CH2C(O)OCH3およびC18までの種々の異性体が包含される。単独でまたは 例えば「ハロアルキル」の如き複合語の中の「ハロゲン」という語は弗素、塩素 、臭素またはヨウ素を示す。さらに、例えば「ハロアルキル」の如き複合語の中 で使用される時には、該アルキルは同一もしくは相異なるハロゲン原子で部分的 または完全に置換されていてもよい。「ハロアルキル」の例には、F3C、Cl O)Oであり、そしてYはHである)は式IIの化合物と式IIIの化合物との縮合に より製造できる。この変換は反応式1に示されている。一般的に有用な方法は酸 スカベンジャー(普通には例えばトリエチルアミンの如き第3級アミン塩基)の 存在下での室温もしくはそれ以下における式IIの化合物と式IIIの化合物の反応 である。この反応は例えば塩化メチレン、テトラヒドロフラン、クロロホルム、 トルエンおよび酸塩化物または塩基と反応しない他の溶媒の如き不活性溶媒の中 で実施できる。この反応は通常は24時間以内に完了する。アミド類、スルホン アミド類およびカルバメート類の生成用の他の有用な方法は Larock,Comprehen sive Organic Transformations,VCH Publishers,Inc.,New York に論じられ ている。 反応式1 反応式2に示されているように、式Iの化合物(ここで、AはA−1であり、 G1はC(X1)N(Y)でありそしてYはHである)は式IIの化合物と式IVのイソシ アナートとの反応により製造できる。典型的な反応は例えば酢酸エチル、塩化メ チレン、テトラヒドロフラン、クロロホルム、 ベンゼンまたはトルエンの如き有機溶媒の中での等モル量のIIおよびIVの組み合 わせを含む。例えばアルカリ金属、第3級アミン、アルカリ金属水酸化物または 金属水素化物の如き塩基を使用することができる。この反応は約−20−100 ℃の範囲の温度において実施でき、−10−30℃の範囲の温度が好ましい。こ の反応は24時間以内に完了する。 反応式2 式Iの化合物(ここで、AはA−1でありそしてYはHである)は反応式3に 示されているように式IIの化合物と式Vのアルデヒドの反応により製造できる。 この反応は例えばメタノールまたはエタノールの如き溶媒中で例えばホウ水素化 ナトリウムまたはシアノホウ水素化ナトリウムの如き還元剤の存在下で実施でき る(Borch et al.,J.Am.Chem.Soc.(1981),93,2897)。反応温度は−30 −200℃に変えることができそして反応は約2−72時間で完了する。 反応式3 或いは、式Iの化合物(ここで、AはA−1であり、A−1はQCH2N(Y) でありそしてYはHである)は反応式4に示されている ような式Iの化合物(ここで、AはA−1であり、A−1はQC(O)N(Y)であ りそしてYはHである)の還元により製造できる。有用な還元剤はアルカリ金属 水素化物である。例えば、0−50℃における例えばテトラヒドロフラン、エー テルまたはジメトキシエタンの如きエーテル系溶媒中での水素化アルミニウムリ チウムを用いる式Iの化合物(AはA−1であり、G1はC(O)でありそしてY はHである)の処理で式Iの化合物(AはA−1でありそしてYはHである)を 生成する。還元は一般的には24時間以内に完了する。アミド類からアミン類へ の他の還元方法に関しては、March,Advanced Organic Chemistry 3rd Ed.,(19 85),pp 1099-1100を参照のこと。 反応式4 式IIの化合物は式VIの化合物と次亜臭素酸ナトリウム(または水酸化ナトリウ ムおよび臭素)との反応により製造できる。この変換は反応式5に示されている 。ホフマン転位の論議は Org.Rxns.(1946),3,pp.267-306 に見られる。典 型的な反応は次亜臭素酸ナトリウムの水溶液に対する式VIの化合物の添加を含む 。反応の温度は0−200℃であり、好適な温度範囲は30−100℃である。 この反応は一般的には24時以内に完了する。或いは、式VIの化合物を還流アセ トニトリル中で[ヒドロキシ(トシルオキシ)ヨード]ベンゼンを用いて処理するこ とにより この変換を行うこともできる。(J.Org.Chem.(1986),51,pp 2669-2671 参 照)。 反応式5 式VIの化合物は式VIIの化合物をアンモニアと反応させることにより製造でき る。この変換は反応式6に示されている。この反応は例えばメタノール、エタノ ール、エーテル、ベンゼンおよびトルエンの如き溶媒の中で実施できる。典型的 な反応は周囲温度において実施され、そして反応は一般的には24時間以内に完 了する。参考文献に関しては、D.W.Jones,J.Chem.Soc.(1969),1729を参 照のこと。 反応式6 式VIIの化合物は反応式7に示されているような商業的に入手できるセリン誘 導体(式VIII)と式IXのイミド酸エステルとの反応により製造できる。この反応 は例えばメタノール、塩化メチレン、クロロホルム、ベンゼン、ジオキサンおよ びテトラヒドロフランの如き溶媒の中で実施できる。水を共溶媒として加えるこ とができる。この反応は0℃から使用される特定溶媒の還流温度までで変わる温 入手できるアミド類から例えば塩化メチレン、ベンゼンまたはトルエンの如き不 活性溶媒中でのテトラフルオロホウ酸トリアルキルオキソニウムを用いる反応に より製造できる。イミド酸エステルの合成は D.A.Neilson により The Chemis try of Amidines and Imidates,Patai and Rappoport,Eds.,Vol.2,(1991) ,pp 425-483 に十分論じられている。 反応式8 式Iの化合物(ここで、AはA−4またはA−5でありそしてX2はアルコキ シまたはチオアルキルである)は反応式9に示されているよう な式Iの化合物(ここで、AはA−4またはA−5でありそしてX2はClまた はBrである)と硫黄または酸素求核剤との反応により製造できる。典型的な反 応は例えばアルカリ金属、第3級アミン、金属水素化物などの如き塩基の存在下 におけるエーテル、テトラヒドロフラン、1,2−ジメトキシエーテル、塩化メ チレン、クロロホルム、N,N−ジメチルホルムアミドおよびジメチルスルホキ シドを含む一般的な有機溶媒中での等モル量の反応物の組み合わせを含む。この 反応は−20−100℃の範囲の温度において実施でき、−10−30℃の範囲 の温度が好ましい。当技術の専門家はこの一般的なタイプの反応を他の求核剤に 拡大できることを認識するであろう。 反応式9 反応式10に示されているように、式Iの化合物(ここで、AはA−4または A−5でありそしてX2はClまたはBrである)は式Iの化合物(ここで、A はA−1またはA−2であり、G1はC(O)であるかまたはG2=NHであり、X1 はOであり、そしてYはHである)と適当なハロゲン化剤、例えば三塩化燐、 レンおよび、キサンの如き有機溶媒の存在下または不存在下における反応物と過 剰のハロゲン化剤との組み合わせを含む。好適な反応温度は35−100℃の範 囲でありそしてこの反応は一般的には24時間以内に完了する。 反応式10 式Iの化合物(ここで、AはA−5でありそしてX2はHである)は式IIの化 合物と式Vの化合物との反応により製造できる。典型的な反応は例えばアセトニ トリル、メタノール、エタノールまたはベンゼンの如き適当な溶媒中での等モル 量の式IIの化合物と式Vの化合物との組み合わせを含む。この反応は酸触媒の存 在下または不存在下で実施することができる。典型的な酸触媒にはアルキルまた はアリールスルホン酸および鉱酸、例えば塩酸、が包含される。反応温度は0℃ から使用される特定溶媒の還流温度までに変えることができる。反応式11はこ の変換を示している。この反応は通常は24時間以内に完了する。 反応式11 式Iの化合物(ここで、AはA−2であり、G2はO、SまたはNHであり、 そしてX1はOである)は式XIの化合物と式XIIの化合物との縮合により製造でき る。一般的に有用な方法は例えばトリエチルアミンの如き酸スカベンジャーの存 在下での室温またはそれ以下における式XIIのアミン、アルコールまたはチオー ルを用いる式XIの酸塩化物の処理である。この反応は例えば塩化メチレン、テト ラヒドロフラン、トルエン、ベンゼンおよびクロロホルムの如き不活性溶媒の中 で実施できる。他の方法は例えばジシクロヘキシルカルボジイミドの如き縮合試 薬の存在下における例えば塩化メチレン、クロロホルム、テトラヒドロフラン、 トルエン、ジメチルホルムアミドおよび酢酸エチルの如き適当な溶媒中での式XI Iの化合物を用いる式XIの酸の処理である。温度は0−200℃の範囲であるこ とができ、好適な温度範囲は20−100℃である。この反応は例えばジメチル アミノピリジンの如き触媒の存在下で実施することができ(Synthesis(1972),4 53 参照)、そして30分間−48時間で完了する。 反応式12 式XIの化合物(ここで、L1はOHである)は反応式13に示されているよう に式VIIの化合物の加水分解により製造できる。典型的な反応は、式VIIの化合物 を例えば水、メタノールまたはエタノールの如き適当な溶媒中で例えば水酸化ナ トリウムまたは水酸化カリウムの如き塩基 で処理することを含む。反応温度は0℃から使用される特定溶媒の還流温度まで に変えることができる。エステル類の加水分解は March,Advanced Organic Che mistry,3rd 度は−70−200℃で変えることができる。ウィッティッヒ反応は Maercker により Org.Rxns.(1965),14,pp 270-490 に論じられている。この反応は1 −48時間で完了する。 反応式14 式XIIIの化合物は等モル量の式XIVの化合物とトリフェニルホスフィンの反応 により製造できる。この変換は反応式15に示されている。この反応は例えばベ ンゼン、トルエン、キシレン、エーテル、テトラヒドロフラン、ニトロメタン、 ニトロベンゼン、アセトニトリル、酢酸エチルおよびジメチルホルムアミドの如 き溶媒の中で実施することができる。反応温度は0−200℃に変えることがで きる。詳細に関しては、Maercker,Org.Rxns.(1965),14,pp 270-490 を参照 のこと。 反応式15 式XIVの化合物は反応式16に示されているようにXVの化合物から製造できる 。この反応は例えばアセトニトリルまたは塩化メチレンの如き適当な溶媒中での XVのアルコールと例えばトリフェニルホスフィンおよび四塩化炭素または四臭化 炭素、トリフェニルホスフィン、イミダゾールおよびヨウ素の如きハロゲン化剤 との組み合わせを含む。(Hooz et al.,Can.J.Chem.(1968),46,86; Lange et al.,Syn.Commun.(1990),20,1473)。反応温度は0−100℃に変える ことができそして 一般的には24時間以内に完了する。 反応式16 式XVの化合物はアルカリ金属水素化物を用いる式VIIのエステルの還元により 製造できる(反応式17)。反応条件は反応式4に関して記載されている通りで ある。 反応式17 式Iの化合物(ここで、AはA−6であり、G6はC2アルキレンであり、そし てQはアリールである)は反応式18に示されているように式XVIの化合物と最 初は塩化チオニルとのそして次にメタノール中の水酸化ナトリウムとの反応によ り製造できる。典型的な反応は式XVIの化合物と塩化チオニルとの未希釈のまた は例えばトルエンもしくは四塩化炭素の如き適当な溶媒を用いる混合を含む。混 合物を30−100℃に0.25−4時間加熱する。溶媒および過剰の塩化チオ ニルを真空下で濃縮しそして生じた粗製塩化物をメタノール中に溶解させそして 水性水酸化ナトリウムで処理する。この第二の反応を次に0.25−4時間にわ たり30−65℃に加熱する。冷却された反応混合物の有機溶媒を用いる抽出で 、AがA−6でありそしてG6がC2アルキレンである式Iの生成 物の単離が可能になる。 反応式18 式XVIの化合物は反応式19に示されているように式XVIIの化合物から製造で きる。式XVIIのアミノアルコールを最初に0−30℃における例えば塩化メチレ ン、クロロホルム、またはテトラヒドロフランの如き適当な溶媒中でのシアン化 トリメチルシリルとの反応によりシリルエーテルに転化させる。(式XVIIの化合 物中のアルコール基はシリルエーテルとして保護されるためそれは次の段階で塩 化ベンゾイルと反応しない。)15−60分後に、シリルエーテルの溶液をトリ エチルアミンの如き有機塩基でそして次に0−30℃において0.5−4時間に わたり適当な塩化ベンゾイルで処理する。水の添加および適当な有機溶媒を用い る抽出により粗製中間体アミドを単離することができる。シリルエーテル保護基 は0−30℃における5−60分間にわたる弗化テトラブチルアンモニウムの溶 液を用いる粗製中間体(テトラヒドロフランのような適当な溶媒中に溶解されて いる)の処理により除去される。水の添加および酢酸エチルのような有機溶媒を 用いる抽出により式XVIの化合物を次に単離する。 反応式19 式XVIIの化合物は反応式20に示されているように式XVIIIの化合物から製造 できる。典型的な反応は40−80℃における0.5−8時間にわたるメタノー ルまたはエタノールのような適当な溶媒中に溶解された式XVIIIのフタルイミド のヒドラジンを用いる処理を含む。 反応式20 式XVIIIの化合物は反応式21に示されているように式XIXの化合物から製造で きる。式XIXのエポキシドをテトラヒドロフランのような適当な溶媒中に溶解さ せそして15−35℃において0.5−4時間にわたり1当量のフタルイミドお よび触媒量(10モル%)のテトラキス−(トリフェニルホスフィン)パラジウム (0)で処理する。 反応式21 式XIXの化合物は反応式22に示されているように製造できる。式XXの化合物 を相転移条件下でヨウ化トリメチルスルホニウムで処理して式XIXのエポキシド を与える(Syn.Commun.(1987),17,503-513)。式XXの化合物は商業的に入手 できるかまたは当技術の専門家により容易に製造することができる。有用な製法 は J.Chem.Soc.Chem.Commun.(1984),1287に見られる。 反応式22 式Iの化合物(ここで、AはA−1であり、YはHであり、QはJである)は 反応式23に示されているように酢酸第二銅の存在下における式IIの化合物と式 XXIのトリアリールビスマスの反応によっても製造できる。この反応は例えば塩 化メチレンの如き溶媒中で室温において24時間以内に実施することができる( Barton et al.Tetrahedron Letts.(1987),28,pp 887-890)。 反応式23 式Iの化合物(ここで、R1およびR2はハロゲンでありそしてR3はアルキル またはアリールである)は式XXIIの化合物と塩基の反応により得られる。式XXII の化合物は置換されたベンズアミドを2−ハロアセトアルデヒドジアルキルアセ タールと反応させることにより製造される(EP−A−594,129参照)。X XIIからIへの転化は室温において5、6時間の期間にわたり行われる。反応式 24がこの変換を開示している。 反応式24 当技術の専門家は反応式7および12に記載されている変換を適当な商業的に 入手できるセリン誘導体を用いることによりAがA−3である式Iの化合物の製 造に拡大できることを認識するであろう。 式Iの化合物の製造に関して以上に記載した一部の試薬および反応条件は中間 体の中に存在するある種の官能基と相容性でないかもしれないことが認められる 。これらの場合には、合成中への保護/脱保護工程の加入が所望する生成物を得 るのを助けるであろう。保護基の使用および選択は化学合成の専門家には明白で あろう。 実施例1 段階A:2,6−ジフルオロベンゼンカルボキシイミド酸メチル 10.62g(0.068モル)の量の2,6−ジフルオロベンズアミドを10 .0g(0.068モル)のテトラフルオロホウ酸トリメチルオキソニウムの70 mLの塩化メチレン中懸濁液に加えた。反応を窒素下で一夜撹拌して行なった。 段階B:2−(2,6−ジフルオロフェニル)−4,5−ジヒドロ−4−オキサゾー ルカルボン酸メチル 9.09g(0.058モル)の量のセリンメチルエステル塩酸塩を一部分ずつ 10.0g(0.058モル)の段階Aの生成物の15mLのEtOHおよび5m LのH2O中の溶液に加えた。混合物を還流下で1.5時間撹拌した。揮発分を減 圧下で蒸発させそして残渣をEtOAc:ヘキサン(1:6)で溶離するシリカ モル)のKOHの100mLのMeOH中溶液を室温において 5時間撹拌した。MeOHを減圧下で蒸発させそして残渣をH2O中に溶解させ た。塩酸をこの溶液に加えて白色固休の沈澱を生成した。この固体を濾過しそし て空気乾燥して、19.6gの生成物を与えた:融点149−150℃;1H N MR(DMSO−d6)δ13.10(br,1H)、7.68(m,1H)、7.2 8(t,2H)、4.95(t,1H)、4.59(2t,2H)。 段階D:2−(2,6−ジフルオロフェニル)−4,5−ジヒドロ−N−(4−メト キシフェニル)−4−オキサゾールカルボキサミド 化合物1,3−ジシクロヘキシルカルボジイミド(0.433g、0.0021 モル)を段階Cの生成物(0.500g、0.0021モル)、4−ジメチルアミ ノピリジン(0.044g、0.00036モル)およびp−アニシジン(0.2 59g、0.0021モル)の3.5mLの塩化メチレン中混合物に加えた。この 混合物を還流下で1.5時間撹拌した。溶媒を蒸発させそして残渣をEtOAc :ヘキサン(1:4)で溶離するシリカゲルカラムを通して、0.881gの固 体を与えた:融点144−146.5℃。1H NMR(DMSO−d6)δ10. 17(s,1H)、7.65(m,1H)、7.58(d,2H)、7.28(t,2 H)、6.90(d,2H)、5.05(t,1H)、4.69(2t,2H)、3. 73(s,3H)。 実施例2 段階A:2−(2,6−ジフルオロフェニル)−4,5−ジヒドロ−4−オキサゾー ルカルボキサミド アンモニアのMeOH中2M溶液中の10.0g(0.041モル)の2−(2, 6−ジフルオロフェニル)−4,5−ジヒドロ−4−オキサゾー ルカルボン酸メチルの溶液を室温において一夜撹拌した。溶媒の蒸発で、9.0 4gの白色粉末を与えた:融点148−149℃;1H NMR(DMSO−d6 )δ7.67(m1H)、7.49(br s,1H)、7.39(br s,1H) 、7.27(t,2H)、4.85(t,1H)、4.56(2t,2H)。 N−[2−(2,6−ジフルオロフェニル)−4,5−ジヒドロ−4−オキサゾリル] −2−フルオロ−4−(トリフルオロメチル)ベンズアミド 塩化チオニル(2.008g、0.017モル)を1.10g(0.005モル) の2−フルオロ−4−トリフルオロメチル安息香酸の5mLのベンゼン中の冷却 された溶液に加えた。混合物を1時間にわたり還流させた。ベンゼンを蒸発させ 2.5g(0.01モル)のN−(1−メトキシ−2−クロロエチル)−2,6− ジフルオロベンズアミドおよび0.7g(0.011モル)のKOHペレット(8 5%)の30mLのメタノール中混合物を室温において一夜撹拌した。生成した 白色スラリーを濾過し、そして濾液を蒸発させて残存溶媒を除去した。残渣を水 /塩化メチレンで抽出した。塩化メチレンの抽出で、1.5g(70%収率)の 1種もしくはそれ以上の本発明の化合物を有効な量で農耕および/または非農 耕感染区域を含む有害生物の環境に、保護しようとする場所に、または抑制しよ うとする有害生物自体に適用することにより、節足動物性有害生物が抑制されそ して農耕、栽培および特殊作物、動物および人間の健康の保護が得られる。それ 故、本発明は1種もしくはそれ以上の式Iの化合物または少なくとも1種の該化 合物を含有する組成物を有効な量で農耕および/または非農耕感染区域を含む有 害生物の環境に、保護しようとする場所に、または抑制しようとする有害生物自 体に適用す ることを含んでなる、葉および土壌成育性の節足動物および線虫有害生物の抑制 並びに農耕および/または非農耕作物の保護方法も含んでなる。好適な適用方法 は噴霧によるものである。或いは、これらの化合物の顆粒状調合物を植物の葉ま たは土壌に適用することもできる。他の適用方法には、直接および残留噴霧、空 気噴霧、種子コート、マイクロカプセル化、全身的吸収、飼料、耳札、巨丸剤、 霧剤、薫蒸剤、アエロゾル、散剤などが包含される。これらの化合物は節足動物 が食べる飼料の中にまたは例えばトラップなどの装置の中に加えることができる 。 本発明の化合物はそれらの純粋な状態で適用できるが、たいていの適用法では 効果的な抑制のために必要な適用割合は、例えば抑制しようとする節足動物の 種類、有害生物の寿命サイクル、成長段階、その寸法、場所、季節、宿主作物ま たは動物、飼育性質、配偶行動、周囲水分、温度などの要素に依存するであろう 。通常の環境下では、1ヘクタール当たり約0.01〜2kgの活性成分の適用 割合が農耕生態系において有害生物を抑制するのに十分であるが、0.001k g/ヘクタールの如き少量でも十分であるかもしれずまたは8kgヘクタールの 如き大量を必要とするかもしれない。非農耕適用に関しては、有効な使用割合は 約1.0〜50mg/平方メートルであるが、0.1mg/平方メートルの如き 少量でも十分であるかもしれずまたは150mg/平方メートルの如き大量を必 要とするかもしれない。 下記の試験は個々の有害生物に対する本発明の化合物の抑制効果を示す。「抑
