Claims (1)
- 【特許請求の範囲】 1. (式中、 R1は水素又は水酸基である、 R2は水素である、 或は、R1とR2は一緒になってR1及びR2を有する炭素原子間の第二結合を形成 する、 R3は−COOH又は−COOR4である、 R4は1ないし6炭素原子を有する、 A、B及びDは同一又は異なり得て、水素、ハロゲン、アルキル、水酸基、ア ルコキシ又は他の置換基からなる群から選ばれる。) のピペラジン誘導体化合物の製造法であって、該方法は、 以下の式 の実質的に純粋な位置異性体を提供すること、及び 実施的に純粋な位置異性体を式 のピペリジン化合物によつてピペリジン誘導体化合物に変換すること、 を含む。 2.該提供が、 式 (式中、 R5は−OR6、−N(R6)2及び−SR6であり、 R6は1ないし6炭素原子を有するアルキルである。) の出発化合物を、式 の位置異性体の第一混合物を生成するのに適した条件下に、式 (式中、Xはハロゲンである。) の化合物によってアシル化すること、 位置異性体の第一混合物を、式 の位置異性体の第二混合物を形成するのに適した条件下に加水分解すること、及 び 位置異性体の第二混合物から式 の実施的に純粋な位置異性体を回収すること、 を含む、請求項1の方法。 3.該回収が、 位置異性体の第二混合物から式 (式中、X+はルイス酸である。) の実質的に純粋な位置異性体塩を結晶化すること、 実質的に純粋な位置異性体塩を分離すること、及び 実施的に純粋な位置異性体塩を式 の実施的に純粋な位置異性体に変換すること、 を含む、請求項2の方法。 4.X+がアルカリ金属塩又は様式NR7R8R9(式中、R7、R8及びR9は独 立して水素或は直鎖又は分枝の、1ないし6炭素原子のアルキル、或はいずれか の位置でフェニル環又は置換フェニル環によって置換されたアルキルである。) のアンモニウム塩である、請求項3の方法。 5.式中、X+がシンコニジンであり、Aが水素である、請求項4の方法。 6.該アシル化を、AlCl3触媒を用いるフリーデルークラフツ反応により実 施する、請求項2の方法。 7.該提供が、式 (式中、X1はハロゲン、トリアルキル錫、トリフラート又は有機金属カップリ ング反応に有用な置換基である。) の出発化合物を、式 の実質的に純粋な位置異性体を生成するのに適した条件下に、式 の化合物によってアシル化することを含む、請求項1の方法。 8.該提供が、式 (式中、R5は−OR6、−N(R6)2及び−SR6であって、R6は1ないし6炭素 原子のアルキルである。) の出発化合物を、式 の位置異性体の第一混合物を生成するのに適した条件下に、式 の化合物でアシル化すること、 式 の位置異性体の第二混合物を形成するのに適した条件下に位置異性体の第一混合 物を加水分解すること、及び 位置異性体の第二混合物から、以下の式 の実質的に純粋な位置異性体を回収すること、 を含む、請求項1の方法。 9.該回収が、 位置異性体の第二混合物から、式 (式中、X+はルイス酸である。) の実質的に純粋な位置異性体塩を結晶化すること、 実質的に純粋な位置異性体塩を分離すること、及び 実質的に純粋な位置異性体塩を、式 の実質的に純粋な位置異性体に変換すること、 を含む、請求項8の方法。 10.X+がアルカリ金属塩又は様式NR7R8R9(式中、R7、R8及びR9は 、水素又は1ないし6炭素原子の直鎖又は分枝のアルキル、或はいずれかの位置 で フェニル環又は置換フェニル環によって置換されたアルキルである。)のアンモ ニウム塩である、請求項9の方法。 11.X+がシンコニジンであり、Aが水素である、請求項10の方法。 12.該アシル化をパラジウム、ニッケル又はその混合物を含有する触媒を使 用して実施する、請求項8の方法。 13.式 のヒドロキシル化ピペリジン誘導体を形成するのに適した条件下にピペリジン誘 導体を還元することをさらに含む、請求項1の方法。 14.ヒドロキシル化ピペリジン誘導体が、式 を有する、請求項13の方法。 15.ヒドロキシル化ピペリジン誘導体が、式 を有する、請求項13の方法。 16.該変換が、以下の式 の実質的に純粋な位置異性体を、式 (式中、Xはハロゲンである。) の第一中間体化合物を形成するのに適した条件下にハロゲン化すること、及び 第一中間体化合物を、以下の式 のピペリジン誘導体を形成するのに適した条件下に式 のピペリジン化合物と反応させること、 を含む、請求項1の方法。 17.該変換が、以下の式 の実質的に純粋な位置異性体を、式 のピペリジン誘導体を形成するのに適した条件下に式 のピペリジン化合物と反応させることを含む、請求項1の方法。 18.ピペリジン誘導体が、式 を有する、請求項1の方法。 19.ピペリジン誘導体が、式 を有する、請求項1の方法。 20.式 又は (式中、 R1は水素又は水酸基である、 R2はハロゲンである、 或はR1とR2は一緒になってR1及びR2を有する炭素原子間の第二結合を形成す る、 R3は−COOH又は−COOR4である、 R4は1ないし6炭素原子を有するアルキルである。 A、B及びDはそれらの環の1又はそれ以上の異なる置換基であることができ 、それぞれ水素、ハロゲン、アルキル、水酸基、アルコキシ又は他の置換基であ る。)の実質的に純粋なピペリジン誘導体化合物、又はその塩。 21.化合物が、式 を有する、請求項20の実質的に純粋なピペリジン誘導体化合物。 22.化合物が、式 を有する、請求項20の実質的に純粋なピペリジン誘導体化合物。 23.化合物が、式 を有する、請求項20の実質的に純粋なピペリジン誘導体化合物。 24.化合物が、式 を有する、請求項23の実質的に純粋なピペリジン誘導体化合物。 25.化合物が、式 を有する、請求項20の実質的に純粋なピペリジン誘導体化合物。 26.化合物が、式 を有する、請求項25の実質的に純粋なピペリジン誘導体化合物。 27.医薬担体及び請求項20の実質的に純粋なピペリジン誘導体化合物を含 む医薬組成物。 28.該実質的に純粋なピペリジン誘導体化合物が有効な抗アレルギー量で存 在する、請求項27の医薬組成物。 29.該組成物が本質的に該実質的に純粋なピペリジン誘導体化合物からなる 、請求項27の医薬組成物。 30.患者に、請求項27の該医薬組成物を有効量、投与することを含む、患 者のアレルギー反応を処置する方法。 31.式 (式中、 R3は−COOH又は−COOR4である、 R4は1ないし6炭素原子を有するアルキルである、 Aはその環の置換基であって、その各々は同一又は異なり得て、水素、ハロゲ ン、アルキル、水酸基、アルコキシ又は他の置換基からなる群から選択される。 )の化合物。
