Claims (14)
- 1.グラム陽性菌の表面上に天然では知見されず、且つ選択的相互作用し得る第 2のアミノ酸配列をコードする第2のDNAフラグメントに操作可能に結合した 、グラム陽性宿主中で操作可能なシグナルペプチドとして操作する第1のアミノ 酸配列をコードする第1のDNAフラグメントを含む組換えDNA配列であって 、第2のDNAフラグメントは、細胞壁拡張及び膜固定配列としてグラム陽性宿 主中に操作可能な第3のアミノ酸配列をコードする第3のDNAフラグメントに 操作可能に結合している該組換えDNA配列。
- 2.前記第2のアミノ酸配列が抗原的に作用し得ることを特徴とする請求項1に 記載の組換えDNA配列。
- 3.前記第2のアミノ酸配列が抗体(免疫グロブリン)またはその活性フラグメ ントであることを特徴とする請求項1に記載の組換えDNA配列。
- 4.前記第3のDNAフラグメントがブドウ球菌タンパク質Aまたは連鎖球菌タ ンパク質Gの細胞壁拡張及び膜固定領域をコードすることを特徴とする請求項1 〜3のいずれか1項に記載の組換えDNA配列。
- 5.前記第1のDNAフラグメントがグラム陽性菌細胞から由来することを特徴 とする請求項1〜4のいずれか1項に記載の組換えDNA配列。
- 6.前記第3のDNAフラグメントがブドウ球菌タンパク質Aの細胞壁拡張及び 膜固定領域をコードすることを特徴とする請求項4または5に記載の組換えDN A配列。
- 7.前記第1のDNAフラグメントがブドウ球菌タンパク質Aのシグナルペプチ ドをコードすることを特徴とする請求項6に記載の組換えDNA配列。
- 8.前記第2のDNAフラグメントガワクチン接種法または抗体の産生に有用な 免疫応答を誘引し得るアミノ酸配列をコードすることを特徴とする請求項1〜7 のいずれか1項に記載の組換えDNA配列。
- 9.請求項1〜8のいずれか1項に記載であり、グラム陽性菌宿主中で複製し得 る組換えDNA配列を含む発現ベクターまたはプラスミド。
- 10.請求項9のベクターまたはプラスミドにより形質転換されたか、請求項1 〜8のいずれか1項に記載の組換えDNA配列を含有するグラム陽性菌細胞。
- 11.一種の特異的な組換え表面レセプター構造体を保持する細胞を同定及び/ または単離し得る特異的相互作用パートナーで細胞の前記異種集団を相互作用さ せる段階を含む、種々の組換え表面レセプター構造体を保持する細胞の異種集団 から請求項10に記載のグラム陽性菌細胞を選択的単離または同定するための方 法。
- 12.前記レセプター構造体が抗体またはその活性フラグメントにより構築され ることを特徴とする請求項11に記載の方法。
- 13.前記特異的構造体を保持する細胞を単離するために固定形の前記相互作用 パートナーを使用することを特徴とする請求項11または12に記載の方法。
- 14.前記相互作用パートナーが固相支持体上で固定されることを特徴とする請 求項13に記載の方法。
