Claims (1)
- 【特許請求の範囲】 1 種子特異性植物プロモーターとして作用でき、植物細胞において脂肪酸又は 脂質の生合成と協奏して前記プロモーターの制御下におかれた遺伝子の発現をす ることもできるプロモーターを含む、組換えDNA構築物。 2 前記プロモーターが、明細書に記載した、すなわち、【配列があります】 の、クローンACP05の少なくとも291bpのポリヌクレオチドを含む、請 求項1に記載のDNA構築物。 3 図1に記載したセイヨウアブラナのACP05遺伝子の少なくとも1kbの PstI−BglII5′上流断片を含む、請求項1に記載のDNA構築物。 4 図1に記載のセイヨウアブラナのACP05遺伝子の少なくとも1kbのB amHI−BglII5′上流断片を含む、請求項1に記載のDNA構築物。 5 大腸菌JM101/pAPIGUS(NC I MB40396)に存在す るプラスミドpAPlGUSに存在する種子特異性植物プロモーターを含む、請 求項1に記載のDNA構築物。 6 植物細胞を形質転換するための、好ましくは脂肪酸の種子特異性の生合成を 修飾するための、請求項1乃至5のいずれか1請求項に記載の種子特異性プロモ ーターを含むDNA構築物の使用。 7 その使用によって続いて植物細胞が、種子において特異的に脂肪酸の修飾さ れた生合成が起こる完全な植物に生長する、請求項6に記載の使用。 8 請求項1乃至5のいずれか1請求項に記載の種子特異性プロモーターを含む DNA構築物を、形質転換できる植物細胞に導入し、生じた形質転換された植物 細胞を完全な植物に成育した後に、導入された種子特異性で時間的に調節する植 物プロモーターにより制御された前記遺伝子の一部を生成する構造遺伝子が、植 物の種子において脂肪酸又は脂質の生合成と協奏して発現され、それにより前記 構造遺伝子に対応するタンパク質を生成する、植物細胞を形質転換する方法。 9 前記構造遺伝子が、脂肪酸又はそれに対応する脂質の種子特異性生合成に必 要なタンパク質をコードする、請求項8に記載の方法。 10 苗木(piantlet)を経由して、種子を有する植物にまで植物細胞 を成育させ、生じる、修飾された組成を有する植物油を含有する種子を回収し、 それにより前記植物細胞の又は植物の細胞又は、種子が、請求項1乃至5のいず れか1請求項に記載のDNA構築物を含有することを含む、植物種子油の生成を 修飾する方法。 11 前記DNA構築物が、前記プロモーターの制御下で、脂肪酸生成又は脂質 生成のための生合成経路において活性なタンパク質をコードする遺伝子を含む、 請求項10に記載の方法。 12 前記DNA構築物が、種子特異性アシルキャリヤータンパク質(ACP) プロモーター、好ましくはブラシカ・ナプス(Brassica napus) 由来の種子特異性アシルキャリヤータンパク質(ACP)プロモーター及び、野 生型遺伝子と異なる、ACPをコードする構造遺伝子から本質的に成るプロモー ターを含む、請求項10又は請求項11に記載の方法。 13 前記DNA構築物が種子と相同性(homologous)である種子特 異性植物プロモーターを含有する場合には、前記DNA構築物は前記種子のゲノ ムにおいて前記プロモーターに関して天然部位でない部位に存在する、請求項1 乃至5のいずれか1請求項に記載のDNA構築物を含む種子。 14 前記DNA構築物が、外来性のタンパク質をコードする、興味を有するD NA配列も含み、それによって興味を有するDNA配列が種子特異性植物プロモ ーターの制御下にある、請求項13に記載の種子。 15 前記種子がブラシカ科に属する植物である、請求項13に記載の種子。 16 所望のタンパク質をコードする構造遺伝子を発現し、植物細胞が、前記構 造遺伝子を含む、請求項1乃至5のいずれか1請求項に記載の組換えDNA構築 物を含有し、前記タンパク質の生成の後に任意に植物細胞から前記タンパク質を 単離することを含む、植物細胞、好ましくは種子細胞において所望のタンパク質 を生成する方法。
