【発明の属する技術分野】本発明は新規な修飾ポリペプ
チド化合物及びその用途に係る。本発明による修飾ポリ
ペプチド化合物は天然型カルシトニン及び合成カルシト
ニン・アナログと同様な生物学的活性を示し且つ安定性
において天然型カルシトニンや合成カルシトニン・アナ
ログよりも優れており、従って医薬品成分としてカルシ
ウム代謝系疾患の予防及び治療に、例えば骨粗鬆症にお
ける疼痛の軽減や骨量減少の抑制、骨形成不全症におけ
る骨折の予防、高カルシウム血症及び骨ぺージェット病
(Paget's disease) の予防及び治療に用いることができ

【従来の技術】カルシトニンは血中カルシウム濃度を低
下させる作用を有するペプチドホルモンであり、既にサ
ケ、ウナギ、アカエイ、ニワトリ、ブタ、ラット、ヒツ
ジ及びヒト由来のものが抽出精製され、それらのアミノ
酸一次配列が明らかにされている。これらのカルシトニ
ンに共通な構造的特徴は何れも 32 個のアミノ酸から構
成され、1 位と 7 位の Cys がジスルフィド結合して環
状構造を呈しており、C末端がプロリン・アミドとなっ
ている点である。これらのカルシトニン類を得るには主
として化学的合成法が行なわれてきたが、反応工程が多
く、合成反応や精製に長時間を要するため、廉価に且つ
大量にカルシトニン類を提供することは極めて困難であ
った。然るに、本出願人会社の研究者等が開発した方法
(特開平 4 - 59795 公報)、即ち、リーダー・ペプチド
に相当する DNA フラグメントを N 末端側に配し、一方
C 末端側には、必要に応じスペーサー・ペプチドをコ
ードする DNA フラグメントを介して複数個のカルシト
ニン又はそのアナログに相当する遺伝子をタンデムに結
合し、このようにして且つ二本鎖になした DNA フラグ
メントをプラスミド等のベクターに組込み、この遺伝子
組換えベクターにて微生物、例えば大腸菌を形質転換さ
せ、この形質転換体を培養することにより融合蛋白乃至
封入体として産生させ、ブロムシアンにて処理すること
によりメチオニン (Met) 部分で切断して個々のフラグ
メントに分離させると共に各フラグメントの C 末端を
ホモセリン (Hse) 又はホモセリン・ラクトン残基に変
じ、次いで酸処理を行った後にアンモニアと反応させる
ことにより C 末端ホモセリン残基をアミド化して (Hse
・NH2) 所望の生物学的活性を有するカルシトニン・ア
ナログになす方法を用いれば、廉価に且つ大量に生産で
