Claims (1)
- 【特許請求の範囲】 1. 少なくとも1つの水溶性活性成分の制御された放出を生じる微細球の形 の薬剤組成物の製法において、 −適当量の水の中に活性成分を溶解する段階と、 −低分子量ポリラクチドから成る放出調節剤を追加的に含むdl−ラクチド− コ−グリコリド型の少なくとも1つのマトリックスコポリマーの塩素化炭化水素 中の溶液をもって、前記のようにして得られた活性成分の水溶液を乳化して、第 1超微細均質エマルジョンを得る段階と、 −界面活性剤を含む外部水相の中で前記の第1エマルジョンを乳化する段階と 、 −溶剤を抽出−蒸発させて微細球を生じ、この微細球を濾過、洗浄および乾燥 後に回収する段階とを含む方法。 2. 前記活性成分が下記物質:カルシトニン、バソプレシン、ソマトスタチ ン、成長ホルモン、プロラクチン、黄体刺激ホルモン、オキシトシン、セクレチ ン、サルブタモール、ゲンタミシン、テトラサイクリン、トブラマイシン、アミ カシン、サリチル酸ナトリウム、ジクロフェナック・ナトリウム、アモキシシリ ン、アンピシリン、ナプロキセン・ナトリウム、ビノレルビン、ビンクリスチン 、ビンデシン、メトトレキサート、シスプラチン、カルムスチン、ドキソルビシ ン、エピルビシン、ダウノルビシン、イフォスファミドおよびその誘導体から選 定される事を特徴とする請求項1に記載の方法。 3. 前記マトリックスコポリマーの中において、ラクチド/グリコリド比が 75:25乃至50:50の範囲内にある事を特徴とする請求項1または2のい ずれかに記載の方法。 4. マトリックスコポリマーの分子量が10,000乃至250,000、好ましくは12, 000乃至34,000の範囲内にある事を特徴とする請求項1乃至3のいずれかに記 載の方法。 5. 調節ポリラクチドの分子量が1000乃至10,000の範囲内、好ましくは 2000の区域にある事を特徴とする請求項1乃至4のいずれかに記載の方法。 6. マトリックスコポリマーと調節ポリラクチドとの比率が10:90乃至 90:10、好ましくは50:50乃至90:10の範囲内にある事を特徴とす る請求項1乃至5のいずれかに記載の方法。 7. 活性成分を保持するための任意物質またはエマルジョンを安定化する任 意試剤を添加する事なく、または粘度を増大するためのなんらかの操作を実施す る事なく、水溶性活性成分を水中に溶解して内部水相を形成する段階と、前記コ ポリマーと、ポリラクチドの低分子量ホモポリマーから成る前記放出調節剤と、 前記塩素化炭化水素とを含む有機相の中で前記内部水相を乳化して、第1(W/ O)エマルジョンを形成する段階と、界面活性剤を含有する第3水相に対して前 記第1エマルジョンを添加して第2(W/O/W)エマルジョンを形成する段階 と、溶剤を常温大気圧で急速に除去して微細球を形成する段階とを含む事を特徴 とする請求項1乃至6のいずれかに記載の方法。 8. 前記第1エマルジョンは超微細で均質である事を特徴とする請求項7に 記載の方法。 9. 第1エマルジョンの水相と有機相との比率が1:100乃至25:10 0の範囲内にある事を特徴とする請求項7に記載の方法。 10. 前記溶剤は、微細球の懸濁液が薄層を成して流れように成された特定 長さの傾斜から成る連続蒸発システムを使用して常温大気圧で急速除去される事 を特徴とする請求項7に記載の方法。 11. 前記塩素化炭化水素は塩化メチレンである事を特徴とする請求項1乃 至10のいずれかに記載の方法。 12. 前記界面活性剤はポリビニルピロリドンとポリオキシエチレンソルビ タン モノオレエートとの混合物である事を特徴とする請求項1乃至11のいず れかに記載の方法。 13. 請求項1乃至12のいずれかに記載の方法を実施する事によって得ら れた微細球。 14. 250μmまたはこれ以下、好ましくは125μmまたはこれ以下、 さらに好ましくは50μmまたはこれ以下の粒径を有する事を特徴とする請求項 13に記載の微細球。
