A method of measuring using a luminescence measuring method is known. This method utilizes the fact that the amount of ATP contained in cells is proportional to the number of cells, and is a very effective method because the measurement time is short and the sensitivity is high.

That is, in order to measure the amount of ATP in cells using this method, luciferase and luciferin are extracted after extraction using a reagent for extracting ATP contained in cells (herein referred to as "extraction reagent"). By using a biochemiluminescence reaction reagent (hereinafter referred to as "luminescence reagent") that uses microbes to generate minute light generated based on biochemiluminescence using a highly sensitive luminescence detector, for example, a photomultiplier tube. ATP is quantified by measuring and utilizing the fact that the amount of light generated is proportional to the amount of ATP. The amount of cells can be measured from the amount of ATP.

Further, the rotation support shaft 132 is attached by a spring member (not shown) in a counterclockwise direction in FIG. 3 so that the roller 136 comes into contact with the tip portion 12B of the support plate 12 of the container holder 10. It is energized. Therefore, when the container holder 10 is now pulled out from the inside of the housing 2, the roller 136 and the operating lever 134 rotate counterclockwise until the operating lever 134 comes into contact with the guide member 18A. Therefore, the shutter member 130 also rotates counterclockwise, and
As indicated by the alternate long and short dash line, the photomultiplier tube 108 is stopped at a position below the photodetection tip portion 108A.
Thus, when the shutter member 130 is pulled out from the inside of the housing 2, the shutter member 130 causes the photodetection tip portion 1 of the photomultiplier tube 108.
Light that enters 08A can be suppressed.

As the sample, commercially available fermented milk was diluted with a phosphate buffer of pH 7.0 to prepare a lactic acid bacterium suspension of 4.4 cells / ml to 4.4 × 10 6 cells / ml. The number of lactic acid bacteria was measured using BPC-added plate count agar medium (manufactured by Eiken Chemical Co., Ltd.).

ことができる。As described in detail above, the method and apparatus for measuring the ATP amount of cells according to the present invention places the filtration membrane, which captures the cells in the sample, in a dish-shaped container having one opening, Since the photomultiplier tube 108 is installed so as to face the opening of this container and the ATP amount of the cells is measured, the filter membrane after cell capture can be folded or the cells on the filter membrane can be folded. It does not require tedious operations such as washing away the cells, not only improves operability, but also solves the risk of contamination from the outside of the filtration membrane and enables highly accurate measurement of the ATP amount of cells. . In addition, the amount of extraction reagent for extracting ATP from the filtration membrane after cell capture is minimized,
As a result, it is possible to avoid a decrease in concentration efficiency,
Furthermore, the amount of luminescence of the sample can be measured directly without going through the glass tube wall or the like, and therefore the amount of ATP of cells can be measured with high accuracy. Due to these effects, the present invention can greatly improve the lower limit of measurement of cells, and thus can greatly contribute to food hygiene, biotechnology, and other microbial tests.
