【発明の詳細な説明】
抗B型肝炎ウイルス活性を有するヌクレオシド本発明の背景
本発明は、ここに開示した活性化合物の一つ以上、またはこれら化合物の一つ
の薬学的に許容し得る誘導体もしくはプロドラッグの有効量の投与を含む、B型
肝炎ウイルス(HBVとも呼ばれる)の処置方法の分野にある。
HBVはダバコに次ぐ2番目のヒト癌の原因である。HBVが癌を誘発するメ
カニズムは知られていないが、それは癌発生を直接引金するか、または感染に関
連した慢性炎症、硬変および細胞再生を通じて間接的に癌発生を引金するものと
推定されている。
B型肝炎は世界的に流行性レベルに達している。宿主が感染に気付かない2な
いし6ケ月のインキュベーション後、HBV感染は腹痛、黄疸およびある種の酵
素の上昇した血中レベルを発生する急性肝炎および肝臓損傷へ発展し得る。HB
Vは肝臓の大量部分が破壊される急速に進行し、時には死に至る疾病の形である
、劇症肝炎を引き起こし得る。患者は典型的には急性ウイルス性肝炎からは回復
する。しかしながら患者の中には血中に高レベルのウイルス抗原が長期間または
永久に持続し、慢性肝炎を発生する。慢性感染は慢性持続性肝炎へ発展し得る。
慢性持続性HBVに感染した患者は発展途上国において最も普通である。199
1年半ばまでに、アジアだけでも約2億2千5百万人、全世界では殆ど3億人の
HBVキャリヤーが存在した。慢性持続的肝炎は疲労、肝硬変、および一次肝臓
癌である肝細胞カルチノーマを引き起こし得る。
西洋工業国においては、HBV感染の高リスクグループはHBVキャリヤーま
たは彼等の血液サンプルと接触にある人々を含む。HBVの流行性は事実後天性
免疫不全症候群に非常に似ており、このことはHBVが何故エイズまたはHIV
関連感染を有する患者間に普通であるかを説明する。しかしながらHBVはHI
Vよりも伝染性である。
遺伝子工学でつくられたタンパクであるα−インターフェロンによる連日処置
は有望であることを示している。ヒト血清から得られたワクチンも患者をHBV
に対し免疫するために開発されている。ワクチンは遺伝子工学によってつくられ
ている。ワクチンは有効であることが発見されているが、ワクチンの製造は、慢
性キャリヤーからのヒト血清の供給が限られ、そして精製操作が長くそして高価
である故に厄介である。さらに異なる血清からつくられたワクチンのバッチ毎に
その安全性を確かめるためにチンパンジーでテストしなければならない。加えて
、ワクチンはウイルスに既に感染した患者を助けない。
ヨーロッパ特許出願No.92304530.6は、1,2−オキサチオランヌ
クレオシドの一グループがB型肝炎感染の処置に有用であることを開示する。2
−ヒドロシメチル−5−(シトシン−1−イル)−1,3−オキサチオランが抗

B型肝炎ウイルスは全世界で流行レベルに達していること、および感染患者に
対して重篤な時には非劇的効果を有する事実に鑑み、このウイルスに感染したヒ
トを処置するための、宿主に対し低い毒性を有する新しい効果的薬剤を提供する
ことに強い必要性が依然存在する。
それ故、本発明の他の一目的はHBVに感染したヒト患者または他の宿主の処
置のための方法および組成物を提供することである。本発明の概要

ヌクレオシドは指示したエナンチオマーとしてそしてその対応するエナンチオマ

の化合物のβ−L−エナンチオマーがここにもっと完全に記載されるように宿主
へHBV処置量において投与される。式中R5は、アルキル化またはハロゲン化
プリンを含む、アデニン、キサンチン、ヒポキサンチンまたは他のプリンである
。
他の具体例においては、本発明は特定的に記載したヌクレオシドのプロドラッ
グのHBV処置量を投与することを含むHBVに感染したヒトの処置方法を含ん
でいる。
ここでいうプロドラッグとは、生体内投与において該ヌクレオシドへ変換され
る、またはそれ自身活性を持っている特定的に記載したヌクレオシドの薬学的に
許容し得る誘導体をいう。非限定な例は
活性化合物の5’およびN4−ピリミジンまたはN6−プリンアシル化もしくはア
ルキル化誘導体である。
記載したヌクレオシド、またはそれらの薬学的に許容し得るプロドラッグもし
くは塩、またはこれら化合物を含んでいる薬学的に許容し得る製剤はHBV感染
および抗HBV抗体陽性およびHBV陽性状態、HBVによって発症した慢性肝
炎、急性肝炎、劇症肝炎、慢性持続性肝炎および疲労のような他の関連する状態
の予防および処置に有用である。これらの化合物または製剤は、抗HBV抗体も
しくはHBV抗原陽性である、またはHBVへ曝露された個人の臨症病状の進行
を予防または阻止するため予防的に使用することもできる。
本発明の一具体例においては、前記活性化合物の一つ以上は、効果的な抗HB
V処置を提供するため一つ以上の他の抗HBV剤と交替にまたは併用して投与さ
れる。交替または併用治療に使用できる交HBV剤の例は、2−ヒドロキシメチ
ル−5−(5−フルオロシトシン−1−イル)−1,3−オキサチオラン(FT
C,WO92/14743を見よ)の(−)−エナンチオマーまたはラセミ混合
物またはその生理学的に許容し得る誘導体または生理学的に許容し得る塩、2−
ヒドロキシメチル−5−(シトシン−1−イル)−1,3−オキサチオラン(B
CH−189または3TCとも呼ばれる。EPA公報No.0382526を見よ
)の(−)−エナンチオマーもしくはラセミ混合物、その生理学的に許容し得る
誘導体または生理学的に許容し得る塩、2’−フルオロ−5−ヨードアラビノシ
ルウラシル(FIAU)のエナンチオマーまたはラセミ混合物、2’−フルオロ
−5−エチルアラビノシルウラシル(FEAU)のエナ
ンチオマーまたはラセミ混合物、カルボビル、またはインターフェロンを含むが
これらに限定されない。
患者へ処置を提供する任意の交替方法を使用できる。交替パターンの非限定例
は、一つの剤の有効量の1〜6週投与の後、第2の抗HBV剤の有効量の1〜6
週投与を含む。交替スケジュールは処置なしの期間を含む。併用療法は一般に二
つ以上の抗HBV剤の有効投与量比の同時投与を含む。
HBVは抗HIV抗体またはHIV抗原陽性の、またはHIVに曝露された患
者にしばしば発見される事実に鑑み、ここに記載した活性抗HBV化合物または
それらの誘導体またはプロドラッグは、適切な状況においては、3’−アジド−
3’−デオキシチミジン(AZT),2’,3’−ジデオキシイノシン(DDI
),2’,3’−ジデオキシシチジン(DDC),2’,3’−ジデオキシ−2
’,3’−ジデヒドロチミジン(D4T),2−ヒドロキシメチル−5−(5−
フルオロシトシン−1−イル)−1,3−オキサチオラン(FTC),または2
−ヒドロキシメチル−5−(シトシン−1−イル)−1,3−オキサチオラン(
BCH−189)のラセミもしくはエナンチオマー形を含む抗HIV剤投与との
併用または交替において投与することができる。化合物のTiboクラス、ネビ
ラピンまたはピリミジノンのような非ヌクレオシドRT阻害剤もクレームした化
合物と併用して投与することができる。
この活性抗HBV剤は抗生物質、他の抗ウイルス化合物、抗カビ剤、または二
次感染の処置のために投与される他の薬剤と併用して投与することができる。図面の簡単な説明

ここで使用する「エナンチオマー的に純粋」なる用語は、少なくとも約95%
,そして好ましくは約97%,98%,99%または100%のヌクレオシドの
単一のエナンチオマーを含んでいるヌクレオシド組成物を意味する。
ここで使用する「アルキル」なる用語は、特にC1ないしC10のメチル、エチ
ル、プロピル、ブチル、ペンチル、ヘキシル、イソプロピル、イソブチル、se
c−ブチル、t−ブチル、イソペンチル、アミル、t−ペンチル、シクロペンチ
ルおよびシクロヘキシルを含むがそれらに限らない。
ここで使用する「アシル」なる用語は、特にアセチル、プロピオニル、ブチリ
ル、ペンタノイル、3−メチルブチリル、ヒドロゲンスクシネート、3−クロロ
ベンゾエート、ベンゾイル、アセチル、
ピバロイル、メシレート、プロピオニル、バレリル、カプロニル、カプリロイル

ここに開示する発明は、一つ以上の上で同定した化合物、またはその生理学的
に許容し得る誘導体、または生理学的に許容し得る塩の有効量を場合により薬学
的に許容し得る担体中において投与することを含む、ヒトまたは他の宿主動物に
おけるHBV感染および同様な態様で複製されるウイルスの処置のための方法お
よび組成物である。本発明の化合物は、抗HBV活性を有するか、または抗HB
V活性を発揮する化合物へ代謝される。
I.活性ヌクレオシドの構成および製造立体化学
ここに開示する方法に使用される化合物は、2’,3’−ジデオキシシチジン
、2’,3’−ジデオキシ−5−(ハロもしくはメチル)シチジン、2−ヒドロ
キシメチル−5−(5−フルオロシトシン−1−イル)−1,3−ジオキソラン
、または2−アミノ−6−(OH,Cl,NH2,またはH)−9−〔(4−ヒ
ドロキシメチ
ル)−テトラヒドロフラン−1−イル〕プリンのエナンチオマーである。
糖またはジオキサラニル部分(以下一般的に糖部分という)の1’−および4
’炭素はキラルであるため、それらの非水素置換基(それぞれCH2ORおよび
ピリミジンまたはプリン塩基)は、糖リング系に関してシス(同じ側)またはト
ランス(反対側)であり得る。それ故4種の光学異性体は以下の立体構造(糖部

HBV感染を処置するための開示した方法において有用なヌクレオシドは、F
DOCのβ−D−エナンチオマーは驚くべきことにFDOCのβ−L−エナンチ
オマーより毒性が少ないことが発見されたためFDOCを除いてβ−L−エナン

2’,3’−ジデオキシシチジン(DDC)は既知化合物である。DDCのD
−エナンチオマーはホフマン、ラロシュによってHIVに感染した人々の処置に
使用のため現在ZALCITABINEの名称で市販されている。米国特許No
s.4,879,277および4,900,828を見よ。
β−D−FDOCのようなエナンチオマー的に純粋なβ−D−ジオキソランヌ
クレオシドはPCT/US91/09124に詳細に開示されているようにして
製造することができる。このプロセスは、糖の1位(これは後で生成したヌクレ
オシドの4’−位になる)について正しいジアストレオマー構造を含む、エナン
チオマー的に純粋な最終生成物のための必要な主体化学のすべてを含んでいる糖
である、(2R,4R)−および(2R,4S)−4−アセトキシ−2−(保護
したオキシメチル)−ジオキソランを1,6−アンヒドロマンノースを最初に合
成することを含んでいる。(2R,4R)−および(2R,4S)−4−アセト

活性ヌクレオシドのモノ,ジおよびトリリン酸誘導体は発表された方法によっ
て製造することができる。モノリン酸エステルはImai et al.,J.

II.ジオキソランヌクレオシドの抗HBV活性
活性化合物がHBVを阻害する能力は種々の実験技術によって測定することが
できる。開示した化合物がHBVの複製を阻止する能力を評価するためここで使
用したアッセイは、Korba and Gerin,Antiviral R
es.,19:55−70(1992)に詳細に記載されている。例証のみの目
的のため、そ
して本発明を限定することなく、β−L−2’,3’−ジデオキシシチジン(β
−L−ddC),および(+)−β−D−2−ヒドロキシメチル−5−(5−フ
ルオロシトシン−1−イル)−1,3−ジオキソラン((+)−β−D−FDO
C)の毒性および抗HBV活性の評価結果が以下に与えられる。(−)−β−L
−2−ヒドロキシメチル−5−(5−フルオロシトシン−1−イル)−1,3−
ジオキソラン(C−)−β−L−FTC)およびβ−D−2’,3’−ジデオキ
シシチジン(β−D−ddC)の毒性および抗HBV活性は対照として含まれて
いる。ここに記載した他の化合物も同様にして評価することができる。
抗HBVアッセイに使用したβ−L−ddCおよびβ−L−5−FddCのサ
ンプルは以下のとおり特徴化された。
抗ウイルス性評価は、1回の実験あたり2培養で細胞の2回の別々の実験(全
部で4培養)で実験した。すべてのプレート中のすべてのウエルは同じ密度でそ
して同時に接種された。
細胞内および細胞外HBV DNAのレベルの本来の変動のため、3.0倍(
HBVビリオンDNAについて)または2.5倍(HBV DNA複製中間体に
ついて)より大きい未処理細胞中のこれらのHBV DNA形の平均レベルより
大きい低下だけを一般に統計的に有意である(P<0.05)と考えた(Kor
ba and Gerin,Antiviral Res.19:55−70,
1992)。各細胞DNA調製物中の統合したHBV DNAのレベル(これら
実験においては細胞あたりベースにおいてそれらはコンスタントに保たれる)が
細胞内HBV DNA形のレベルを計算するために使用され、それによってブロ
ットハイブリダイゼーションアッセイにつきものの技術的変動を排除した。
未処理細胞中の細胞外HBVビリオンDNAについての典型的値は50ないし
150pg/ml培地(平均約76pg/ml)である。未処理細胞中のHBV D
NA複製中間体は50ないし100pg/μg細胞DNA(平均約74pg/μ
g細胞DNA)範囲である。一般に抗ウイルス性化合物の処理による細胞内HB
V DNAレベルの低下は、HBVビリオンDNAのレベルの低下よりもより顕
著ではなく、そしてより遅く発生する。
参照のため、ハイブリダイゼーション分析がこれらけの実験のために実施され
た態様は、2〜3ゲノムコピー/細胞に対して約1.0pg細胞内HBV DN
A/μg細胞DNAおよび3×105ウイルス粒子/mlに対し1.0pg細胞外
HBV DNA/ml培地の
当量を与える。
どんな観察された抗ウイルス効果も細胞生存性に対する一般的効果かどうかを
評価するために毒性分析が実施された。使用した方法は、HSV(herpes
simplex ウイルス)およびHIVを含む種々のウイルス/宿主系にお
ける細胞生存性のための標準的な広く使用されるアッセイである、中性赤色染料
の摂取に基いていた。
テスト化合物はDMSO中40mMストック溶液の形(ドライアイス上で凍結
)で使用された。
テストサンプルの日常部分標本は、めいめいの個々の部分標本が1回だけの凍
結解凍サイクルを受けるように−20℃にされ、凍結された。日常部分標本は解
凍され、室温で媒地中に懸濁され、そして直ちに細胞培養物へ添加された。化合
物は抗ウイルス活性については0.01ないし10μMにおいてテストされた。
化合物は1ないし30μMの濃度において毒性のためにテストされた。結果は表
1に与えられている。
実施例2 化合物の毒性
2.2.15細胞培養物(肝炎ビリオンで形質転換したHepG2細胞)中の
ウイルスの成育を阻止する活性化合物の能力を評価した。表1に示すように、1
00μMの濃度ではテスト化合物のどれについても有意な毒性(未処理細胞中で
見られる染料摂取レベルの50%以上の低下)が見られなかった。化合物は30
0μMにおいて中程度毒性であったが、すべての3化合物はβ−D−ddCより
もこの濃度において少ない毒性を示した。β−L−ddCおよびβ−L−Fdd
CのIC50はβ−D−ddCのそれの約2倍であることが見える。
毒性分析は96ウエル平底組織培養プレートで実施された。毒性分析のための
細胞が培養され、そして抗ウイルス性評価に使用したのと同じスケジュールでテ
スト化合物で処理された。各化合物は各自3培養について4種の濃度においてテ
ストされた。中性赤色染料の摂取が毒性の相対的レベルを決定するために使用さ
れた。510nM(A510)において内部化された染料の吸収を定量分析に使用
した。値は、テスト化合物と同じ96ウエルプレート上に維持した未処理細胞の
9個の別の培養物中の平均A510値(±標準偏差)のパーセントとして提供され
る。プレート40上の9対照培養物中の染料摂取のパーセントは100±3であ
った。150−190μMβ−D−ddCにおいては染料摂取の2倍の減少(未
処理培養物に見られるレベルに対して)がこれらのアッセイにおいて典型的に観
察される(Korba and Gerin,Antiviral Res.1
9:55−70,1992)。
実施例3 抗B型肝炎ウイルス活性
陽性処理対照、β−D−2’,3’−ジデオキシシトシン(β−D−ddC)
は、使用した濃度においてHBV DNA複製の有意な低下を誘発した。以前の
研究は、β−D−ddCの9〜12μMにおいてHBV RIの90%低下(未
処理細胞中の平均レベルに関して)がこのアッセイシステムにおいて観察される
ことを示している(Korba and Gerin,Antiviral R
es.19、55−70,1992)。これは表1に提示したデータと一致する
。
表1に提示したデータは、すべての3化合物(β−L−FddC,β−L−d
dCおよびβ−D−FDOC)はHBV複製の強力な阻害剤であり、HBVビリ
オンDNA複製をβ−D−ddC処理後に観察されるそれと匹敵する、またはそ
れよりも大きく低下させる。
III.薬剤組成物の製造
ここに開示した化合物およびそれらの薬学的に許容される塩、プロドラッグ、
および誘導体は、HBV感染、および抗HBV抗体陽性およびHBV陽性状態、
HBVによって発生した慢性肝臓炎症、黄疸、急性肝炎、劇症肝炎、慢性持続性
肝炎および疲労のような他の関連する状態の予防および処置に有用である。これ
ら化合物または製剤は、抗HBV抗体またはHBV抗原陽性である、またはHB
Vへ曝露された個人の臨床的疾病の進行を予防もしくはおくらせるために予防的
に使用することもできる。
これら症状に罹っているヒトは、ここに記載した活性化合物の一つまたは混合
物、またはそれらの薬学的に許容し得る誘導体もしくは塩のHBV処置量を場合
により薬学的に許容し得る担体もしくは
希釈剤中において患者へ投与することによって処置することができる。活性化合
物は任意の適切なルートにより、例えば経口、非経口、静脈内、皮内、皮下また
は局所的に液体または固体形で投与することができる。
活性化合物は、処置される患者に重大な毒性効果を生ずることなく治療的に有
効な量を患者へ放出するのに十分な量において薬学的に許容し得る担体または希
釈剤に含められる。
前述した状態のすべてのための活性化合物の好ましい投与量は、1日あたり約
1ないし60mg/kg体重,好ましくは1ないし20mg/kg,さらに一般的には1
日あたり0.1ないし約100mg/kg体重の範囲であろう。薬学的に許容し得る
誘導体の有効投与量範囲は放出すべき親ヌクレオシドの重量に基いて計算するこ
とができる。もし誘導体自身が活性を示すならば、有効投与量は誘導体の重量を
使用して上のようにして、または当業者に既知の他の手段によって推計すること
ができる。一具体例において、活性化合物は、HIV効能についての3’−アジ
ド−3’−デオキシチミジ(AZT),2’−3’−ジデオキイノシン(DDI
),2’−3’−ジデオキシシチジン(DDC),または2’,3’−ジデヒド
ロチミジン(D4T)のための製品能書または医師デスク参考文献に記載されて
いるように投与される。
化合物は、単位投与形あたり活性成分7ないし3000mg,好ましくは70な
いし1400mgを含んでいる任意の適当な単位投与形で便利に投与される。
理想的には、活性成分は活性化合物のピーク血漿濃度約0.2ないし70μM
,好ましくは約1.0ないし10μMを得るように投
与すべきである。これは、例えば場合により食塩水中の活性成分の0.1ないし
5%溶液の静注により、または活性成分のボーラスとして投与により達成するこ
とができる。
活性化合物は薬学的に許容し得る塩の形で提供することができる。ここで使用
する薬学的に許容し得る塩または複塩なる用語は、親化合物の所望の生物学的活
性を保持し、そしてもしあっても最低の望ましくない毒性効果を発揮する塩また
は複塩をいう。そのような塩の非限定例は、(a)無機酸(例えば塩酸、臭化水
素酸、リン酸、硝酸等)で形成した酸付加塩、および酢酸、シュウ酸、酒石酸、
コハク酸、リンゴ酸、アスコルビン酸、安息香酸、タンニン酸、パモエ酸、アル
ギン酸、ポリグルタミン酸、ナフタレンスルホン酸、ナフタレンジスルホン酸、
およびポリガラクツロン酸のような有機酸で形成した酸付加塩、(b)ナトリウ
ム、カリウム、亜鉛、カルシウム、ビスマス、バリウム、マグネシウム、アルミ
ニウム、銅、コバルト、ニッケル、カドミウム等のカチオンで、またはN,N−
ジベンジルエチレンジアミン、アンモニウムまたはエチレンジアミンから形成し
た有機カチオンで形成した塩基付加塩、または(c)(a)と(b)の組合せ例
えばタンニン酸亜鉛塩等である。
活性化合物の修飾、特にN6またはN4および5’−0位における修飾は活性種
のバイオアベイラビリティおよび代謝速度に影響し、活性種の放出の制御を提供
することができる。
薬物組成物中の活性化合物の濃度は、薬物の吸収、不活性化、および排泄速度
、それに当業者に既知の他のファクターに依存するであろう。投与量値は緩和す
べき状態の重篤度によっても変化することを留意すべきである。どの特定の対象
についても、特定の投与量
計画は個人の必要性および組成物を投与する人またはそれを監督する人の職業的
判定に従って時間的に調節すべきこと、そしてここに述べた濃度範囲は例示であ
って特許請求した組成物の範囲もしくは実際を限定することを意図しないことを
理解すべきである。活性成分は一度に投与するか、または種々の時間間隔で投与
すべきより少量の投与に分割することができる。
活性化合物の好ましい投与モードは経口である。経口組成物は一般に不活性希
釈剤または食し得る担体を含んでいる。それらはゼラチンカプセル中に包むかま
たは錠剤に圧縮することができる。経口治療投与の目的のため、活性化合物は賦
形薬と混合され、そして錠剤、トローチまたはカプセルの形で使用される。薬学
的に許容し得る結合剤および/またはアジュバンド材料を組成物の一部として含
めることができる。
錠剤、丸剤、カプセル、トローチ等は以下の成分または同様な性格の化合物を
含むことができる。微結晶性セルロース、トラガントゴムまたはゼラチンのよう
なバインダー;デンプンまたは乳糖のような賦形剤;アルギン酸、プリモゲルま
たはコーンスターチのような崩壊剤;ステアリン酸マグネシウムまたはステロー
トのような滑沢剤、ショ糖またはサッカリンのような甘味剤;ペパーミント,サ
リチル酸メチル、またはオレンジ香料のような香料。投与単位形態がカプセルの
時、それは上のタイプの材料に加え、脂肪油のような液状担体を含むことができ
る。加えて投与単位形態は投与単位の物理的形態を修飾する種々の他の材料、例
えば糖衣、シエラックまたは他の腸溶剤のコーティングを含むことができる。
活性化合物またはその薬学的に許容し得る塩もしくは誘導体は、
エリキサー、懸濁液、シロップ、ウエハー、チューインガム等の成分として投与
することができる。シロップは活性成分に加え、甘味剤としてショ糖およびある
種の防腐剤、染料および着色料、および香料を含んでもよい。
活性化合物、またはその薬学的に許容し得る誘導体もしくは塩は所望の作用を
損なわない他の活性物質、または所望の作用を補完する物質、例えば抗生物質、
抗カビ剤、抗炎症剤、または抗HBV、抗サイトメガロウイルスもしくは抗HI
V剤を含む他の抗ウイルス剤と混合することもできる。
非経口、皮内、皮下または局所適用のために使用される溶液または懸濁液は以
下の成分を含むことができる。注射用蒸留水、食塩水、固定化油、ポリエチレン
グリコール、グリセリン、プロピレングリコールまたは他の合成溶媒のような無
菌希釈剤;ベンジルアルコールまたはメチルパラベンのような抗バクテリア剤;
アスコルビン酸または重亜硫酸ナトリウムのような抗酸化剤;エチレンジアミン
テトラ酢酸のようなキレート剤;酢酸塩、クエン酸塩またはリン酸塩のような緩
衝剤および塩化ナトリウムまたはデキストロースのような等張化剤。非経口製剤
はガラスもしくはプラスチック製のアンプル、使い捨てシリンジまたは多数投与
バイアルに包装することができる。
もし静脈投与するならば、好ましい担体は生理食塩水またはリン酸緩衝化食塩
水(PBS)である。好ましい具体例において、活性化合物は、インプラントお
よびマイクロカプセル化放出システムを含む制御された放出製剤のような、体内
からの急速な排除に対して化合物を保護する担体と製剤化される。エチレン−酢
酸ビニル、ポ
リ無水物、ポリグリコール酸、コラーゲン、ポリオルソエステルおよびポリ乳酸
のような生分解性の生体適合性ポリマーを使用することができる。そのような製
剤の製造方法は当業者には明らかであろう。このような材料はAlza Cor
poration およびNova Pharmaceuticals,Inc
から商業的に得ることもできる。
リポソーム懸濁液(ウイルス抗原に対するモノクローナル抗体をもって感染細
胞へ標的するリポソームを含む)も薬学的に許容し得る担体として好ましい。こ
れらは、例えば米国特許No.4,522,811(これはその全体を参照として
ここに取入れる)に記載されている、当業者に既知の方法によって製造すること
ができる。例えば、リポソーム製剤は適切な脂質(ステアロイルフォスファチジ
ルエタノールアミン、ステアロイルフォスファチジルコリン、アラカドイルフォ
スファチジルコリン、およびコレステロールのような)を無機溶媒に溶解し、次
に溶媒を蒸発し、容器表面上に乾燥した脂質の薄いフィルムを残すことによって
製造することができる。活性化合物、またはそのモノリン酸エステル、ジリン酸
エステルおよび/またはトリリン酸エステル誘導体の水溶液を次に容器へ導入す
る。容器は容器の側部から脂質材料を遊離し、そして脂質凝塊を分散するために
手で渦巻し、それによってリポソーム懸濁液を形成させる。

- 【特許請求の範囲】 1.構造式: (式中、R1は水素、フッ素、臭素、塩素、沃素またはメチルであり、R2はO H,Cl,NH2またはHである。)のヌクレオシド、またはその薬学的に許容 し得る塩であって、その反対のβ−(DまたはL)エナンチオマーを含まない9 5%以上純度の化合物のHBV処置量を投与することよりなる、ヒトまたは他の 宿主動物におけるHBV感染の処置方法。 2.構造式: (式中、R1は水素、フッ素、臭素、塩素またはメチルであり、R2は水素;C1 −C20アルキル;エステル基の非カルボニル部分が直鎖、分岐もしくは環状C1 −C20アルキル、フェニルもしくはベンジルから選ばれたアシル;天然に存在す るまたは非天然存在アミノ酸;メトキシメチルを含むアルコキシアルキル;ベン ジルを含むアラルキル;フェノキシメチルのようなアリールオキシアルキル;場 合によりハロゲン,C1−C4アルキルまたはC1−C4アルコキシで置換されたフ ェニルを含むアリール;コハク酸のようなジカルボン酸;メタンスルホニルを含 むアルキルもしくはアラルキルスルホニル;またはモノ、ジもしくはトリリン酸 エステルであり、 R4は水素;C1−C20アルキル;エステル基の非カルボニル部分が直鎖、分 岐もしくは環状C1−C20アルキル;フェニルもしくはベンジルから選ばれたア シル;メトキシメチルを含むアルコキシアルキル;ベンジルを含むアラルキル; フェノキシメチルのようなアリールオキシアルキル;場合によりハロゲン,C1 −C4アルキルまたはC1−C4アルコキシで置換されたフェニルを含むアリール である。)のヌクレオシドであって、その反対のβ−(DまたはL)エナンチオ マーを含まない95%以上純度のヌクレオシドのHBV処置量を投与することよ りなる、ヒトまたは他の宿主動物におけるHBV感染の処置方法。 3.担体が経口放出に適している請求項1または2の方法。 4.担体はカプセルよりなる請求項1または2の方法。 5.担体は錠剤の形である請求項1または2の方法。 6.投与は非経口である請求項1または2の方法。 7.アルキル基はメチル、エチル、プロピル、ブチル、ペンチル、ヘキシル、イ ソプロピル、イソブチル、sec−ブチル、t−ブチル、イソペンチル、アミル 、t−ペンチル、シクロペンチルおよびシクロヘキシルよりなる群から選ばれる 請求項2の方法。 8.アシル基はアセチル、プロピオニル、ブチリル、ペンタノイル、3−メチル ブチリル、ヒドロゲンスクシネート、3−クロロベンゾエート、ベンゾイル、ア セチル、ピバロイル、メシレート、プロピオニル、バレリル、カプロニル、カプ リロイル、カプリニル、ラウリル、ミリスチル、パルミチル、ステアリルおよび オレイルよりなる群から選ばれる請求項2の方法。 9.請求項1または2のヌクレオシドのHBV処置量を、2−ヒドロキシメチル −5−(5−フルオロシトシン−1−イル)−1,3−オキサチオランの(−) −エナンチオマーまたはラセミ混合物、2−ヒドロキシメチル−5−(シトシン −1−イル)−1,3−オキサチオランの(−)−エナンチオマーまたはラセミ 混合物、2’−フルオロ−5−ヨードアラビノシルウラシル(FIAU)のエナ ンチオマーまたはラセミ混合物、2’−フルオロ−5−エチルアラビノシルウラ シル(FEAU)のエナンチオマーまたはラセミ混合物、カルボビル、またはイ ンターフェロンよりなる群から選ばれた化合物と交替投与において投与すること よりなる、ヒトまたは他の宿主動物におけるHBV感染の処置方法。 10.請求項1または2のヌクレオシドのHBV処置量を、2−ヒドロキシメチル −5−(5−フルオロシトシン−1−イル)−1,3−オキサチオランの(−) −エナンチオマーまたはラセミ混合物、2−ヒドロキシメチル−5−(シトシン −1−イル)−1, 3−オキサチオランの(−)−エナンチオマーまたはラセミ混合物、2’−フル オロ−5−ヨードアラビノシルウラシル(FIAU)のエナンチオマーまたはラ セミ混合物、2’−フルオロ−5−エチルアラビノシルウラシル(FEAU)の エナンチオマーまたはラセミ混合物、カルボビル、またはインターフェロンより なる群から選はれた化合物と併用して投与することよりなる、ヒトまたは他の宿 主動物におけるHBV感染の処置方法。 11.ヌクレオシドはβ−L−2’,3’−ジデオキシシチジン(β−L−ddC ),β−L−2’,3’−ジデオキシ−5−フルオロシチジン(β−L−Fdd C)、および(+)−β−D−2−ヒドロキシメチル−5−(5−フルオロシト シン−1−イル)−1,3−ジオキソラン((+)−β−D−FDOC)よりな る群から選ばれる請求項1の方法。 12.ヌクレオシドはβ−L−2’,3’−ジデオキシシチジン(β−L−ddC ),β−L−2’,3’−ジデオキシ−5−フルオロシチジン(β−L−Fdd C)、および(+)−β−D−2−ヒドロキシメチル−5−(5−フルオロシト シン−1−イル)−1,3−ジオキソラン((+)−β−D−FDOC)よりな る群から選ばれる請求項2の方法。
