【発明の詳細な説明】 治療用ペプチド含有のミクロエマルジョン 発明の分野 本発明は治療薬含有の医薬上許容される油中水(w/o)自己乳化性ミクロエ マルジョン、その調製法およびその使用に関する。 発明の背景 ミクロエマルジョンは、一般に、界面活性分子の界面膜により安定化された2 つの不混和性液体の熱力学的に安定した、等方性透明分散系と定義できる。ミク 良するための錠剤形成アジュバント、およびc)吸収エンハンサーとしての使用 を包含する。DSSはヘパリン(エンゲル・アール・エッチ(Engel,R.H.)ら、 ェイ・ファーム・サイ(J.Pharm.Sci)58,706−710、1969)、イ ンスリン(デュポン・エイ(Dupont,A)ら、ウゲルクリフト・ラゲル(Ugeskrif t Lager)119,1461−1463、1957)およびフェノー ルジョン組成物を記載する。適当な油は、ポリオキシエチレン脂肪酸エステルお よび/またはポリオキシエチレン脂肪性アルコールエーテルからなる界面活性剤 を含むミリスチン酸イソプロピル、2−オクチルドデカノールおよびパラフィン 油を包含する。用いる補助界面活性剤は水相であるポリオキシエチレングリセロ ール脂肪酸エステル/水である。塩基性薬剤の場合、イオン対形成化合物はサル ファートである。 クロエマルジョンに対する先駆体として新規かつ有用である。したがって、本発 明は、 (a)中鎖脂肪酸トリグリセリドと低HLB界面活性剤を有する親油性相であっ て、中鎖脂肪酸トリグリセリドの低HLB界面活性剤に対する割合が5:1〜1 .5:1である親油性相と; (b)ポリオールおよび i)サルファートまたはその医薬上許容される塩(脂肪性サルファート、ア リールサルファート、脂肪性−アリールサルファートまたはその混合物); ii)スルホナートまたはその医薬上許容される塩(脂肪性スルホナート、ア リールスルホナート、脂肪性−アリールスルホナートまたはその混合物); iii)スルホスクシナートまたはその医薬上許容される塩(脂肪性スルホス クシナート、アリールスルホスクシナート、脂肪性−アリールスルホスクシナー トまたはその混合物);または ルエーテルまたはポリエチレングリコール長鎖アルキルエステルである。 本発明にて用いる適当な中鎖脂肪酸トリグリセリドは、天然、半合成または合 成のものであり、種々の中鎖脂肪酸トリグリセリド、および/または中鎖および 長鎖脂肪酸トリグリセリドの混合物を包含する。かかる混合物は、中鎖および長 鎖脂肪性アシルトリグリセリドの物理的混合物だけでなく、例えば、エステル交 換により、中鎖および長鎖脂肪性アシル基を含むように化学的に修飾されたトリ グリセリドも包含する。 好ましい中鎖脂肪性アシルトリグリセリドの場合、脂肪酸組成物は、所望によ りカプリン(C10)酸と混合していてもよいカプリル(C8)酸(例、50〜1 00%(w/w)のカプリル酸および0〜50%(w/w)のカプリン酸)のト リグリセリドである。適当な例は、商品名 MYRITOL;CAPTEX(カ たは脂肪酸をいい、「中鎖」について前記したように所望により置換されていて もよい。 本発明のミクロエマルジョンにおいて、非イオン性高HLB界面活性剤は、通 常、多量の水性相を可溶化し得るミクロエマルジョンを生成するための補助高H LB界面活性剤として配合される。しかし、かかるミクロエマルジョンは、一般 に、非イオン性高HLB界面活性剤が不在のミクロエマルジョンよりも比較的粘 性である。 本発明にて用いられる適当な低HLB界面活性剤は、脂肪性アシルモノグリセ リド、脂肪性アシルジグリセリド、ソルビタン中鎖または長鎖脂肪性アシルエス テルおよび中鎖遊離脂肪酸、ならびにその混合物を包含する。適当なモノおよび ジグリセリドは、各々、異なる脂肪性モノおよびジグリセリドの混合物を包含し 、その脂肪酸基は中鎖または長鎖あるいはその混合物である。 適当な中鎖脂肪性アシルモノおよびジグリセリドはカプリル酸およびカプリン 酸から形成される。適当な混合物は、約50〜100%のカプリル酸および約0 〜50%のカプリン酸からなる。モノおよびジグリセリドの混合物は、好ましく は少なくとも50重量%、より好ましくは少なくとも70重量%のモノグリセリ ドからなる。適当なこれらの商業上の供給源は、CAPMUL(カールシャム・ リピッド・スペシャルティーズ(Karlsham Lipid Specialities))の商品名で 入手可能な製品、例えば、モノグリセリド(77%)、ジグリセリド(21%) および遊離グリセロール(1.6%)からなり、脂肪酸組成が、カプロン酸(3 %)、カプリル酸(67%)およびカプリン酸(30%)であるCAPMULM CMならびにモノグリセリド(70〜90%)、シグリセリド(10〜30%) および遊離グリセロール(2〜4%)を有し、脂肪酸組成が、少なくとも98% のカプリル酸からなるCAPMUL C8を包含する(オレアートとして表示さ れる製造業者のデータ;実際のC8/10モノおよびジグリセリド含量は、各々 、約45%)。 本発明の好ましい具体例において、低HLB界面活性剤は、少なくとも約80 重量%の、好ましくは少なくとも約90重量%の、さらに好ましくは少なくとも 約95重量%のカプロン酸、カプリル酸、カプリン酸モノグリセリドまたはその 混合物を含むモノおよびジグリセリドの混合物、好ましくはカプロン酸、カプリ ル酸、カプリン酸モノグリセリドまたはその混合物を含有する。これら界面活性 剤の製品は、例えば、Imwitor 308(Huls America,Inc.)(約80− 90重量%のカプリル酸モノグリセリド含有);およびグリセロールモノカプリ リン(シグマ・ケミカルズより1−モノオクタノイル−ラク−グリセロールとし て製造)(約99重量%のカプリル酸モノグリセリド含有);およびグリセロー ルモノカプラート(シグマ・ケミカルズより1−モノデカノイル−ラク−グリセ ロールとして製造)(約99重量%のカプリン酸モノグリセリド含有)を包含す る。 適当な長鎖脂肪酸モノグリセリドは、グリセロールモノオレアート、グリセロ ールモノバルミタートおよびグリセロールモノステアラートを包含する。適当な 市販品の例は、MYVEROLの商品名で入手可能な製品、例えば、MYVER OL 18−92(サンフラワー油モノグリセリド)および18−99(ナタネ 油モノグリセリド)、MYVATEXおよびMYVAPLEX(各々、イースト マン・コダック・ケミカルズ(Eastman Kodak Chemicals),ニューヨーク州、 ロチェスターから入手)を包含する。さらに有用な長鎖脂肪性アシルモノグリセ リド含有製品は、ARLACEL 186(ICI・アメリカズ・インコーポレ ーション(ICI Americas Inc.)より入手可能)であり、これはグリセロールモ ノオレアートに加えて、プロピレングリコール(10%)を含む。MYVERO L 18〜92の主な脂肪酸は、オレイン酸(19%)、リノール酸(68%) およびパルミチン酸(7%)であり、一方、MYVEROL18−99の主な脂 肪酸はオレイン酸(61%)、リノール酸(21%)、リノレン酸(9%)およ びパルミチン酸(4%)である。適当には、このような長鎖モノグリセリドにお いて、主な脂肪酸成分は、C18の飽和、モノ不飽和またはポリ不飽和脂肪酸、好 ましくはC18のモノ不飽和またはポリ不飽和脂肪酸である。加えて、Myvat ex SMGの商品名で入手可能な製品であるモノグリセリドのジアセチル化お よびジスクシニル化製品も有用である。 本発明において用いるのに適当なソルビタン長鎖エステルは、SPAN 80 およびARLACEL 80の商品名で市販されているソルビタンモノオレアー トならびにSPAN 83およびARLACEL 83の商品名で市販されている ソルビタンセスキオレアートを包含する。本発明において用いるのに適当なソル ビタン中鎖エステルは、ソルビタンカプリラート、ソルビタンカプラート、ソル ビタンラウラートを包含する。 本発明において用いられる適当な中鎖脂肪酸は、カプリル酸およびカプリン酸 およびその混合物である。 適当には、低HLB界面活性剤は、約2〜8のHLB平均値を有する。CAM PULMCM、MYVEROL 18〜99、ARLACEL 80、ARLAC EL 83およびARLACEL 186製品のHLB値は、各々、約5.5〜6 、3.7、4.3、3.7および2.8である。1−モノカプリリンの推定HLBは 約8.0である。 本発明の好ましい具体例において、ミクロエマルジョンは、中鎖脂肪性アシル 成分、例えば、カプリル酸およびカプリン酸から誘導されるもの、特に、カプリ ル酸からの誘導体からなる。従って、好ましいミクロエマルジョンは、CAPT EX355、810、CAPTEX8000、特にCAPTEX8000;CA PMUL MCMまたはCAPMVL C8、特にCAPMUL C8からなる。 本発明の自己乳化性(w/o)ミクロエマルジョンにおける低HLB界面活性 剤(中鎖脂肪酸モノグリセリドおよび/または中鎖脂肪酸ジグリセリドであり、 親油性相の成分である)の使用は粒径減少をもたらし、治療薬の吸収を促進する と考えられる。 したがって、好ましい具体例において、本発明は、中鎖脂肪酸トリグリセリド がカプリル酸またはカプリルおよびカプリン酸トリグリセリドの混合物であり、 低HLB界面活性剤が中鎖脂肪酸がカプリル酸またはカプリルおよびカプリン酸 の混合物であり、所望により少量の中鎖脂肪酸、特にカプリル酸トリグリセリド およびカプリル酸モノグリセリドまたはジグリセリドあるいはその混合物を含有 していてもよい中鎖脂肪酸モノグリセリドまたはジグリセリドあるいはその混合 物であるミクロエマルジョンを提供する。 本発明にて用いる高HLB界面活性剤は、脂肪性、アリールまたは脂肪性−ア リールサルファート、スルホナートまたはスルホスクシナートである。該脂肪性 基は中鎖または長鎖アルキルまたはジアルキル基である。中鎖部は、前記したよ うに、C6〜C12、好ましくはC8〜C10を含有し、長鎖部はC12〜C24を、好ま しくはC14〜C18を含有する。脂肪性鎖は分枝または分枝していなくてもよく、 所望により、置換されていてもよく、飽和、モノ不飽和またはポリ不飽和であっ てもよい。好ましくは、脂肪性基は中鎖長である。 サルファート、スルホナートおよびスルホスクシナート基と連結するのに2つ の可能な形態がある。1つは脂肪性アシルエステル結合であり、他方はエステル 結合である。脂肪性またはアリール基がサルファート、スルホナートまたはスル ホスクシナートと脂肪性アシルエステル結合を形成する場合、すなわち、付加的 なカルボニル[C(O)]基を含有する場合、脂肪性(またはアリール)基はエ ステル結合の脂肪酸部として区別されるようになる。脂肪性アシルエステル結合 は、例えば、C6-12−C(O)−OS(O)3Na+である。これは、本明細書に てエステル結合と称される、C6-12C−O−S(O)3Na+の一般に認識され ている結合と異なる。この用語は、C6-12OH基とスルホン酸のスルホンエステ ルを形成するO−S(O)2−Na+基と反応させることに由来する。好ましくは 、該結合はエステル結合であり、脂肪性アシルエステル結合ではない。より好ま しくは、結合基は中鎖アルキルまたはジアルキル基である。 本明細書にて用いる場合、「アリール」サルファート、スルホナートまたはス ルホスクシナートなる語は、フェニルまたはナフチル基を意味する。適当なアリ ールスルホナートは、例えば、ベンゼンスルホナートを包含する。 サルファート、スルホナートおよびスルホスクシナートと連結するについて1 以上の部位が可能である場合、該化合物は脂肪性およびアリール結合の両方の混 合物、例えばドデシルベンゼンサルファートであってもよい。本明細書にて用い るこの組み合わせを「脂肪性−アリール」と称する。好ましくは、この例におけ る該脂肪性基は中鎖アルキルまたはジアルキル基である。 適当な長鎖アルキルまたはジアルキルサルファート、スルホナートおよびスル ホスクシナートは、限定されるものではないが、ミリスチン(C14)、パルミ チン(C16)、パルミトレイン酸(C16:1)ステアリン(C18)、オレ イン酸(C18:1)、バセエン(vaceenic)酸(トランスオレイン酸)または リノール酸(C18:2)を包含する。適当には、アルキルおよびジアルキル部 は、異なる中鎖および長鎖部の混合物、例えば、C12とC16基を有するジア ルキルサルファートである。 好ましくは、サルファート、スルホナートおよびスルホスクシナートは中鎖ア ルキルまたはジアルキル誘導体である。適当な中鎖基は、限定されるものではな いが、オクチル(C8)、デシル(C10)、ドデシル(C12)、イソオクトアノ イル、またはジオクチル、ジデシルもしくはジドデシルを包含する。好ましくは 、中鎖基はオクチル、デシル、ドデシルまたはジオクチルである。さらに好まし 適当には、サルファート、スルホナートまたはスルホスクシナートは、医薬上 許容される水溶性の塩、例えば、ナトリウムおよびカリウム塩のようなアルカリ 金属塩、またはアンモニウムもしくはN(R)4(Rはアルキルである)誘導体 と称される第四級アンモニウム塩であるか、またはエタノールアミンまたはトリ エタノールアミンのような第一級および第二級(プロトン化)アミン塩である。 好ましくは、該塩はアルカリ金属塩である。適当には、該塩は中鎖アルキルまた はジアルキルサルファート、例えばオクチル、デシルまたはドデシルサルファー トであるか、またはジアルキルスルホスクシナートの塩であり、そのうちナトリ ウムジオクチルスルホスクシナートおよびナトリウムドデシルサルファートが好 ましい。ナトリウムジオクチルスルホスクシナート(DSS)およびナトリウム ド デシルサルファート(SDS)は、各々、41および40の推定HLB値を有す る。 本明細書にて用いる「ポリオール」なる語は、すなわち2またはそれ以上のヒ ドロキシル基を含有する多価アルコールである。そのような化合物は、限定され るものではなく、エチレングリコール、プロピレングリコールまたはポリエチレ ングリコール(PEG)を包含する。PEGはまた高分子単位としてエチレング リコールを有するポリグリコールをいう。好ましくは、該ポリオールは3または それ以上のアルコール単位を有し、PEGのグリセロールである。さらに好まし くは、ポリオールは、分子量が400であるPEG(PEG−400)である。 他の適当な多価アルコールは、限定されるものではないが、エチレングリコール (2−OH単位)、ソルビトール(6−OH)およびマンニトール(6−OH) を包含する。 サルファート、スルホナートまたはスルホスクシナートのポリオールに対する 割合は、約10:1ないし1:1であり、好ましくは5:1ないし1:1、さら に好ましくは5:1ないし2:1、最も好ましくは約1:1である。 商業上入手可能なDSS製品は、あらゆるUSP等級を包含し、DSS(10 0%)、DSS85%界面活性剤(15%安息香酸ナトリウム)またはPEG4 00中のDSS(50%)を包含する。 適当には、本発明にて用いる混合物は、ポリオールと、少なくとも1つの高H LB界面活性剤(サルファート、スルホナートまたはスルホスクシナートあるい はその医薬上許容される塩)とからなるものである。サルファート部またはその 医薬上許容される塩は、脂肪性サルファート、アリールサルファート、脂肪性− アリールサルファートまたはその混合物である。スルホナートまたはその医薬上 許容される塩は、脂肪性スルホナート、アリールスルホナート、脂肪性−アリー ルスルホナートまたはその混合物である。スルホスクシナートまたはその医薬上 許容される塩は、脂肪性スルホスクシナート、アリールスルホスクシナート、脂 肪性−アリールスルホスクシナートまたはその混合物である。高HLB界面活性 剤は前記サルファート、スルホナートまたはスルホスクシナートのいずれかとそ の医薬上許容される塩の混合物であってもよい。かかる混合物はサルファート基 、スルホナート基またはスルホスクシナート基の2成分または3成分系混合物と することができる。 好ましい高HLBは中鎖アルキルまたはジアルキルサルファート、スルホナー トまたはスルホスクシナートあるいはその医薬上許容される塩である。 サルファート、スルホナートおよびスルホスクシナートである高HLB界面活 性剤の総重量は、処方中に存在する高HLB界面活性剤の総重量の少なくとも5 0%、より好ましくは100%に近い値である。処方中に他の高HLB界面活性 剤を配合していもよいことがわかるであろう。適当には、高HLB界面活性剤の 総重量は約5〜約30%(w/w)および約70から100%(w/w)未満の 範囲である。さらに好ましくは、高HLB界面活性剤の総重量は5〜約25%( (c)ポリエチレングリコール長鎖アルキルエーテルのようなPEGグリセロ ールエーテル、例えば、ポリエチル化グリコールラウリルエーテルおよびPEG 脂肪性アルコールエーテル; (d)長鎖アルキルエステルのようなPEGグリセロールエステルおよびPE G脂肪酸エステル、例えば、PEG−モノステアラート。 界面活性剤系は、加えて、限定されるものではないが、以下に示すような他の 界面活性剤を含有してもよい: とも処方中に用いた高HLB界面活性剤のHLB値を有し、薬剤が親油性相より も親水性相中に優先的に溶解する化合物をいうものである。かかる物質はペプチ ドおよび非ペプチドの両方を含む。適当なペプチドは、小ペプチドだけでなく大 ペプチド/ポリペプチドおよび蛋白質も包含する。適当なこのようなペプチドは 、好ましくは約100〜10000、より好ましくは約100〜約6000の分 子量を有する。特に好ましいのは、2〜35アミノ酸基を有するペプチドである 。高分子量のペプチド、分子量が10000より大きく、約50000までのペ プチドも本発明のミクロエマルジョンに応用できる。 適当な小ペプチドは、約2〜約10、より好ましくは約2〜約6個のアミノ酸 基を有する。好ましい小ペプチドは、RGD含有ペプチド、例えば平均分子量が 用いて、活性成分の一体性および組成物の安定性を失うことなく、PEG(40 0)と高HLB界面活性剤、特に脂肪族サルファート、スルホナートまたはスル ホスクシナートの脂肪性アシルまたはジアシル誘導体の比率を適当に修飾するこ とにより薬剤の溶解を促進してもよい。 水性親水性相は、水または等張セイライン溶液を含むのが適当であり、さらに 選択された親油性相と不混和性の医薬上許容される溶媒、例えば、ポリエチレン グリコール、プロピレングリコール、ソルビトール、マンニトールおよび他のモ ノまたはジサッカリドを含んでもよい。 中鎖脂肪酸トリグリセリド、低および高HLB界面活性剤および親水性相のす べての混合物から本発明の範囲に含まれる安定な自己乳化性ミクロエマルジョン が得られるわけではないことは当業者には容易に理解できる。しかし、適当な割 合は、第1図に示す相図により当業者には容易に決定できる。この系は、中鎖脂 肪酸トリグリセリド(油)、低HLB界面活性剤、高HLB界面活性剤および水 性/親水性相の4成分からなるため、疑似三相図を用いる。この場合、3つの変 数だけがあり、その各々を三角形の1つの辺で表されるように、2成分、例えば 油と低HLB界面活性剤の割合を一定に保つ。すなわち、第1図において、(1 )は一定割合Xの油と低HLB界面活性剤の混合物を表し、(2)は親水性(水 性)相を表し、(3)は高HLB界面活性剤を表す。例えば、点「A」は50% 油+低HLB界面活性剤、20%水性相および30%高HLB界面活性剤の混合 物を表す。 本発明のミクロエマルジョンが存在する相図の領域は、油および低HLB界面 活性剤の混合物(一定割合)を高HLB界面活性剤および親水性相に対して滴定 することにより決定され、相分離点、曇点および透明点を明記する。清澄、透明 な処方は、安定したミクロエマルジョンの形成を示すものである。用いる成分の 個々の特性に従って、室温で液体およびゲル状の処方が得られる。 安定で透明な系が得られたら、色素可溶化、水中の分散性および導電率の測定 などの簡単な試験を行って、ミクロエマルジョンが(o/w)または(w/o) 型のいずれであるかを決定する。水溶性色素は(o/w)ミクロエマルジョン中 に分散し、一方(w/o)ミクロエマルジョン中では元の形態のままである。同 様に、(o/w)ミクロエマルジョンは一般に水中に分散するのに対して、(w /o)ミクロエマルジョンは、一般に、分散しない。加えて、(o/w)ミクロ エマルジョンは電気を通すのに対して、(w/o)は通さない。系の等方性は偏 光下での試験により確認できる。ミセル特性であるミクロエマルジョンは等方性 であり、したがって、偏光下で試験した場合、非複屈折である。 中鎖油(CAMTEX8000)および低HLB界面活性剤(CAMTEXC 8)を2:1の割合で、高HLB界面活性剤(DSS/PEG400)を1:1 の割合で、かつ水を含有する系の代表的疑似三相図を第2図に示す。油+低HL B界面活性剤の混合物は成分(1)で示されており、水は成分(2)で、高HL B界面活性剤は成分(3)で示されている。この系は、ミクロエマルジョン領域 として相図に示される、清澄、透明なミクロエマルジョンの2つの範囲を付与し 、その範囲は、通常、領域(A)と(C)に細分割される。 不安定な処方は領域(A)と(C)の間の範囲にて生成されるが、領域(A) から領域(C)の連続した範囲にて、PEG400のDSSに対して別の割合お よび/または油/低HLB界面活性剤の別の割合を用いることにより、安定した ミクロエマルジョンが得られる。 この細分割は、主に、過剰量の水の存在下、伝導度、粘度および希釈度の違い によるものである。 本発明の範囲内にあるミクロエマルジョンは疑似三相図の領域(A)および( C)にあるものである。粘度および伝導度は共に領域(C)にて増加する。過剰 の分散相(セイラインまたは水)の存在下で、領域(A)のミクロエマルジョン は元の(w/o)を示す、混濁したエマルジョン(o/w)になる。対照的に、 領域(C)からのミクロエマルジョンは水中油の等方性系(ミクロエマルジョン )の清澄な特性を保持する。 したがって、さらなる態様において、本発明は、種々の成分の相対的割合が第 2図のような疑似三相図の領域(A)および(C)の範囲内にある、前記の安定 な自己乳化性ミクロエマルジョンを提供する。 一般に、代表的系において、油+低HLBが、ミクロエマルジョンの約5%〜 約25%の範囲にて、および約70%から100%未満の範囲にて存在し、高H LB界面活性剤が約5%から約30%および約70%から100%未満の範囲に て、および水性親水性相、すなわち水またはセイラインが20%(w/w)未満 で存在する場合、安定した清澄、透明な液体ミクロエマルジョンが得られる。 相図の代表的な範囲を作成するこのプロセスにより、本発明の範囲内にある安 定した自己乳化性ミクロエマルジョンを得る種々の成分の適当な量を決定できる 。 適当には、一の場合において、中鎖脂肪酸トリグリセリドおよび低HLB界面 活性剤の合計がミクロエマルジョンの約70ないし100%(w/w)からなる 。もう一つ別の場合において、中鎖脂肪酸トリグリセリドと低HLB界面活性剤 の合計がミクロエマルジョンの約5ないし約25%(w/w)からなる。中鎖脂 肪酸トリグリセリドと低HLB界面活性剤を種々の割合で合し、混合してもよい 。中鎖脂肪酸トリグリセリドの低HLB界面活性剤に対する割合が約5:1ない し約1.5:1、好ましくは約4:1ないし約2:1の範囲にある場合、有用な (w/o)ミクロエマルジョンが得られる。中鎖脂肪酸トリグリセリドの低HL B界面活性剤に対する割合が5:1に向かって増加するにしたがって、ミクロエ マルジョン存在範囲の領域(C)がますます支配的となることが判明した。 適当には、一の場合において、高HLB界面活性剤は、約5ないし約30%の 範囲にて存在してもよく、もう一つ別の場合において、70%ないし100%未 満にて存在する。加えて、高HLB界面活性剤をポリオールと10:1ないし1 :1にて、好ましくは1:1にて混合してもよい。 適当には、親水性相は、ミクロエマルジョンの0よりもほんの少し大きな値か ら約20%(w/w)、好ましくは約0.1〜約15%(w/w)からなる。 一般に、高HLB界面活性剤の相対量の増加が、親水性相の相対量の増加に匹 敵することは当業者に容易に理解される。 本発明の好ましいミクロエマルジョンにおいて、中鎖脂肪酸トリグリセリドの 低HLB界面活性剤に対する割合は、好ましくは、4:1および2:1である。 本発明のミクロエマルジョンは実質的に不透明でない、即ち光学的顕微鏡手段 で見た場合、透明または乳白色である。均質状態において、これらは偏光下で試 験した場合、光学的に等方性(非複屈折)である。低温および外界温度で、長時 間にわたって、相分離、白濁または沈殿を生じることなく、優れた安定性を示す 。処方は、種々の温度、例えば4℃、外界温度、37℃および50℃、好ましく は4℃または外界温度で安定な形態で貯蔵できる。 本発明のペプチド含有ミクロエマルジョンは、対応するペプチド不含ミクロエ マルジョンの安定性(貯蔵寿命)と類似した安定な特性を示す。安定な(w/o )ミクロエマルジョンは水性相のpHが約3から約8まで変化する場合に形成さ れ、低または高pHで高い溶解度を示す薬剤に関して有利である。該ミクロエマ ルジョンは種々の粘度を示す。相対的に多量の高HLB界面活性剤を含むミクロ エマルジョン、領域(C)DSS/PEG400(1:1)は、この物質の高い 粘度のためにより粘度が高くなる傾向がある。 好ましくは、本発明のミクロエマルジョンの小滴または粒子の直径は、例えば レーザー光散乱技法による数平均直径として測定した場合、150nm未満、よ り好ましくは100nm未満、さらに好ましくは50nm未満、最も好ましくは 5〜35nmの範囲である。 種々の相は、所望により、さらに以下の成分をある少量にて含んでもよいが、 これに限定されない: i)酸化防止剤、例えば没食子酸n−プロピル、ブチル化ヒドロキシアニソ ール(BHA)およびその混合異性体、d−α−トコフェロールおよびその混合 異性体、アスコルビン酸、プロピルパラベン、メチルパラベンおよびクエン酸( モノ水和物); ii)安定化剤、例えばヒドロキシプロピルセルロース; iii)抗菌薬、例えば安息香酸(ナトリウム塩); iv)プロテアーゼ阻害剤、例えばアプロチニン。 本発明のミクロエマルジョンは、その成分を接触させた場合、自発的または実 質的に自発的に、即ち実質的にエネルギーを供給することなく、例えば均質化お よび/またはミクロ流動化または他の機械的攪拌により与えられるような高い剪 断エネルギーの非存在下で形成される。したがって、該ミクロエマルジョンは適 当な量を穏やかに手動で混合するかまたは必要ならば確実に混合するように攪拌 する簡単なプロセスにより容易に調製できる。好ましくは、薬剤を親水性相中に 、直接またはそのストック溶液の希釈化により溶解させ、これを次にあらかじめ 混合した油および低HLB界面活性剤の組合せに、続いて高HLB界面活性剤( またはその逆)に混合しながら添加する。別法として、油、低HLB界面活性剤 、高HLB界面活性剤および薬剤不含親水性相を混合することにより、まず薬剤 不含ミクロエマルジョンを調製し、ついでこれにさらに薬剤を溶解した親水性相 を添加する。ミクロエマルジョンを調製する間、すべての成分を溶解させるのに より高い温度(40−60℃)を必要するかもしれないが、好ましい系は室温で 処方できる。ペプチドのような熱不安定性活性成分の場合、外界温度での処方が 特に有利である。 本発明のミクロエマルジョンは治療剤からなる医薬組成物であり、したがって 、ヒトを含む動物に投与する治療法を包含するものである。 従って、別の態様において、本発明は有効量の治療薬を配合した前記ミクロエ マルジョンを、これを必要とする患者に投与することからなる治療法を提供する ものである。 投与に対する治療効果を得るのに必要な薬剤の量は、もちろん、選択した薬剤 、症状の性質および程度、および治療を受ける動物によって変わることは当業者 には理解でき、最終的には医師の裁量による。さらには、薬剤の最適量および各 投与の間隔は治療する症状の性質および程度、投与の形態、経路および部位、治 療を受ける患者によって決定され、このような最適値は常套手段により決定でき る。また、治療の最適経路、即ち投与する回数も通常の治療法決定試験を用いて 容易に確認できることは明らかである。 本発明はまた、医薬の製造における、前記した中鎖脂肪酸トリグリセリド、低 HLB界面活性剤、高HLB界面活性剤、治療薬および親水性相の使用を提供す る。 本発明のミクロエマルジョンは、経口、局所、直腸、膣内または他の全身投与 に用いることができる。したがって、該ミクロエマルジョンはそれに適した形態 にされる。例えば、経口投与用の医薬組成物はソフトゼラチンカプセルにされ、 ある種の医薬組成物の粘性は直接局所投与するのに適当である。経口および局所 投与用の組成物が特に好ましい。 本発明を以下の記載例(薬剤不含組成物)および実施例(薬剤含有組成物)お よび生物学的例により説明するが、これに限定されない。 代表的組成物の相図 疑似三相図を以下の代表的系について作成した: 中鎖脂肪酸トリグリセリド(油) CAPTEX8000 低HLB界面活性剤 CAPMUL C8 高HLB界面活性剤 DSS/PEG400(1:1) 水性相 水 油の低HLB界面活性剤に対する割合は2:1であった。 また、油の低HLB界面活性剤に対する割合が5:1、4:1または3:1で ある前記系について疑似三相図を作成できる。 他の別の疑似三相図は、高HLB界面活性剤がDSS(純粋)またはDSS+ 15%安息香酸ナトリウムをPEG400または別のポリオールに10:1ない し1:1の範囲にて溶かし、2:1ないし5:1の割合で前記の代表的系につい て作成できる。 本発明のミクロエマルジョンが存在する相図の領域は、油および低HLB界面 活性剤(一定割合)の混合物を高HLB界面活性剤および水性相に対して滴定す ることにより決定し、相分離点、曇点および透明点を明記する。得られた相図を 第2図に示す。偏光下で試験した場合、非複屈折が観察された。 前記した例を用いて、PEG400の代わりにプロピレングリコールまたはグ リセロールのような他のポリオールを用いて別の処方を調製してもよい。また、 DSS高HLB界面活性剤をナトリウムドデシルサルフファート(SDS)に変 えてもよい。 実施例 本明細書の記載に従って、以下の組成を有する処方を調製できる。 これらのミクロエマルジョンは、親水性相をジオクチルナトリウムスルホスク シナートのPEG中溶液に加えることにより処方した。その後、この混合物に油 +低HLB界面活性剤の混合物を加え、手動により十分に混合し、清澄な透明ミ クロエマルジョンを得た。 別法として、親水性相を、油および低HLB界面活性剤の適当な重量の混合物 に加え、ついでそれに高HLB界面活性剤を穏やかに攪拌しながら(マグネティ ックホットプレートスターラー)加えることができる。 活性成分配合のミクロエマルジョンは、適量の薬剤を適量の水性相に溶かすか 、または好ましくは、ストック溶液を用いることにより、相前記したと同様の方 法にて調製でき、ついで必要ならば、渦巻き状に攪拌しながら希釈して、完全に 溶解させる。 生物学的例カルセインの生物学的利用能 標準的無意識ラット実験(ウォーカー(Walker)ら、ライフ・サイエンス(Li fe Science)47,29−36,1990)を用いて、試験化合物であるカルセ イン[5(6)−カルボキシ−フルオロセイン、分子量=623]を前記のいず れかの実施例におけるミクロエマルジョンとして投与した場合の十二指腸内生物 学的利用能を評価し、同一化合物を同一経路であるが、等張トリス緩衝液として 投与した場合に得られる評価と比較した。血漿試料中の化合物の濃度を蛍光分光 学を用いて測定する。ミクロエマルジョン1kg当たり約1.25マイクロモル または1.0mlを十二指腸内(i.d.)投与した後、生物学的利用能を測定し 、等張トリス緩衝液として投与した場合の生物学的利用能と比較する。 前記した生物学的利用能測定は、当業者にとって十分に理解できるものである が、便宜上、以下に繰り返し記載する: 吸収実験について用いる場合、雄ラットを一夜絶食させる。溶液またはミクロ エマルジョンからの化合物(この場合、カルセイン)の静脈内(i.v.)または 十二指腸内(i.d.)投与を常法に従って行う。 i.v.投与の場合、絶食させたラットをロムパン(Rompun)(5mg/kg) およびケトセット(ketset)(35mg/kg)の混合物を腹腔内注射して麻酔 し、静脈用カテーテルを装着する。一日、ラットを手術から回復させる。カテー テルを装着したラットを化合物の投与前18時間絶食させた。各化合物を側尾− 静脈投与により投与する。0.5mlのアリコートの血液試料を0、1、3、5 、15、30、45、60、90、120、150および180分で採集した。 0分の試料は投与の15分前に採集する。1600xgで5分間遠心分離に付す ことにより、血漿を全血から取り出し、ついで試料当たり250μlのアリコー トを−20℃で貯蔵する。血液ペレットを12.5単位のヘパリン処理したセイ ラインで復元し、静脈用カテーテルを介してラットに戻す。実験後、ラットをペ ントハルビタールを静脈内投与して殺す。 i.d.投与の場合、静脈用カテーテルに加えて、十二指腸用カテーテルを外科 的に麻酔ラットに装着し、動物を4ないし5日間手術より回復させる。化合物を 溶液またはミクロエマルジョンより十二指腸用カテーテルを介して投与する。血 液試料(0.5mlアリコート)を静脈用カテーテルを介してヘパリン処理した エッペンドルフ(eppendorf)管にて0、10、30、60、120、180、 240および1440分で採集する。0分試料は投与の15分前に採集する。i .v.投与プロトコルにて前記したように、血漿を分析用に採集し、血液をラット に戻す。各ラットの時間経後の大便を、軟便、軟便/水便または粘性のランクで 評価する。 吸収実験の終わりに(投与後、4〜6時間または24時間)、二酸化炭素を用 いて窒息させて殺し、採血する。ついで、腹部切開を行い、完全なGI管を摘出 し、50x倍率の顕微鏡で観察する。 カルセインの血漿中濃度を、パーキン・エルマー・LS・50発光分光計を用 い、490および515nmの励起および放射波長で蛍光により測定する。生物 学的利用能(%F)を、以下の式を用いて、i.d.またはi.v投与後のAUC (血漿中濃度−時間曲線下の面積)より算定する: %F=(AUCid/AUCiv)x(用量iv/用量id)x100 本発明の処方を以下の方法により活性成分を有するまたは有しないGI刺激評 価について試験する。ラットにおける経口投与/GI刺激評価 この試験において使用するのに適当なラットは、雄スプレーグ−ドーリ((Sp rague-Dawley)、帝王切開出産、無ウイルス抗体;チャールス・リバー・ラボラ トリース(Charles River Laboratories))である。ラットを実験の前日一夜絶 食させる。所望の投与量のミクロエマルジョンを10mg/kgを越えない量で 強制飼養により投与する。実験の終了時に動物を二酸化炭素を用いて窒息させる ことにより殺し、採血する。腹部切開を行い、胃および十二指腸粘膜の全体的観 察を裸眼および顕微鏡(ニコンSMZ−10型双眼顕微鏡)を用いて行う。 本発明の一態様は、経口投与によりGI管に沿って障害があったとしてもほと んどないペプチドを含むかまたは含まないw/o自己乳化性ミクロエマルジョン の処方である。例えば本発明の処方を、強制飼養により経口投与する(好ましく は1処方につきラット3匹)。24時間後、動物を採血して、腹部切開により、 裸眼および顕微鏡の両方で観察する。動物の胃および十二指腸の両方の粘膜表面 を調べて、裸眼で病変がないかどうかを見る。ラットにおけるRGDペプチドの経口生物学的利用能 以下に記載する操作において、前記したようにして処方した、例えばミクロエ マルジョン1g当たり3mgのペプチド(例えば、RGDフィブリノゲン受容体 拮抗物質含有ペプチド)を含有するミクロエマルジョンを以下の方法にて経口生 物学的利用能に関して試験する。 a)セイライン中ペプチドの静脈内(iv)投与 絶食したラットに腹腔内(ip)注射を行い、外科手術により大腿動脈カテー テルを設置する。ラットを1日の間、手術から回復させた。カテーテルを挿入し たラットを実験前18時間絶食させる。各ラットに側尾静脈投与により以下のよ うにして調製した溶液から3mgのペプチドを投与する: 10.84mgのペプチドを0.9%セイライン溶液で8mlにする。0.5m lアリコートの血液試料を0、1、3、5、10、15、30、45、60、9 0、120、150および180分に採集する。0分の試料を投与する15分前 に採取する。16000Xgで5分間遠心分離することにより血漿を全血から取 り出し、次に該血漿を−20℃で1試料当たり250μlのアリコートにて貯蔵 する。血液ペレットをヘパリン処理したセイライン溶液で復元し、カテーテルを 介して適切なラットに戻す。実験後、ラットにペントバルビタールを静脈内投与 して殺す。 b)ミクロエマルジョン中ペプチドの十二指腸内(id)投与 絶食したラットに麻酔カクテルを腹腔内注射し、外科手術により頸静脈および 十二指腸用カテーテルを挿入する。ラットを4〜5日間、手術から回復させる。 カテーテルを挿入したラットを実験前18〜20時間絶食させる。各ラットにミ クロエマルジョン中またはセイライン溶液中のいずれかのペプチド10mgを投 与する。0、10、30、60、120、180、240および1440分にヘ バリン処理したエッペンドルフ管中に頚静脈カテーテルを介して0.5mlアリ コートの血液試料を集める。0分の試料を十二指腸用カテーテルにより投与の1 5分前に採取する。血漿を分析のために採集し、血液を前記静脈内投与(項a) において記載したようにラットに戻す。1440分後、ラットをペントバルビタ ールの静脈内投与により殺し、採血し、全体的観察のためにGI管を摘出する。 c)ペプチド血漿濃度の分析 HPLC分析用の試料の前後に標準体を置く。0〜200ngのペプチドにつ いて50μlアリコート、1000〜2000ngのペプチドについて25μl アリコート、10000ngのペプチドについて15μlアリコート、各サンプ ルの50μlアリコートをポストカラム蛍光検出により分析する。蛍光クロマト グラフィーデータを集め、ネルソン・クロマトグラフィー・データ・システム (Nelson Chromatography Data System)を用いて積分する。ピーク面積(Y) およびペプチド標準濃度(X)を用いて、式:勾配=(X*Yの合計)/(X2 の合計)から原点を通した直線の勾配を決定する。勾配は、試料に関するピーク 面積比およびペプチド血漿濃度間の関係を表わす。 d)生物学的利用能の計算 まず、0〜240分での血漿濃度曲線下の面積(AUC)を各ラットについて 測定する。十二指腸内投与の場合、以下の式により、静脈内投与の平均AUCを 用いて各動物に関して生物学的利用能(%)を決定する [(AUCid/AUCiv)*(用量iv/用量id)]*[100]。 ついで、ペプチド用量のフィブリノゲン受容体拮抗物質を配合した前記処方を 含有するミクロエマルジョンを十二指腸内投与した後のラットにおけるRGDペ プチドに関する経口生物学的利用能のデータを、前記方法により得ることができ る。 適用可能ならば、本発明の処方をin vivo活性について試験する。活性成分の 1つとして、フィブリノゲン受容体拮抗物質を本明細書にて用い、血小板凝集試 験を利用し、ミクロエマルジョン由来のペプチドの薬理活性を測定する。これら 研究を以下に示すように行う。イヌにおける経口投与/血小板凝集試験 この試験において用いたイヌは、オス雑種(Mongrel)(即ち混合種)である 。イヌを実験の前日一夜絶食させる。留置カテーテル用に選択した頭部静脈を以 下のようにして準備する:該当部分の毛をまず剃り、70%アルコール中に浸し たガーゼで清浄にする。留置カテーテルを頭部静脈中に置き、3.8%クエン酸 ナトリウムを満たしたルエル(luer)ロックアダプターにつなぐ。カテーテルを しっかりテープで固定する。血液サンプルを採取する場合、0.3mlの血液を 実際の試料採取の前に個別の1ccシリンジ中に採取し、ルエルロックアダプター 中に含まれるクエン酸ナトリウムにより血液サンプルが希釈されるのを防ぐ。次 に2.7mlの血液を3ccシリンジ中に抜き取り、0.3mlの3.8%クエン酸 ナトリウムを含み、適当な時点で標識したベノジェクト(Venoject)真空管中に いれる。3.8%クエン酸ナトリウム中血液試料を含む管を静かに2、3回倒立 させて、成分を混合し、次に全血凝集試験のために1mlを採取する。残りの血 液試料をエッペンドルフ管に移し、遠心分離により上清血漿を取り出して新しい 管に移し、ついでその後のHPLC分析のために凍結させ、ペプチド含量を測定 する。 0時点の直後に血液試料を採取し、ペプチドを含むかまたは含まない適量のミ クロエマルションを、サイズ12のゼラチンカプセルを用いてイヌに経口投与す る。 血液試料を次にクロモ−ログ(Chromo-Log)全血凝集検出計を用いて血小板凝 集阻害に関して試験する。試料を試験する前に装置を37℃に加温し、プローブ を蒸留水および柔らかいブラシで清浄にする。プローブを凝集検出計に取付け、 セイライン溶液のキュベット中にいれ、凝集検出計中のサイドキュベットウェル 中にて加温する。実際の検定に関しては、ベノジェクト真空管中の0.3mlの 3.8%クエン酸ナトリウムと混合した2.7mlの血液試料の1mlをキュベッ トに添加し、凝集検出計のウェル中に入れる。攪拌棒をキュベット中にいれ、9 00rpmにセットする。プローブを試験キュベット中にしっかりといれ、蓋を 閉める。ベースライン、ゼロおよび目盛りをセットする。目盛りは20を5オー ムに等しく設定する。攪拌キュベットを5分間静置し、この時点で、5μlのコ ラーゲンを攪拌した全血に添加し、キュベット中5μl/mlの最終溶液を得る 。 一旦、勾配の変化がコラーゲン添加のベースラインに達したら反応を2分間モ ニターし、2分間の勾配を用いて変化をオーム/分で計算する。変化(オーム/
