Claims (31)
- 1.ウイルスのコートタンパク質の一部として既定外来ペプチドを含む植物ウイ ルスの集合粒子。
- 2.外来ペプチドが生物学的機能性ペプチドであり、その生物学的適用がそれよ り大きな分子もしくは粒子と組合されたそのペプチドの供与を要求するか又はそ れにより高められる、請求項1に記載のウイルス粒子。
- 3.ペプチドが抗原である、請求項1又は2に記載のウイルス粒子。
- 4.抗原がウイルス抗原である、請求項3に記載のウイルス粒子。
- 5.抗原が動物(ヒトを含む)ウイルス抗原である、請求項4に記載のウイルス 粒子。
- 6.外来ペプチドが植物ウイルスのコートタンパク質の露出表面に組み込まれた 、請求項1〜5のいずれか一項に記載のウイルス粒子。
- 7.植物ウイルスがRNAウイルスである、請求項1〜6のいずれか一項に記載 のウイルス粒子。
- 8.ウイルスコートタンパク質がβ−バレル構造を有する、請求項1〜7のいず れか一項に記載のウイルス粒子。
- 9.外来ペプチドが植物ウイルスのループ接合βシートに挿入された、請求項8 に記載のウイルス粒子。
- 10.植物ウイルスがコモウイルスである、請求項1〜9のいずれか一項に記載 のウイルス粒子。
- 11.コモウイルスがカウピーモザイクウイルス(CPMV)である、請求項1 0に記載のウイルス粒子。
- 12.外来ペプチドが植物ウイルスのβB−βCループに挿入された、請求項9 、10又は11に記載のウイルス粒子。
- 13.挿入された外来ペプチドが現存ループヘの付加である、請求項9〜12の いずれか一項に記載のウイルス粒子。
- 14.挿入された外来ペプチドが現存ループの一部の置換である、請求項9〜1 2のいずれか一項に記載のウイルス粒子。
- 15.外来ペプチドが口蹄疫ウイルス(FMDV)に由来する動物ウイルス抗原 である、請求項1〜14のいずれか一項に記載のウイルス粒子。
- 16.外来ペプチドがヒト免疫不全ウイルス(HIV)に由来する動物ウイルス 抗原である、請求項1〜14のいずれか一項に記載のウイルス粒子。
- 17.外来ペプチドがヒトライノウイルス(HRV)に由来する動物ウイルス抗 原である、請求項1〜14のいずれか一項に記載のウイルス粒子。
- 18.免疫原成分として請求項3〜17のいずれか一項に記載された集合植物ウ イルス粒子を含むヒト又は動物病原体に対する保護用ワクチン。
- 19.免疫原成分として請求項5に記載されたウイルス粒子を含む動物ウイルス ワクチン。
- 20.コートタンパク質についてコードする植物ウイルス核酸を修正するために 外来ペプチドについてコードするヌクレオチド配列を導入し、植物、植物組織、 植物細胞又は原形質体を修正ウイルス核酸で感染させ、修正ウイルスの集合粒子 を収集することからなる、請求項1〜17のいずれか一項に記載された植物ウイ ルス粒子の産生方法。
- 21.導入されるヌクレオチド配列がコートタンパク質の露出部分についてコー ドする植物ウイルス核酸のその部分で押入される、請求項20に記載の方法。
- 22.外来ペプチドについてコードするDNA配列をコートタンパク質の露出部 分についてコードする植物ウイルスのRNAに相当するcDNA中に導入し、こ うして修正されたcDNAからそのRNA転写物を産生させ、必要であれば植物 体で全ウイルス粒子の増殖及び集合に要求されるいずれか他のRNAと一緒に植 物、植物組織、植物細胞又は原形質体を転写物で接種し、修正ウイルスの集合粒 子を収集することからなる、RNA植物ウイルスに適用される、請求項20に記 載の方法。
- 23.外来ペプチドについてコードするDNAを植物ウイルスcDNAから切出 されたDNA断片中に導入し、修正形で植物ウイルスcDNAを再構成するよう に修正断片を組換える、請求項22に記載の方法。
- 24.塵生された修正ウイルス又はそれから抽出されたRNAが追加収量の修正 ウイルスを産生するために植物で継代される、請求項22又は23に記載の方法 。
- 25.コートタンパク質の露出表面に相当する部位で外来ペプチドについてコー ドするDNA配列を含むCPMVコートタンパク質cDNAの断片。
- 26.SstI断片である、請求項25に記載の断片。
- 27.請求項25又は26に記載された断片を含むベクター。
- 28.コートタンパク質の露出表面に相当する部位で外来ペプチドについてコー ドするDNAインサートを有するCPMV M RNAの全鎖長cDNAを含む ベクター。
- 29.請求項25〜28のいずれか一項に記載された断片又はベクターのRNA 転写物。
- 30.請求項29に記載されたキャップ化RNA転写物。
- 31.請求項18又は19に記載されたワクチンを投与することからなる病原体 に対する動物(ヒトを含む)の保護方法。
