表わされる光学活性エポキシ化合物に誘
導し、次いで水、あるいは塩基性水溶液と反応さ
せることを特徴とする、一般式

アミノプロパンジオール類に立体配置を反転する
方法に関するものである。
光学活性N−置換アミノプロパンジオールに関
して、その立体配置が(S)体の化合物は光学活
性のβ−受容体遮断薬の有用な合成中間体である
ことが知られている。Nの置換基についてはイソ
プロピル基およびt−ブチル基が薬効の面から重
要である。
一般に、不斉炭素を有する化合物を、光学分
割、あるいは不斉加水分解によつて得た場合、工
業的には不必要な光学異性体をラセミ化させて、
もう一度利用するか、望ましくは不必要な光学異
性体を必要な光学異性体に反転させる方法が必須
である。一般式()で表わされる光学活性N−
置換アミノプロパンジオールの有効な所望とする
光学活性体への反転方法の報告例はなく、さらに
有効なラセミ化方法さえ見出されていない。また
光学活性2−アミノアルコール類の立体配置を変
換する方法としては特開昭49−75545、特開昭55
−20785が知られているが、光学活性N−置換ア
ミノプロパンジオールには適用できなかつた。
本発明者らは、この点に関し検討し、光学活性
なN−置換アミノプロパンジオールから誘導され
るエポキシ化合物を水あるいは塩基性水溶液で処
理すると驚くべきことに不斉炭素の立体配置が反
転すると同時に、一挙に所望する立体配置を有す
るN−置換アミノプロパンジオールが生成するこ
とを見出して本発明を完成させた。
具体的に本発明の反転方法を式で示すと式1の
通りに表わされる。

ロパンジオール化合物は、本出願人の特許出願
(特開昭59−31692、同59−31693、同59−78696)
により得られる光学活性5−ヒドロキシメチル−
N−置換オキサゾリジン−2−オンをアルカリ加
水分解して容易に取得できる。
次に、立体配置の保持された光学活性エポキシ
化合物()への変換方法は種々の公知の反応を
組み合わせて達成できる。例えば式2および式3
の方法によつて行われる。
即ち、式2に示すように、光学活性N−置換ア
ミノプロパンジオールをHBr−酢酸と反応させ、
次いでナトリウムメチラート等の塩基でエポキシ
誘導体に変換し、ついでクロル炭酸アルキルやク
ロル炭酸ベンジル等でアミノ基をカルバメート基
にして光学活性エポキシ化合物()を得ること
ができる。式3では、HBr−酢酸の替りにp−
トルエンスルホニルクロリド等で光学活性のトシ
レート体に誘導し、ついでナトリウムメチラート
等の塩基で式2と同様のエポキシ誘導体に変換で
きる。これらの光学活性N−置換アミノプロパン
ジオールから化合物()のエポキシ化合物に変
換する工程では不斉炭素の立体配置は保持されて
いる。
本発明における反転工程の原料の光学活性エポ
キシ化合物()の置換基R1はC1〜C4の低級ア
ルキル基から選択することができる。たとえばメ
チル、エチル、プロピル、イソプロピル、ブチ
ル、t−ブチル基等が挙げられるが、薬効の高い
β−受容体遮断薬に対応するアルキル基としてイ
ソプロピル基とt−ブチル基が好適である。化合
物()の置換基R2はアミノ基をカルバメート
基にする際の試薬により規定される。この試薬と
してはクロル炭酸メチル、クロル炭酸エチル、ク
ロル炭酸ベンジル又は、クロル炭酸置換ベンジル
が使用できる。これらに対応する置換基R2はメ
チル、エチル、ベンジルおよび置換ベンジル基で
ある。置換ベンジル基としてはp−メトキシベン
ジル、p−ニトロベンジル、o−メトキシベンジ
ル、o−ニトロベンジル等の置換ベンジル基が使
用できる。上記の光学活性エポキシ化合物()
のアミノ基の置換基R2は反転工程において脱離
する基であり、工業的には、R2がメチルおよび
エチル基が好ましい。
これらの光学活性エポキシ化合物()を水あ
るいは塩基性水溶液で処理することにより反転し
たN−置換アミノプロパンジオールに変換させる
ことができる。
本発明で使用する塩基としては、水酸化ナトリ
ウム、水酸化カルシウム、水酸化カリウム、水酸
化バリウム、水酸化アルミニウム、炭酸ナトリウ
ム、炭酸水素ナトリウム、炭酸カリウム、炭酸カ
ルシウム、炭酸バリウム等の無機塩基が使用でき
る。溶媒としては水が好ましいが、ベンゼン、ト
ルエンなどの芳香族炭化水素;メタノール、エタ
ノールなどのアルコール類;塩化メチレン、クロ
ロホルムなどのハロゲン化炭化水素;ジオキサン
エチルエーテル、テトラヒドロフランなどのエー
テル類およびジメチルフオルムアミド、ジメチル
スルホキシド、アセトニトリル等の極性溶媒と水
との混合系も使用できる。
反応は均一系でも不均一系でも進行し、反応温
度は室温から使用する溶媒の還流温度まで選択す
ることができる。好ましくは室温から約50℃の範
囲である。
本反応において塩基を使用する場合は、反応系
の規定度が約3規定まで使用できる。これらの反
応条件で本反応を実施すると、反応は数時間から
48時間の範囲で終了できる。反応後、反応液を水
酸化ナトリウム水溶液などのアルカリ水溶液でア
ルカリ性にして、塩化メチレンや酢酸エチルなど
の有機溶媒で反応物を抽出できる。分液した有機
層を減圧下に濃縮することにより目的とする立体
配置が反転した光学活性N−置換アミノプロパン
ジオールが得られる。目的物が結晶の場合は、析
出する結晶を過することにより容易に取得する
ことができる。必要によつてはアセトン等の溶媒
で再結晶し精製することもできる。また、目的物
が液体の場合は、そのままでは純度の高いものが
得られるが、必要に応じて蒸留あるいはカラムに
より精製することもできる。
以下、実施例により本発明を詳細に説明する。
参考例 1
(5R)−3−t−ブチル−5−ヒドロキシメチ
ル−オキサゾリジン−2−オン〔a〕20 D=−44.7°
(C=1、CHCl3)3.0gを6N−NaOH 30mlに溶
かし、95℃で3時間撹拌する。反応後、反応液を
塩化メチレンで抽出する。塩化メチレン層を無水
硫酸マグネシウムで乾燥する。塩化メチレンを減
圧留去すると(R)−3−t−ブチルアミノ−1,
2−ジヒドロキシプロパンの白色結晶2.4gが得
られる。

57.6°(C=1、CHCl3)4.0gを6N−NaOH 30ml
にとかし、90℃で3時間撹拌する。反応後、反応
液を塩化メチレンで抽出する。塩化メチレン層を
無水硫酸マグネシウムで乾燥する。塩化メチレン
を減圧留去すると(R)−3−イソプロピルアミ
ノ−1,2−ジヒドロキシプロパンの液体3.0g
が得られる。

実施例 1
参考例1で得られた(R)−3−t−ブチルア
ミノ−1,2−ジヒドロキシプロパン2.4gを臭
化水素−酢酸溶液(25%)40mlに加え、室温で3
時間撹拌する。反応後、反応液を減圧濃縮すると
結晶が析出する。この結晶をエーテルで洗浄して
(R)−2−アセトキシ−1−ブロモ−3−t−ブ
チルアミノプロパンのHBr塩4.9gが得られる。
(R)−2−アセトキシ−1−ブロモ−3−t−ブ
チルアミノプロパンのHBr塩2gをメタノール
10mlに溶かし、この溶液にナトリウムメトキシド
(28%メタノール溶液)2.5gを冷却しながら滴下
する。滴下後30分撹拌する。反応後、反応液を減
圧濃縮する。水20mlを加え塩化メチレンで抽出す
る。塩化メチレン層を無水硫酸マグネシウムで乾
燥する。塩化メチレンを減圧留去すると(R)−
3−t−ブチルアミノ−1,2−エポキシプロパ
ンの液体が得られる。これを全量、塩化メチレン
5mlにとかし、さらに水5ml加えたものに、クロ
ル炭酸エチル0.8gを塩化メチレン5mlにとかし
た溶液と炭酸カリウム1.1gを水5mlにとかした
溶液を冷却下、同時に滴下し、室温で3時間撹拌
する。反応後、反応液に2N NaOH 10mlを加え
て塩化メチレンで抽出する。塩化メチレン層を無
水硫酸マグネシウムで乾燥する。塩化メチレンを
減圧留去すると(R)−N−エトキシカルボニル
−3−t−ブチルアミノ−1,2−エポキシプロ
パン1.1gの液体を得る。
得られた(R)−N−エトキシカルボニル−3
−t−ブチルアミノ−1,2−エポキシプロパン
1.0gをNaOHでPH12に調製した水溶液に加え、
室温で7時間撹拌する。反応後、反応液に水酸化
ナトリウムを加え、塩化メチレンで抽出する。塩
化メチレン層を無水硫酸マグネシウムで乾燥す
る。塩化メチレンを減圧留去すると結晶が得られ
る。これをヘキサンで洗浄し取すると(S)−
3−t−ブチルアミノ−1,2−ジヒドロキシプ
ロパンの白色結晶0.65gが得られる。

実施例 2
参考例2で得られた(R)−3−イソプロピル
アミノ−1,2−ジヒドロキシプロパン3.0gを
ピリジン塩酸塩2.9gとピリジン10mlを含む溶液
にとかす。この溶液に冷却下、パラトルエンスル
ホン酸クロライド4.4gを塩化メチレン10mlにと
かした溶液を滴下する。5時間室温で撹拌する。
反応後、反応液に炭酸カリウム1.6gを水50mlに
して加える。この溶液から過剰のピリジンを減圧
留去した後、炭酸カリウム3.7g加え塩化メチレ
ンで抽出する。塩化メチレン層を無水硫酸マグネ
シウムで乾燥する。硫酸マグネシウムを過で除
いた溶液にp−トルエンスルホン酸4.3gを加え、
塩化メチレンを減圧留去すると粗(R)−1,2
−ジヒドロキシ−3−イソプロピルアミノプロパ
ン−1−p−トルエンスルホナートのp−トルエ
ンスルホン酸塩9.4gが得られる。次に、この塩
をイソプロピルアルコールで再結して得た塩の一
部5.8gをメタノール50mlにとかし、この溶液に
ナトリウムメトキシド(28%メタノール溶液)
5.2gを冷却しながら滴下する。滴下後30分撹拌
する。反応後、反応液を減圧濃縮する。水50mlを
加え、塩化メチレンで抽出する。塩化メチレン層
を無水硫酸マグネシウムで乾燥する。塩化メチレ
ンを減圧留去すると(R)−3−イソプロピルア
ミノ−1,2−エポキシプロパンの液体が得られ
る。これを全量、塩化メチレン10mlにとかし、さ
らに水10ml加えたものに、クロル炭酸エチル1.8
gを塩化メチレン10mlにとかした溶液と炭酸カリ
ウム2.2gを水10mlにとかした溶液を、冷却下、
同時に滴下し、室温で3時間撹拌する。反応後、
反応液に2N NaOH 20ml加えて塩化メチレンで
抽出する。塩化メチレン層を無水硫酸マグネシウ
ムで乾燥する。塩化メチレンを減圧留去すると、
(R)−N−エトキシカルボニル−3−イソプロピ
ルアミノ−1,2−エポキシプロパンの液体を
1.8g得る。
得られた(R)−N−エトキシカルボニル−3
−イソプロピルアミノ−1,2−エポキシプロパ
ン1.0gをNaOHでPH12に調整した水溶液に加え、
室温で6時間撹拌する。反応後、反応液に水酸化
ナトリウムを加え、塩化メチレンで抽出する。塩
化メチレン層を無水硫酸マグネシウムで乾燥す
る。塩化メチレンを減圧留去すると液体0.65gを
得る。このものを、さらに蒸留により精製すると
(S)−3−イソプロピルアミノ−1,2−ジヒド
ロキシプロパンの液体0.58gを得る。

参考例1と同様にして得られる(R)−N−エ
トキシカルボニル−3−t−ブチルアミノ−1,
2−エポキシプロパン1.0gを水10mlに加え48時
間室温で撹拌する。反応後、反応液に水酸化ナト
リウムを加え塩化メチレンで抽出する。塩化メチ
レン層を無水硫酸マグネシウムで乾燥する。塩化
メチレンを減圧留去する結晶が得られる。これを
ヘキサンで洗浄し取すると(S)−3−t−ブ
チルアミノ−1,2−ジヒドロキシプロパンの白
色結晶0.65gが得られる。

実施例1と同様にして得られる(R)−N−エ
トキシカルボニル−3−t−ブチルアミノ−1,
2−エポキシプロパン1.0gを0.01N K2CO3 20
mlに加え、10時間室温で撹拌する。反応後、反応
液に水酸化ナトリウムを加え塩化メチレンで抽出
する。塩化メチレン層を無水硫酸マグネシウムで
乾燥する。塩化メチレンを減圧留去すると結晶が
得られる。これをヘキサンで洗浄し取すると
(S)−3−t−ブチルアミノ−1,2−ジヒドロ
キシプロパンの白色結晶0.60gが得られる。

実施例1と同様にして得られた(R)−3−t
−ブチルアミノ−1,2−エポキシプロパン1.0
gをアセトン5mlにとかし、さらに水5mlを加え
たものにクロル炭酸エチル0.8gをアセトン5ml
にとかした溶液と、炭酸カリウム1.1gを水5ml
にとかした溶液を、冷却下、同時に滴加し、室温
で12時間撹拌する。反応後、反応液からアセトン
を減圧留去し、水酸化ナトリウムを加え、塩化メ
チレンで抽出する。塩化メチレン層を無水硫酸マ
グネシウムで乾燥する。塩化メチレンを減圧留去
すると結晶が得られる。これをヘキサンで洗浄
し、取すると(S)−3−tブチルアミノ−1,
2−ジヒドロキシプロパンの白色結晶0.7gが得
られる。
