Abstract
Description
Claims (14)
- 1.Campylobacter jejuniのゲノムの核酸塩基配列と特異 的にハイブリダイズするヌクレオチド配列において、ヌクレオチド配列No.1 と、これと相補的な配列と、この配列の少なくとも1つの塩基の突然変異、挿入 、欠失または置換に起因して変化した配列との中から選択されることを特徴とす る塩基配列。
- 2.請求項1に記載のヌクレオチド配列の全部または一部を含むヌクレオチド配 列。
- 3.ヌクレオチド配列No.2と、これと相補的な配列と、この配列の少なくと も1つの塩基の突然変異、挿入、欠失または置換に起因して変化した配列との中 から選択される請求項1に記載のヌクレオチド配列を有するヌクレオチド配列。
- 4.請求項1〜3のいずれか一項に記載の配列の1つの増幅生成物。
- 5.請求項1〜3のいずれか一項に記載のヌクレオチド配列を含むことを特徴と するクローニングベクター。
- 6.請求項1〜4のいずれか一項に記載のヌクレオチドより選択された少なくと も20の連続するヌクレオチドを有することを特徴とするCampylobac ter jejunjの核酸に対して特異的な核酸プローブ。
- 7.ヌクレオチド配列No.1と、ヌクレオチド配列No.2と、これらの相補 的な配列と、これらの配列の少なくとも1つの塩基の突然変異、挿入、欠失また は置換に起因して変化した配列との中から選択される請求項6に記載のCamp ylobacter jejuniの核酸に対して特異的な核酸プローブ。
- 8.担体上に固定されてキャプチャープローブとして使用される請求項6または 7に記載の核酸プローブ。
- 9.請求項1〜4のいずれか一項に記載の配列より選択されるヌクレオチドを1 8〜30個、好ましくは18〜22個有するオリゴヌクレオチドペアで構成され ることを特徴とするCampylobacterjejuniの核酸配列の増幅 に特異的なオリゴヌクレオチドプライマーペア。
- 10.下記配列を有するオリゴヌクレオチドペアで構成されることを特徴とする 請求項9に記載のCampylobacter jejunjの核酸配列の増幅 に特異的なオリゴヌクレオチドプライマーペア:5,【配列があります】3, 5,【配列があります】3,
- 11.下記段階を特徴とする生物試料中のCampylobacter jej uniの存在を検出する方法: a)生物試料と請求項9または10に記載のプライマーペアとをプライマーがC ampylobacter jejuniの核酸にハイブリダイズするような条 件下で接触させ、この場合、必要に応じて、生物サンプル中に含まれるCamp ylobacter jejuniの核酸を予めハイブリダイゼーションし易い 状態としておき、b)Campylobacter jejuniに属する核酸 を増幅させ、c)プライマーを両側に有する断片に対応するCampyloba cterjejuniの核酸の断片が増幅されたことを検出し、d)特異的プロ ーブのハイブリダイゼーション、シーケンシングまたは制限部位分析等によって 増幅された断片を必要に応じて確認する。
- 12.下記段階を特徴とする食物試料中のCampylobacter jej uniの存在を検出する方法: a)低速遠心分離で粗食物組織を除去し、b)食物試料と請求項9または10に 記載のプライマーペアとをプライマーがCampylobacter jeju njの核酸にハイブリダイズするような条件下で接触させ、この場合、必要に応 じて、食物サンプル中に含まれるCampylobacter jejumiの 核酸を予めハイブリダイゼーションし易い状態としておき、c)Campylo bacter jejuniに属する核酸を増幅させ、d)プライマーを両側に 有する断片に対応するCampylobacterjejuniの核酸の断片が 増幅されたことを検出する。
- 13.下記要素を有することを特徴とする生物試料中または食物試料中のCam pylobacter jejuniの存在を検出するためのキット:1)請求 項9または10に記載のプライマーペア、2)Campylobacter j ejumiの核酸を増幅させる試薬、3)必要に応じて増幅した断片の配列を確 認するもの、特に、請求項6〜8に記載の核酸プローブ。
- 14.Campylobacter jejuni株を特定のCampylob acter jejuni株として特定・分類するための疫学的手段としての請 求項6〜8のいずれか一項に記載の核酸プローブの使用。
