- 【特許請求の範囲】 1.デバイス内の反応チャンバーと検出チャンバーの間で流体サンプルを移送す る方法であって、 a)反応チャンバーと検出チャンバーをもつデバイスを準備し、反応チャンバー と検出チャンバーをこれらのチャンバーの間の流体連通手段によって相互に連結 し、前記反応チャンバー内に反応サンプルを配置し、推進剤とサンプルが混合す るように又はサンプルが推進剤と流体連通手段の間に位置するように前記反応チ ャンバー内に推進剤も配置され、更に前記推進剤が膨張するように誘導可能であ る段階と、 b)推進剤を膨張するように誘導してより大きい容積を占有させ、サンプルを流 体連通手段を介して検出チャンバーに圧入する段階とを含む前記方法。 2.前記反応チャンバーが細長形チャンバーであり、前記誘導する段階が前記推 進剤に熱を加える段階を含む請求項1に記載の方法。 3.前記熱が前記細長形反応チャンバーの推進剤を含む局在領域に加えられる請 求項2に記載の方法。 4.前記推進剤が前記反応サンプルと混合するように前記反応チャンバー内に配 置される請求項3に記載の方法。 5.前記推進剤は該推進剤と流体連通手段の間にサンプルが位置するように前記 反応チャンバー内に配置される請求項3に記載の方法。 6.前記反応チャンバーが核酸増幅用熱サイクリングチャンバーであり、前記誘 導する段階が熱サイクリング反応の完了後に実施される請求項1に記載の方法。 7.前記反応チャンバーが核酸増幅用細長形熱サイクリングチャンバーであり、 前記誘導する段階が前記推進剤を加熱する段階を含む する段階が前記推進剤を加熱する段階を含む請求項1に記載の方法。 8.前記推進剤が前記反応サンプルと混合するように前記反応チャンバー内に配 置される請求項7に記載の方法。 9.前記推進剤は該推進剤と流体連通手段の間にサンプルが位置するように前記 反応チャンバー内に配置される請求項7に記載の方法。 10.前記推進剤が蒸発可能な流体である請求項1に記載の方法。 11.前記推進剤が蒸発可能な流体である請求項7に記載の方法。 12.前記蒸発可能な流体が反応サンプル自体である請求項1に記載の方法。 13.前記蒸発可能な流体が反応サンプル自体である請求項8に記載の方法。 14.前記推進剤が空気である請求項1に記載の方法。 15.前記推進剤が空気である請求項9に記載の方法。 16.前記誘導する段階が非機械的手段によって実施される請求項1に記載の方 法。 17.前記誘導する段階が非機械的手段によって実施される請求項6に記載の方 法。 18.前記誘導する段階が前記推進剤を蒸発させることによって実施され、前記 蒸発が前記推進剤中で核形成部位を使用することによって局在化される請求項1 0に記載の方法。 19.前記核形成部位が微粒状物質を含む請求項18に記載の方法。 20.前記核形成部位がガラス又はプラスチックマイクロビーズを含む請求項1 8に記載の方法。 21.前記反応チャンバーが少なくとも二つの異なる長手方向セグメントをもつ 細長形チャンバーであり、更に、 a)第1の長手方向セグメントを断続的に加 熱して前記熱サイクリング反応を実施する段階と、 b)別の長手方向セグメントを加熱して前記推進剤の膨張を誘導し、前記反応サ ンプルを前記検出チャンバーに移送する段階とを含む請求項6に記載の方法。 22.前記反応チャンバーが長手方向セグメントをもつ細長形チャンバーであり 、更に、 a)前記長手方向セグメントに第1の最大量の熱を断続的に加えて前記熱サイク リング反応を実施する段階と、 b)前記長手方向セグメントに前記第1の最大量を上回る熱を加えて前記推進剤 の膨張を誘導し、前記反応サンプルを前記検出チャンバーに移送する段階とを含 む請求項6に記載の方法。 23.前記熱サイクリング反応がリガーゼ連鎖反応又はポリメラーゼ連鎖反応を 含む請求項6に記載の方法。 24.前記熱サイクリング反応がリガーゼ連鎖反応又はポリメラーゼ連鎖反応を 含む請求項7に記載の方法。 25.標的核酸を含む疑いのあるサンプルと複数のプライマー又はプローブを反 応チャンバー内で反応させて前記標的核酸を増幅する段階と、前記サンプルを前 記反応チャンバーと別個の検出領域に移送し、増幅標的の存在又は量を検出する 段階とを含むところの標的核酸の増幅及び検出方法において、前記サンプルを前 記反応チャンバーから前記検出領域に推進するために熱を使用することによって 前記サンプルを検出領域に移送する段階を含む改良。
