【発明の詳細な説明】 〔産業上の利用分野〕 本発明は、感染症の診断システム及び当該システムに用
いるための微生物の検出と微生物の同定の方法に関す
る。

にはこれらの血液検体からの菌の培養の成功率は極めて
低い。しかも、菌血症を疑われた時点で、大量に抗生物
質を投与されている場合には、たとえ血液中に菌が含ま
れていたとしても、増菌・発育できない場合が多く、そ
れ故、C・B陽性になる割合は極めて小さい。

を利用したハイブリダイゼーションによるもの〔特表昭
61−502376号参照〕でその特異性に基づいた正確性の高
い方法等が開発されているが、いずれも、菌の分離及び
増菌培養が不可欠である。

養を菌が発育するまで続けるわけであるので、C・B陽
性になった場合ですら一週間以上かかる場合が多い。従
って、このことが従来法でのC・B陽性での死亡率を高
くしている要因となっている。例えば、「日本感染症学
誌」、vol.58,No.2,pp.122、1984年の報告によれば、血
液培養陽性率が28.6%(163/569件)でも、その内死亡
率が84.6%(138/163件)にまで到っていることが報告
されている。

sis)、リステリア(Listeria)、サルモネラ(Salmone
lla)、ブルセラ(Brucella)、レジオネラ・ニューモ
フィラ(Lationella pneumothila)等があり、本発明の
診断方法に適用可能である。

ら、検出が迅速かつ確実である。

ン法により迅速かつ正確に菌を同定するから、全体とし
て精々1〜2日でずっと的確な検出を実現するという作
用を有し、画期的である。また、菌体成分がほとんど消
化されて検出されない場合でも、その未変性DNA(RNA)
をハイブリダイスし検出できる。

用)に載せ、ピペットの腹等で延ばし、完全に乾くまで
風乾させる。次に、カルノア固定数(エタノール:クロ
ロホルム:酢酸=6:3:1)中にスライドグラスを10分間
浸し、細胞の固定を行う。

ら、室温で10分間の洗浄を2回行い、さらにAP9.5(100
mM Tris・HCl pH9.5,100mM NaCl,10mM MgCl2)中で軽く
振盪させながら室温で10分間の洗浄を3回行う。

実施例2:ビオチン化プローブを用いたインサイテュハイ
ブリダイゼーションによる細胞内外における菌(感染
菌)の検出・同定(以下、「プローブ法」と称する) 上記Aの方法に従って作成した各スライドグラス固定サ
ンプルを、PBS溶液中に室温で10分間以上浸して再水和
を行い、サポニン及びTX−100を各々0.25%(w/v)含む
PBS溶液に10分間軽く振盪させながら室温で10分間処理
する。

2N酸塩を含む生理食塩液で20分間洗う。次に、0.1Mトリ
エタノールアミン−HCl緩衝液(pH 8.0)に0.5%無水酢
酸を含む溶液中に20分間浸す。

ト(Bathesda Resesrch Laboratories)を用いる。

サンプルを調製した。

ず)。

針を与えることができ、これにより、死亡率の低減が期
待される。

発明の診断方法では、患者よりの血液サンプルの採取が
一度で良く厳密な無菌操作も不要で、しかもその採取量
も例えば、デキストラン法では約1mlと極めて少量で良
いので、患者に及ぼす肉体的に或いは精神的な苦痛が低
減できる。

【特許請求の範囲】[Claims] 【請求項1】感染症原因菌の同定方法であって、下記工
程、すなわち: (a)血液、腹水、透析液、尿、および膿からなるグル
ープから選択された生体成分から調製した食細胞を多量
に含む検体を固定し、 (b)感染症原因菌の遺伝子断片を含む細菌DNAプレー
ブと、前記検体とのハイブリダイゼーションを行い、お
よび (c)前記ハイブリダイゼーションにて、前記食細胞画
分に対してハイブリッドを形成した前記細菌DNAプロー
ブを検出する、 工程を含むことを特徴とする感染症原因菌の固定方法。

れた標識分子を結合したビオチンを接触させ、および (d)前記アビジン様蛋白質を介して前記検体に結合し
ている前記ビオチンに結合した前記標識分子を検出す
る、 工程を含むことを特徴とする感染症原因菌の検出方法。
[Claim 3] A method for detecting infectious disease-causing bacteria, comprising the following steps: (a) immobilizing a specimen containing a large amount of phagocytes prepared from a biological component selected from the group consisting of blood, ascites, dialysis fluid, urine, and pus; (b) contacting the specimen with an avidin-like protein; (c) contacting the specimen contacted with the avidin-like protein with biotin bound to a labeling molecule selected from the group consisting of enzymes, antibodies, dyes, and gold; and (d) detecting the labeling molecule bound to the biotin bound to the specimen via the avidin-like protein.

ブを検出する、 工程をさらに含む請求項3に記載の方法。
