As a result of intensive study to solve the problems of the conventional method, the present inventor discovered that non-α-vitamin E, which has low biological activity,
We have discovered a method for producing highly purified α-tocopherol using tocopherol as a starting material, which is easy to industrialize and has a high yield. The method of the present invention is to obtain α-tocopherol by hydrogenating non-α-tocopherol and formaldehyde or a polymer thereof in the presence of an alkali metal hydrogen sulfate or an aromatic sulfonic acid. That is, according to the present invention, after dissolving the raw materials non-α-tocopherol and formdehyde in a solvent, hydrogenation is performed using a conventional hydrogenation catalyst such as a palladium-carbon catalyst in the presence of an aromatic sulfonic acid or an alkali metal hydrogen sulfate salt. α-tocopherol conversion easily proceeds simply by heating under pressure, and high purity α-tocopherol can be obtained in high yield. The non-α-tocopherol used as a raw material for the present invention may be any type or a mixture thereof,
Can be used regardless of purity. Moreover, the coexistence of α-tocopherol does not hinder the reaction.
That is, the main starting material is the scum produced in the steam refining of vegetable oils. This deodorized scum is currently obtained primarily from soybean oil. Halaformaldehyde is particularly suitable as the formaldehyde or its polymer used in the present invention, and it is preferable to use it in an excess amount of 3 to 10 times the molar amount relative to non-α-tocopherols. The alkali metal hydrogen sulfate used in the present invention is Na,
Any of K, Li, etc. may be used, but potassium hydrogen sulfate is preferred. In addition, aromatic sulfonic acids are benzene,
Mono- or polysulfonic acid of an aromatic carbon monocyclic compound such as toluene or a salt thereof, preferably monosulfonic acid, particularly toluenesulfonic acid, and more preferably p-toluenesulfonic acid.
and these for non-α-tocopherols
It is convenient to use about 0.3 to 0.5 times the molar amount. Hydrogenation can also be carried out in the presence or absence of a catalyst, and any conventional hydrogenation catalyst can be used, but a palladium-carbon catalyst is most suitable. In the method of the present invention, a non-α-tocopherol and formaldehyde or a polymer thereof react in the presence of an alkali metal hydrogen sulfate or an aromatic sulfonic acid to produce a hydroxymethylated non-α-tocopherol, which is then It is presumed that it is added and converted into α-tocopherol. Therefore, a two-step method can be considered in which a hydroxymethylated product of non-α-tocopherol is produced and isolated or hydrogenated without isolation, but the hydroxymethylated product is unstable, so a two-step method is adopted. Since there is no advantage to
- A non-α-
Converted to tocopherol. In the method of the present invention, an organic solvent inert to the reaction is used, and methanol is particularly preferably used. The amount of solvent used is preferably about equivalent to four times the amount of the starting material to be treated. The optimum reaction temperature is 140° to 170°C.
In reduction by hydrogenation, it is more convenient to carry out under pressure than normal pressure, and the reaction proceeds satisfactorily at the above-mentioned preferred temperature of about 20 kg/cm 2 . The reaction time may be appropriately determined depending on the reaction temperature, the amount of reaction raw materials, etc. [Effects] According to the method of the present invention, α-tocopherol can be obtained very selectively and in high yield without producing any impurities as a by-product, as is clear from the following examples. It can be separated and purified by simple purification means such as vacuum distillation, which is extremely advantageous industrially. [Examples] Next, Examples are given to further specifically explain the present invention, but the present invention is not limited to the following Examples. Example 1 Soybean oil tocopherol concentrate (tocopherol content 97.6%, composition ratio γ-tocopherol; 74
%: δ-tocopherol; 26%) (see Figure 1)
Dissolve 30.0 g in 80 ml of methanol, add 14.0 g of paraformaldehyde, 5.0 g of paratolumenesulfonic acid and 1.1 g of 5% palladium-carbon, and add hydrogen.
The reaction was carried out at a temperature of 160° C. for 10 hours under a pressure of 20 Kg/cm 2 . After the completion of the reaction, the palladium-carbon was filtered off, the filtrate was concentrated under reduced pressure, the obtained oil was extracted with isopropyl ether, washed with water until neutral, and the isopropyl ether layer was concentrated under reduced pressure. Concentration gave 30.2 g of a viscous brown oil. Analysis by high performance liquid chromatography revealed that the oil contained 91.5 g of α-tocopherol, with no β-, γ-, or δ-tocopherol peaks found (see Figure 2). Recovery rate 94.4% Example 2 Soybean oil tocopherol concentrate (total tocopherol content 97.6%, composition ratio γ-tocopherol 74%;
δ-tocopherol 26%) 30.0g in methanol
Dissolve in 120 ml, add 11.0 g of paraformaldehyde, 5.0 g of potassium hydrogen sulfate and 1.0 g of 5% palladium-carbon, and heat at 150°C under a pressure of 20 kg/cm 2 of hydrogen.
The reaction was carried out at a temperature of 12 hours. After the reaction was completed, post-treatment was carried out in the same manner as in Example 1 to obtain 30.6 g of a concentrated oil. When the oil was analyzed by high performance liquid chromatography, the purity of α-tocopherol was found to be
It was 90.9%, and no peaks of β-, γ-, and δ-tocopherol were found. Recovery rate 95.0%. Example 3 Soybean oil tocopherol concentrate (total tocopherol content 62.3%, composition ratio α-tocopherol 10%;
γ-tocopherol 67%; δ-tocopherol 23
%) 30.0g in methanol 120ml, paraformaldehyde 10.0g, paratoluenesulfonic acid
2.5g of palladium and 1.0g of 5% palladium-carbon were added, and hydrogen was added at a temperature of 150℃ under a pressure of 20Kg/ cm2.
The reaction was carried out for 12 hours. After the reaction was completed, post-treatment was carried out in the same manner as in Example 1 to obtain 30.2 g of a concentrated oil. Analysis of the oil by high performance liquid chromatography revealed that it contained 56.1% α-tocopherol, and no peaks for β-, γ-, and δ-tocopherol were found. Recovery rate was 90.6%. Example 4 Soybean oil tocopherol concentrate (total tocopherol content 62.3%, composition ratio α-tocopherol 10%;
γ-tocopherol 67%; δ-tocopherol 23
%) 30.0g in methanol 120ml, paraformaldehyde 10.0g, potassium hydrogen sulfate 2.5g,
and 1.0 g of 5% palladium-carbon were added, and the reaction was carried out at a temperature of 150° C. for 12 hours under a pressure of 20 kg/cm 2 of hydrogen. After the reaction was completed, post-treatment was carried out in the same manner as in Example 1 to obtain 30.3 g of a concentrated oil. Analysis of the oil by high performance liquid chromatography revealed that it contained 56.7% α-tocopherol, and no peaks for β-, γ-, and δ-tocopherol were found. Recovery rate was 91.9%. Reference Example 1 (Supplementary test of Example 2 of JP-A-56-68677) Soybean oil tocopherol concentrate (total tocopherol content 90.3%; composition ratio α-tocopherol 3.0)
%; γ-tocopherol 62.8%; δ-tocopherol 34.2%) (see Figure 3) 12.1g to toluene 102
ml and acetic acid 18.0 ml, add 4.1 g of paraformaldehyde and 1.4 g of 10% palladium-carbon, and heat at 100°C under a pressure of 50 kg/cm 2 of hydrogen.
A time reaction was performed. 10% palladium after the reaction
After carbon was filtered off, the filtrate was washed with water until it became neutral, and the organic solvent layer was concentrated under reduced pressure to obtain 13.0 g of a viscous brown oil. Analysis by gas chromatography revealed that the oil contained 54.5% α-tocopherol, with several % of unreacted non-α-tocopherols (see Figure 4). Recovery rate 65%. Reference example 2 Soybean oil tocopherol concentrated oil (97.6% total tocopherol, composition 74% non-γ-tocopherol; δ-
Tocopherol 26%) 30.0g in methanol 120ml
10.0 g of paraformaldehyde, 5.0 g of phosphorus pentoxide, and 1.0 g of 5% palladium on carbon.
The reaction was carried out at a temperature of 175° C. for 4 hours under a pressure of 40 kg/cm 2 of hydrogen. After the reaction was completed, the palladium-carbon was filtered off, and the filtrate was concentrated under reduced pressure. The resulting oil was extracted with isopropyl ether and washed with water until it became neutral, but separation from the aqueous layer was poor and it took a long time. Needed to stand still.
After washing with water, the isopropyl ether layer was concentrated under reduced pressure to obtain 29.8 g of a viscous brown oil. When this oily substance was analyzed by gas chromatography, it was found that it contained 79.6% α-tocopherol, and no peaks for β-, γ-, or δ-tocopherol were found, but a few peaks were found just before α-tocopherol eluted. % impurity peaks that were difficult to purify by distillation were found. Recovery rate 81.0%.
