〔従来の技術〕
従来知られているペナム誘導体は比較的広範囲
な抗菌スペクトルを有しているが、耐性菌に対す
る効果が弱く必ずしも満足すべきものではない。
〔発明が解決しようとする問題点〕
このような状況下において、広範囲な抗菌スペ
クトルを有し、かつβ−ラクタマーゼに対し安定
で、耐性菌に対しても強い抗菌活性を示すペナム
誘導体の開発が望まれていた。
〔問題点を解決する手段〕
本発明者らは、上記問題点を解決すべく鋭意研
究を行つた結果、一般式〔〕で表わされるペナ
ム誘導体およびその塩が広範囲な抗菌スペクトル
を有し、グラム陽性菌、グラム陰性菌に対し優れ
た抗菌活性を示し、かつβ−ラクタマーゼに対し
安定な性質を有すると同時に、低毒性であつて人
および動物の疾病に対し優れた治療効果を発揮す
ること並びに他のペナム誘導体へ導く中間体とし
ても有用であることを見出し、本発明を完成する
に至つた。
本発明の一般式〔〕で表わされるペナム誘導
体およびその塩はそのテトラゾリル基がペナム環
の2位に直接結合しているところに構造上の特徴
を有する。
以下、さらに本発明を詳細に説明する。
本発明において特にことわらない限り、“低級
アルキル”とは、直鎖または分枝鎖状C1〜5アルキ
ル、たとえば、メチル、エチル、n−プロピル、
イソブチルなどを意味する。
R1におけるアミノ保護基としては、後述する
アミノ保護基が挙げられ、またアシル基としては
ペニシリンおよびセフアロスポリン分野で通常使
用されているもの、たとえば、ホルミル、α−ア
ミノ−α−フエニルアセチル、α−フエニルアセ
チル、α−フエノキシアセチル、α−(2−チエ
ニル)アセチル、α−(3−ブロモフエニル)ア
セチル、α−カルボキシ−α−フエニルアセチ
ル、α−(1−テトラゾリル)アセチル、α−(5
−メトキシ−2−チエニル)アセチル、α−シア

式〔〕で表わされる化合物を得るには、通常
ペニシリンおよびセフアロスポリン分野で用い
られる反応と同様の条件で実施され、たとえ
ば、トリチル基を用いて保護する場合、ザ・ジ

(ウ) アミド化
一般式〔〕で表わされる化合物もしくはそ
の塩またはそれらのカルボキシル基における反

物またはその塩を反応させ、一般式〔〕で表
わされる化合物またはその塩を得るには、当該
分野で用いられるアミド化と同様の条件で実施
され、たとえば、特開昭50−64295号などに記
載の方法またはそれに準じた方法によつて行う
ことができる。
(エ) イミノクロル化
一般式〔〕で表わされる化合物に塩基の存
在下で、ハロゲン化剤を反応させることによ
り、一般式〔〕で表わされる化合物を得るこ
とができる。ここで用いられるハロゲン化剤と
しては、たとえば、ホスゲン、トリクロロメチ
ルクロロホルメート、塩化チオニル、五塩化リ
ンなどが挙げられ、ホスゲンまたはトルクロロ
メチルクロロホルメートが特に好ましい。また
使用される塩基としては、たとえば、トリメチ
ルアミン、トリエチルアミン、ジイソプロピル
エチルアミン、N,N−ジメチルアニリン、N
−メチルモルホリン、ピリジンなどが挙げら
れ、特にピリジンが好ましい。この反応を溶媒
中で行う場合、反応に不活性なものであれば特
に限定されないが、具体的には、塩化メチレ
ン、クロロホルム、四塩化炭素、テトラヒドロ
フラン、ジオキサンなどが挙げられ、特に塩化
メチレンが好ましい。
ハロゲン化剤の使用量は一般式〔〕で表わ
される化合物に対して1〜3倍モル程度、塩基
の使用量はハロゲン化剤に対して1.0〜1.2倍モ
ル程度である。この反応は−20〜30℃、好まし
くは25℃で2〜5時間実施すればよい。
このようにして得られた一般式〔〕で表わ
される化合物は単離せずにつぎの反応に付すこ
とができる。
(オ) カルバミル化および脱水
一般式〔〕で表わされる化合物またはその
塩をカルバミル化して一般式〔〕で表わされ
る化合物を得、ついでこれを脱水反応に付して
一般式〔XI〕で表わされる化合物を得るには、
通常のカルバミル化および脱水反応を用いるこ
とができ、たとえば、ザ・ジヤーナル・オブ・
アメリカン・ケミカル・ソサエテイー(The

54581号などに記載の条件もしくはそれらに準
じた方法によつて行うことができる。
以上説明した各工程において得られる化合物ま
たはそれらの塩に異性体などが存在する場合、通
常の方法でそれぞれ単離および精製することがで
きる。
また、アシル化反応において、R1aの基中に反
応に活性な基が存在するときは、反応に際し、通
常の保護基で任意に保護しておくこともでき、反
応後常法によりその保護基を脱離させることもで
きる。

参考例 1
(1) (3S,5R,6R)−3−(p−ニトロベンジル
オキシカルボニル)−7−オキソ−6−フエニ
ルアセタミド−4−チア−1−アザビシクロ
〔3.2.0〕ヘプタン14.10gを塩化メチレン282ml
に懸濁させ、N,N−ジメチルアニリン12.16
mlを加え−60℃に冷却する。この中に、五塩化
リン9.99gを加え−40〜−30℃で1時間反応さ
せる。ついで、反応液を−50℃まで冷却し、無
水メタノール90mlを加え0℃まで昇温させ、30
分間反応させた後、水100mlを加え、同温度で
30分間撹拌する。析出晶を取し、塩化メチレ
ン42mlおよび水28mlで順次洗浄した後、乾燥さ
せれば、融点99〜100℃(分解)を示す(3S,
5R,6R)−6−アミノ−3−(p−ニトロベン
ジルオキシカルボニル)−7−オキソ−4−チ
ア−1−アザビシクロ〔3.2.0〕ヘプタンの塩
酸塩10.10g(収率87.8%)を得る。
(2) (3S,5R,6R)−6−アミノ−3−(p−ニ
トロベンジルオキシカルボニル)−7−オキソ
−4−チア−1−アザビシクロ〔3.2.0〕ヘプ
タンの塩酸塩9.90gを酢酸エチル100mlおよび
水75mlに懸濁させ、炭酸水素ナトリウムでPH
7.0に調整する。有機層を分取し、飽和食塩水
30mlで洗浄した後、無水硫酸マグネシウムで乾
燥させる。減圧下に溶媒を留去し、得られた残
留物を塩化メチレン100mlに溶解させ、0℃に
冷却する。この中に、トリエチルアミン5.46ml
を加えた後、トリチルクロリド11.40gを加え、
0℃で3時間反応させる。ついで、反応液に水
50mlを加え、有機層を分取した後、飽和食塩水
40mlで洗浄し、無水硫酸マグネシウムで乾燥さ
せる。ついで、減圧下に溶媒を留去し、得られ
た残留物をカラムクロマトグラフイー[メルク
製シリカゲル60、No.7734、溶出溶媒;ベンゼ
ン:酢酸エチル=100:1〜50:1(容量比)〕
で精製すれば、アモルフアス状の(3S,5R,
6R)−6−トリチルアミノ−3−(p−ニトロ
ベンジルオキシカルボニル)−7−オキソ−4
−チア−1−アザビシクロ〔3.2.0〕ヘプタン
10.40g(収率67.5%)を得る。

る。ついで、反応液を1N塩酸でPH7.0に調整
し、減圧下に溶媒を留去る。得られた残留物に
水170mlおよびジエチルエーテル43mlを加え、
水層を分取した後、酢酸エチル170mlを加え、
1N塩酸でPH2.0に調整する。有機層を分取し、
水50mlおよび飽和食塩水40mlで順次洗浄した
後、無水硫酸マグネシウムで乾燥させる。つい
で、減圧下に溶媒を留去すれば、融点159〜165
℃(分解)を示す(3S,5R,6R)−3−カル
ボキシ−7−オキソ−4−チア−6−トリチル
アミノ−1−アザビシクロ〔3.2.0〕ヘプタン
4.00g(収率61.8%)を得る。

1時間反応させた後、水20mlを加え、有機層を
分取る。さらに、有機層を水15mlおよび飽和食
塩水10mlで順次洗浄した後、無水硫酸マグネシ
ウムで乾燥される。ついで、減圧下に溶媒を留
去し、得られた残留物をカラムクロマトグラフ
イー〔メルク製シリカゲル60、No.7734、溶出溶
媒;ベンゼン:酢酸エチル=20:1〜10:1
(容量比)〕で精製すれば、(3S,5R,6R)−3
−〔N−(2−メトキシカルボニルエチル)カル

溶解させ、−65℃に冷却する。これにN,N−
ジメチルアニリン9.94mlおよび五塩化リン5.31
gを順次添加し、−60〜−50℃で1時間反応さ
せた後、無水メタノール48mlを加え、1.5時間
を要して室温まで昇温する。ついで、氷冷下で
反応液に水100mlを加え、10分間撹拌した後、
炭酸水素ナトリウムでPH7.0に調整する。有機
層を分取し、飽和食塩水100mlで洗浄した後、
無水硫酸マグネシウムで乾燥させる。この溶液
に氷冷下、ピリジン1.59mlを加え、ついで、ト
リチルクロリド5.47gを10分間を要して分割添
加し、同温度で20分間撹拌する。反応液を水80
mlおよび飽和食塩水60mlで順次洗浄した後、無
水硫酸マグネシウムで乾燥させる。減圧下に溶
媒を留去し、得られた残留物をカラムクロマト
グラフイー〔和光シリカゲルC−200、溶出溶
媒;ベンゼン:酢酸エチル=50:1〜30:1

下に溶媒を留去すれば、アモルフアス状の

ン5.43mlを加え−60℃に冷却する。さらに五塩
化リン4.46gを加え、−60〜−50℃で1時間反
応させる。ついで、無水メタノール25mlを−50
℃で加え、0℃まで昇温させ、1時間反応させ
る。水20mlを加え、炭酸水素ナトリウムでPH
1.0に調整し、水層を分取する。さらに、有機
層に水5.0mlを加え水層を分取し、これを先の
水層と合し、これに塩化メチレン20mlを加え、
炭酸水素ナトリウムでPH7.5に調整する。有機
層を分取し、さらに、水層を塩化メチレン10ml
ずつで10回、抽出操作を繰り返す。抽出液と先
の有機層を合し、無水硫酸マグネシウムで乾燥
させた後、減圧下に溶媒を留去する。得られた
残留物にジエチルエーテルを加え、結晶化させ
れば、融点117〜121℃(分解)を示す(3R,
5R,6R)−6−アミノ−3−シアノ−7−オ

シクロ〔3.2.0〕ヘプタン40mgをN,N−ジメチ
ルホルムアミド1.5mlに懸濁させ、トリエチルア
ミン0.026mlを加えた後、−10℃まで冷却する。一
方、D−α−(4−エチル−2,3−ジオキソ−
1−ピペラジンカルボキサミド)フエニル酢酸60
mgを塩化メチレン2.0mlに懸濁させ、−20℃でN−
メチルモルホリン0.023mlおよびクロル炭酸エチ
ル0.018mlを加え、−30〜−20℃で1時間反応させ
る。−10〜−5℃でこの反応液を先に調整したN,
N−ジメチルホルムアミド溶液中へ滴下する。滴
下終了後、同温度で1時間反応させた後、メタノ
ール1.0mlを加え、室温まで昇温させ、同温度で
30分間撹拌する。ついで、減圧下に溶媒を留去
し、得られた残留物に水5.0ml、酢酸エチル5.0ml
を加え、1N塩酸でPH2.0に調整し、有機層を分取
する。さらに水層を酢酸エチル3mlで3回抽出
し、抽出液の先の有機層と合わせ、これに水5.0
mlを加え、炭酸水素ナトリウムでPH7.5に調整す
る。水層を分取し、酢酸エチル10mlを加え、1N
塩酸でPH2.0に調整し、有機層を分取する。さら
に水層を酢酸エチル5.0mlで2回抽出し、抽出液
を先の有機層と合わせ、これを飽和食塩水5mlで
洗浄した後、無水硫酸マグネシウムで乾燥する。
ついで、減圧下に溶媒を留去し、得られた残留物
にジエチルエーテルを加え、結晶を取すれば、

実施例 10
(1) (3R,5R,6R)−3−〔1−(2,4−ジメ
トキシベンジル)テトラゾール−5−イル〕−
7−オキソ−4−チア−6−アミノ−1−アザ
ビシクロ〔3.2.0〕ヘプタンのp−トルエンス
ルホン酸塩700mgを塩化メチレン15mlおよび水
10mlの混合溶媒に懸濁させ、飽和炭酸水素ナト
リウム水溶液でPH7.0に調整し、有機層を分取
する。有機層をさらに飽和食塩水5mlで洗浄し
た後、無水硫酸マグネシウムで乾燥させる。
(2) 一方、D−α−(4−エチル−2,3−ジオ
キソ−1−ペピラジンカルボキサミド)フエニ
ル酢酸440mgを塩化メチレン4.5mlに懸濁させ、
−20℃でN−メチルモルホリン0.17mlおよびク
ロル炭酸エチル0.14mlを加え、−20〜−15℃で
1時間撹拌する。−15〜−10℃でこの反応液を
(1)の溶液中へ滴下した後、この混合液を30分間
を要して室温まで昇温する。ついで、水10mlを
加え、飽和炭酸水素ナトリウム水溶液でPH7.0
に調整し、有機層を分取する。この有機層に水
10mlを加え、1N塩酸でPH2.5に調整し、有機層
を分取する。水10mlおよび飽和食塩水10mlで順
次洗浄した後、無水硫酸マグネシウムで乾燥さ
せ、減圧下に溶媒を留去する。得られた残留物
をカラムクロマトグラフイー〔和光シリカゲル
C−200、溶出溶媒;ベンゼン:酢酸エチル=
1.2(容量比)〕で精製すれば、(3R,5R,6R)
−6−〔D−α−(4−エチル−2,3−ジオキ
ソ−1−ペピラジンカルボキサミド)フエニル
アセタミド〕−3−〔1−(2,4−ジメトキシ
ベンジル)テトラゾール−5−イル〕−7−オ
キソ−4−チア−1−アザビシクロ〔3.2.0〕
ヘプタン650mg(収率74.8%)を得る。
融点:172〜175℃(分解)(再結溶媒:塩化メ

実施例 11
(3R,5R,6R)−6−〔D−α−(4−エチル
−2,3−ジオキソ−1−ペピラジンカルボキサ
ミド)フエニルアセタミド〕−3−〔1−(2,4
−ジメトキシベンジル)テトラゾール−5−イ
ル〕−7−オキソ−4−チア−1−アザビシクロ
〔3.2.0〕ヘプタン500mgを、あらかじめ1N塩酸お
よび1M酢酸ナトリウムにてPH3.0に調整した緩衝
溶液1mlとアセトニトリル9mlの混合溶媒に溶解
させる。室温で硝酸セリウム(IV)アンモニウ
ム1.24gを加え、炭酸水素ナトリウム水溶液でPH
0.7〜1.0に調整し、10分間反応させる。ついで、
酢酸エチル30mlおよび水15mlを順次加え、1N塩
酸でPH2.0に調整し、有機層を分取する。さらに
水層を酢酸エチル5mlで2回抽出し、抽出液を先
の有機層と合し、これに水10mlを加え、炭酸水素
ナトリウムでPH7.5に調整する。水層を分取し、
酢酸エチル10mlを加え、1N塩酸でPH2.0に調整
し、有機層を分取する。さらに、水層を酢酸エチ
ル5mlで2回抽出し、抽出液を先の有機層と合
し、これを飽和食塩水10mlで洗浄した後、無水硫
酸マグネシウムで乾燥させる。ついで、減圧下に
溶媒を留去し、得られた残留物に酢酸エチル2ml
を加え、結晶化させた後、取すれば、(3R,
5R,6R)−6−〔D−α−(4−エチル−2,3
−ジオキソ−1−ペピラジンカルボキサミド)フ
エニルアセタミド〕−7−オキソ−3−(5−テト
ラゾリル)−4−チア−1−アザビシクロ
〔3.2.0〕ヘプタン97mg(収率25%)を得る。
この化合物を等量の重炭酸水素ナトリウムを含
む水に溶解させ、凍結乾燥すれば、(3R,5R,
6R)−6−〔D−α−(4−エチル−2,3−ジオ
キソ−1−ペピラジンカルボキサミド)フエニル
アセタミド〕−7−オキソ−3−(5−テトラゾリ
ル)−4−チア−1−アザビシクロ〔3.2.0〕ヘプ
タンのナトリウム塩を得る。
なお、この化合物の物性(IR値)は、実施例
8で得られたものと一致した。
実施例 12
(3R,5R,6R)−3−シアノ−7−オキソ−
4−チア−6−トリチルアミノ−1−アザビシク
ロ〔3.2.0〕ヘプタン1.0gをクロロホルム20mlに
溶解させ、N−メチルピペリジン塩酸塩0.65g、
N−メチルピペリジン0.90mlおよびナトリウムア
ジド0.31gを順次添加し、30分間加熱還流する。
ついで、室温まで冷却した後、反後液に水10mlを
加え、1N塩酸でPH1.0に調整し、有機層を分取す
る。さらに、有機層を水10mlおよび飽和食塩水5
mlで順次洗浄した後、無水硫酸マグネシウムで乾
燥させる。減圧下に溶媒を留去すれば、(3R,
5R,6R)−7−オキソ−3−(5−テトラゾリ
ル)−4−チア−6−トリチルアミノ−1−アザ
ビシクロ〔3.2.0〕ヘプタン1.0g(収率91%)を
結晶として得る。
この化合物の物性値(融点、IR)は、実施例
6で得られた化合物と一致した。[Table]
