- 【特許請求の範囲】 1.第一および第二の平行で平らな表面、および該第一および第二表面の間を伸 びる縁部を有する平らな導波管をであって、該縁部が光を受けるための受光区域 を有し、該表面の少なくとも一方がその上に固定された複数の捕獲分子を有し、 該捕獲分子がそれぞれ分析物を選択的に結合する結合部位を有する導波管と、該 受光縁部に隣接して該導波管に一体化されている半円筒形のレンズとからなるこ とを特徴とする均質固相蛍光免疫検定用装置。 2.所望の波長領域の光線を与え、かつ該光線を前記レンズ中に向ける様に配置 された光源、 前記導波管表面の少なくとも一方の上に形成された1個以上の貯蔵部であっ て、該貯蔵部のそれぞれが、緩衝液、少なくとも1種の前記分析物の複数の分子 、および前記光線から生じた消失性光で刺激したときに蛍光を放射し得る複数の トレーサー分子を含む試料溶液を含む様に形成された貯蔵部、および 本質的に前記蛍光からなる蛍光信号を直接検出する様に配置された検出手段 であって、前記表面に対して平行で前記表面から離れた平面上に前記蛍光が衝突 するように前記蛍光を収集する配置されたレンズ手段を含む検出手段 をさらに含む請求項1の装置。 3.間隔を置いて配置された前記固定捕獲分子の複数のパッチを有し、かつ前記 検出手段が、該パッチの個々の一つにそれぞれ対応する複数の蛍光信号を同時に 個別に収集する様に形成されている請求項2の装置。 4.前記検出手段が、前記離れた平行な平面内で互いに間隔を置いて配置された 複数の光検出素子、および前記蛍光信号のそれぞれを該光検出素子のそれぞれの 一つに集める様に配置されたレンズ手段を有する画像形成検出器を含む請求項3 の装置。 5.前記導波管および前記レンズが、光学プラスチックで形成されかつ一体成形 されている請求項1または2の装置。 6.前記半円筒形レンズが、シート状の光線を非ゼロ角度で前記受光縁部の中に 向ける方向で配置された焦点区域を有し、該角度が前記導波管および水性媒体の 間の界面で光の全内部反射をする臨界角度未満である請求項1、2または5の装 置。 7.前記導波管が、末端縁部を形成する鋸歯状部分を有し、該末端縁部が前記受 光縁部および前記半円筒形レンズと反対側の末端に配置されている請求項1また は6の装置。 8.前記導波管が請求項9の光学基材である請求項1、2、3又は4の装置。 9.前記導波管が、請求項10による光学基材であり、かつ導波管厚さを有し、 さらに、前記光源と前記半円筒形レンズの間に位置して、光線を一般的に前記導 波管の厚さの約2倍未満の厚さを有するシート状光線に形状変換するための光線 形状変換手段を含む請求項2、3又は4の装置。 10.光信号により対応する分析物を検出するために、該分析物を特異的に捕獲 するのに有用な捕獲分子が結合されている、固相検定に有用な光学基材であって 、該基材に対する非特異的な結合の量を適度に低くする被覆を施した区域を有す る光学表面を含み、該被覆が、ポリメタクリロイル重合体からなるヒドロゲル、 シリ ル−誘導体化されたポリエチレングリコール、アビジン、および本質的に親水性 残基からなる少なくとも1種の親水性重合体ブロックに隣接する、本質的に疎水 性残基からなる少なくとも1種の疎水性重合体ブロックを含むブロック共重合体 からなる群から選択されることを特徴とする光学基材。 11.前記被覆区域に共有結合された1種類以上の捕獲分子種をさらに含み、該 1種類以上の種のそれぞれが、対応する分析物に特異的に結合する様に構築され ている請求項10の光学基材。 12.光応答性結合部分および捕獲分子を共有結合できるタンパク質結合部分を 有する架橋剤をさらに含み、該架橋剤が、アリールアジド、フッ素化アリールア ジド、およびベンゾフェノンからなる群から選択され、該架橋剤が、前記光応答 性結合部分を介して前記被覆に共有結合する請求項10の光学基材。 13.前記架橋剤の前記タンパク質結合部分を介して前記被覆区域に共有結合さ れた1種類以上の捕獲分子種をさらに含み、該1種類以上の種のそれぞれが、対 応する分析物に特異的に結合する様に構築されている請求項12の光学基材。 14.前記被覆が、一般式 (PEO)x(PPO)y(PEO)z (式中、PEO=ポリエチレンオキシドおよびPPO=ポリプロピレンオキシ ドであり、x、yおよびzのそれぞれが約30〜約150である) を有するブロック共重合体である請求項10、12又は13の光学基材。 15.前記捕獲分子のそれぞれが、それぞれの中に反応により配置 されたチオール基を有し、および前記捕獲分子が該チオール基を介して前記被覆 に結合される、請求項12又は14の光学基材。 16.前記架橋剤がベンゾフェノンマレイミドである請求項12又は14の光学 基材。 17.前記光学基材が、シリカ系材料であり、および前記光学表面と前記被覆の 間で前記区域に塗布された下塗りをさらに含み、該下塗りが、少なくとも1個の 反応性シリル基を有するシリカ親和性剤を含む請求項10、12又は16の光学 基材。 18.請求項10の光学基材の製造方法であって、光学基材の区域を、ポリメタ クリロイル重合体からなるヒドロゲル、アビジン、メトキシポリ(エチレングリ コール)トリメトキシシラン、および本質的に親水性残基からなる少なくとも1 種の親水性重合体ブロックに隣接する、本質的に疎水性残基からなる少なくとも 1種の疎水性重合体ブロックを含むブロック共重合体、からなる群から選択され た被覆で被覆する工程を含む方法。 19.光応答性結合部分および捕獲分子を共有結合できるタンパク質結合部分を 有する、アリールアジド、フッ素化アリールアジド、およびベンゾフェノンから なる群から選択された光親和力架橋剤を用意する工程、 該被覆した光学表面を、該光応答性結合部分を活性化するのに適した光で照 射する手段を用意する工程、および 該前記照射手段を操作し、該被覆した光学表面を該架橋剤の存在下で照射し 、該光応答性結合部分を活性化し、それによって該架橋剤を該被覆に共有結合さ せる工程 をさらに含む請求項18の方法。 20.対応する分析物を特異的に結合する様にそれぞれ構築され、分子内にそれ ぞれ反応により配置されたチオール基を含む1種類以上の捕獲分子種を用意する 工程、および 該捕獲分子を、該チオール基を介して前記被覆に結合させる工程 をさらに含む請求項18又は19の方法。 21.対応する分析物を特異的に結合する様にそれぞれ構築された1種類以上の 捕獲分子種を用意する工程、および 該捕獲分子を、前記架橋剤の前記タンパク質結合部分に結合させる工程 をさらに含む請求項19の方法。 22.複数の捕獲分子種を用意し、前記照射手段を用意する工程が、前記導波管 の前記被覆区域の複数の部分を個別に順次照射する手段を用意する工程を含み、 および前記照射工程が、前記被覆区域の前記部分を前記架橋剤の存在下で選択的 に順次照射する工程である請求項21の方法。 23.前記捕獲分子を前記架橋剤に結合させる前記工程が前記照射工程の前に行 なわれる請求項21または22の方法。 24.前記の個別に順次照射する工程が、 遊離架橋剤の存在下で前記被覆区域の前記部分の一つを照射する工程、 前記捕獲分子種の1種類を前記架橋剤に結合させる工程、および 部分を前記照射する工程および捕獲分子の1種を結合する工程を交互に繰り 返す工程 を含み、該交互に繰り返す工程が、前記部分のそれぞれに対して前記複数の種 と異なった種で行なわれる請求項22又は23の方法。 25.分析物に特異的な捕獲分子を光学基材に結合させ、該分析物が該捕獲分子 に結合することにより、試料中の該分析物の存在に対応する蛍光信号を発生させ る固相蛍光免疫検定を行なう方法であって、 請求項1のバイオセンサーを用意する工程、 半円筒形レンズを通して導波管中に光線を送る様に配置された光源を用意す る工程、 導波管表面と平行なかつ導波管表面から離れた面上に当たる蛍光を検出する ために配置された検出手段を用意する工程、 表面上に固定した捕獲分子を有する該導波管表面を、緩衝液、分析物の存在 下で該捕獲分子に結合することができかつ光線で刺激したときに蛍光信号を放射 できる複数のトレーサー分子、および未知量の分析物分子を含む溶液と接触させ る工程、 該導波管表面と該接触溶液とを維持し、該トレーサー分子を該分析物分子お よび該捕獲分子と結合させる工程、 光源を作動させ、該導波管から消失性光を発生させ、該トレーサー分子を刺 激する工程、 検出手段を作動させ、該トレーサー分子から放射された該蛍光信号を検出す る工程、および 該放射された蛍光信号を解析し、試験試料中の分析物の量を決定する工程 を含むことを特徴とする方法。 26.前記導波管バイオセンサーを用意する工程において、前記導波管が請求項 10、11または12の光学表面を有し、前記用意された検出手段が、複数の光 検出器素子および被覆区域の複数の部分からの光を該光検出器素子のそれぞれ対 応する部分に向ける方向に配置されたレンズ手段を含む請求項25の免疫検定方 法。 27.結合していないトレーサーを前記表面から洗い流すための洗浄工程なしに 行なわれる請求項25の免疫検定方法。
