【発明の詳細な説明】[Detailed description of the invention]

(産業上の利用分野) 本発明はポリウレタン相中に珪素重合体が凝集
偏在した粒子構造を形成することなく、均一に分
散して成る抗血栓性材料に関するものである。 (従来の技術) ポリシロキサン−ポリウレタン系の抗血栓性物
質としては、ポリウレタンにポリシロキサンを相
分離状態で分散させた組成物〔特開昭57−
139351、139352号公報;今井庸二、渡辺昭彦、人
工臓器、第11巻、209頁(1982年)〕やポリウレタ
ンとポリシロキサンを、水の存在下で、室温架橋
剤と反応させて安定なエマルジヨンとして抗血栓
性物質(特願昭57−41865号)などが提案されて
いる。 (発明が解決しようとする問題点) ポリウレタンとポリシロキサンをブレンドした
素材では、相分離状態のポリシロキサンは通常
0.2〜10μ程度の凝集偏在した光学顕微鏡オーダー
で観察できる分散単位であり、この分散粒子形は
不均一であり、抗血栓性も満足できるものでな
く、抗血栓性のレベルも一定しない。 (問題点を解決するための手段) 本発明は上記の問題点を解決した新規な抗血栓
性材料の提供を目的とするものである。 本発明になる抗血栓性材料は、ポリウレタンと
珪素重合体とから構成され、珪素重合体がポリウ
レタン相中に凝集偏在した分離単位を形成するこ
となく、均一に分散して成る抗血栓性材料であ
る。 本発明の抗血栓材料は、第1図に示した従来の
ポリウレタンと珪素重合体をブレンドして成る該
材料(超薄切片の透過電子顕微鏡写真)で確認さ
れる珪素重合体がミクロンオーダーの凝集偏在し
た分散単位は存在せず、珪素重合体は第2図に示
した如く均一に分散したものである。 本発明の抗血栓性材料性はポリウレタンと珪素
重合体とを混合することによつては得ることはで
きない。本発明の該材料を得る一つの方法はポリ
ウレタン溶液に縮合重合可能な低分子含珪素化合
物を添加混合して成る溶液から流延などにより被
膜を形成させる方法である。 本発明で使用するポリウレタンは特に限定され
ず、ポリエステル系ポリウレタンおよびポリエー
テル系のポリウレタンであつて良い。 ポリエステル系ポリウレタンは例えばエチレン
グリコール、ジエチレングリコール等のグリコー
ルまたはトリメチロールプロパン、グリセリン等
の多価アルコールとアジビン酸、コハク酸等の多
価カルボン酸との間で末端水酸基のポリエステル
を合成し、これとエチレンジイソシアネート、
2,4−トリレンジイソシアネート、4,4−ジ
フエニルメタンジイソシアネート等のポリウレタ
ンの製造に従来から使用されているイソシアネー
ト基含有化合物とを反応させて両末端がイソシア
ネート基であるプレポリマーとし、公知の鎖延長
剤で鎖延長して調製してもよい。 また、ポリエーテル系ポリウレタンはたとえば
ポリエチレングリコール、ポリプロピレングリコ
ール又はこれらの共重合物テトラメチレングリコ
ール、ポリテトラメチレングリコールあるいはア
ルキレンオキシドとプロピレングリコール、1,
2,6−ヘキサントリオール等の多価アルコール
とを反応させて得られるポリオールと前記のイソ
シアネート基含有化合物とを反応させて末端イソ
シアネートのプレポリマーとし、これらを公知の
鎖延長剤で分子鎖を伸長させて調製してもよい。 尚、本発明に使用するポリウレタンを調製する
に際し、鎖延長剤としてジアミン、たとえばエチ
レンジアミン、ジエチレンジアミン、ヘキサメチ
レンジアミン等を用いてポリウレタンウレアとし
てもよい。又鎖延長剤としてはエチレングリコー
ル、ブタンジオール等の公知のジオールを用いて
もよいことは勿論である。 本発明で使用する縮合重合可能な低分子の含珪
素化合物としては、シリコーン樹脂やゴムの製造
に使用される室温架橋剤やシランカツプリング剤
などの内で、水によつて活性化されて縮合重合し
て架橋珪素重合体に転化し得る一分子中に少なく
とも2ケの官能基を有する化合物が特に好まし

ヒドロフラン、ジオキサンなどの環状エーテル類
やジメチルホルムアミド、ジメチルアセトアミ
ド、ジメチルスルホキシドのような1種以上の有
機極性溶媒であり、本抗血栓性材料の医療器への
使用の観点から、塗布膜からの溶剤の除去(蒸
発、水洗など)が容易なものが好ましい。 また上記の溶媒に程度の大きい影響を及ぼさな
い範囲で少量の他の溶剤、例えばアルコールやア
セトンなどが含まれることは差しつかえない。 溶液中のポリウレタンの濃度は1〜50重量%で
あり、好ましくは3〜40重量%、更に好ましくは
5〜30重量%である。この範囲よりポリウレタン
濃度が低いときは粘度が低くすぎて成膜時に塗布
回数の増加や形成フイルムの極薄などの問題を生
じ、又この範囲を超えると粘度が高すぎてフイル
ム厚の調整が困難となる。 含珪素化合物とポリウレタンを溶解した均一溶
液を、スプレー、流延などの方法により基体上に
塗布し、成膜することにより本発明の抗血栓材料
が得られる。成膜中に含珪素化合物は空気中の水
分により活性化されて架橋ポリシロキサンに転化
される。 本発明の抗血栓性材料は含珪素化合物とポリウ
レタンを溶解した均一な溶液に所定量の水を添加
して、該溶液中で含珪素化合物の縮合を行わせた
後成膜することによつても得ることができる。 本発明の抗血栓性材料、ポリウレタンとポリシ
ロキサンとを混合して得られる該材料とは異な
り、既に記載した如くポリシロキサンは数ミクロ
ンオーダーの凝集偏在した分散単位を形成せずに
ポリウレタン相中に均一に分散している。第1図
〜第4図の電子顕微鏡写真に基づいて本発明の抗
血栓性材料と従来のポリウレタンとポリシロキサ
ンをブレンドして得られる材料との違いを説明す
る。いずれもポリウレタン90重量%とポリシロキ
サン10重量%から構成されているが、第1図及び
第3図は従来のブレンド系の抗血栓性材料であ
り、第2図及び第4図は本発明の抗血栓性材料で
ある。第1図及び第2図は両材料の超薄切片の通
常の電子顕微鏡写真(透過像)であり、第3図及
び第4図は第1図、第2図と同視野のポリシロキ
サンのSi元素の面分析によるSi元素の特性X線
(Kα線)による電子顕微鏡写真である。第3図の
白点部が凝集偏在している部分がポリシロキサン
相であり、第1図の黒い部分に対応する。 このように、従来のブレンド系の抗血線性材料
ではポリシロキサンはポリウレタン相に凝集偏在
して分散しているが、本発明の該材料では、第2
図に示した如くポリシロキサンはこの様な凝集偏
在した分散単位を形成せず、ポリウレタン相に均
一に分散している。 本発明において、均一分散とは、抗血栓性材料
の超薄切片を分析電子顕微鏡を用いて、500Å×
500Å以下の範囲でSi元素分析を行ない、その任
意の部分でのSi濃度差が少なくとも10%以下であ
るような分散を意味しており、この場合、透過像
においても少なくとも500Å以上のSi元素の存在
を示す高電子密度の部分は存在せず(第2図)、
Si元素に関する面分析(第4図)においてSi元素
の存在密度を示す自点が特に偏在しない一様な分
布の結果が得られる。 本発明の抗血栓性材料が何故に上記の如き構造
を有するかは解明されていないが一つの理由とし
ては以下のことが考えられる。 一般にポリウレタンと高分子量ジメチルシリコ
ーンは相溶性が劣るが、本発明に用いる低分子硅
素化合物はポリウレタンとの相溶性が良好であ
り、縮合反応の際、ポリウレタン主鎖間をぬうよ
うにして反応が進行するために生成する珪素重合
体は特に偏在分離することがないのであろう。 本発明の抗血栓性材料は従来のポリウレタン/
ポリシロキサンブレンド系の該材料に比べて、極
めて優れた抗血栓性を示すので、人工心臓;人工
腎臓回路や人工心肺回路のような血液回路;血管
内留置カテーテル、カニユーレ、各種血液バイパ
スチユーブ;血液バツグなどあらゆる血液接触医
療器具の血液接触面を構成するのに最適な材料で
ある。 以下実施例により本発明を具体的に説明する。 実施例 1 ポリテトラメチレングリコール(平均分子量
2500)とメチレンビス(4−フエニルイソシアネ
ート)を用い、連鎖拡張剤としてブタンジオール
を用いて公知の方法で調製したポリエーテル系ポ
リウレタン100重量部を水分7ppm以下にまで脱水
したジオキサン/テトラヒドロフラン混合溶剤
(重量混合比1/2)に35℃で、撹拌下に溶解し、
粘稠な溶液(重合体濃度10重量%)を調製した。
この溶液に蒸留直後のジメチルジアセトキシシラ
ン/メチルトリアセトキシシラン混合物(重量混
合比20/80)をポリウレタン90重量部に対して10
重量部の割合となる様に加え均一に混合した。こ
の溶液をガラス板に流延して被膜を形成せしめ
た。 又、上記の低分子珪素化合物に代えてポリジメ
チルシロキサンを用い、ポリウレタン/ポリジメ
チルシロキサン(90/10重量%)ブレンドの被膜
を形成せしめた。この2種の被膜の超薄切片を作
り電子顕微鏡で観察した。第1図にブレンド系
の、第2図に本発明の材料の透過像の写真を示し
た。又同一切片の面分析によるSi元素の特性X線
による像の写真を第3図(ブレンド系)、第4図
(本発明例)に示した。既に記した通り、第3図
と第4図の比較から、本発明の材料はポリシロキ
サンがポリウレタン相中に凝集偏在した分散単位
を形成することなく均一に分散していることが分
る。 この2種の被膜を用いてリーホワイト試験(臨
床検査法提要−86、金井泉、金井正光編著、金
原出版)を行い、血液凝固時間を測定した。ブレ
ンドでは53分、本発明材料では120分であつた。 実施例 2 ポリエチレングリコール(平均分子量650)と
4,4′−ジフエニルメタンジイソシアネートを用
い、連鎖拡張剤としてプロピレンジアミンを用い
ポリエーテルポリウレタンウレアを調製した。こ
のポリエーテルポリウレタンウレア100重量部を
充分に脱水したジメチルアセトアミドに室温で撹
拌下に溶解させ重合体濃度9.0%の粘稠な溶液と
した。この溶液にジメチルジメトキシシランとメ
チルトリメトキシシラン等の混合物を第1表に示
すような割合で添加し、均一透明な溶液を得た。
実施例1と同様にガラス板上に流延して成膜した
フイルム及び試験砕に塗布してリーホワイト試験
を行つた結果を第1表にまとめた。
