- 【特許請求の範囲】 1.試料中の分析物の存在又はレベルを検出する方法であって、下記の工程: (a)該試料を、分析物/第2リガンド複合体が形成されるように該分析物と 結合することのできる第2リガンドと混合し、 (b)工程(a)で生成した混合物を、第1リガンド/第2リガンド複合体が 形成されるように第2リガンドと結合し得る第1リガンドを結合して有する固相 と接触させ、該接触は、工程(a)で形成した該分析物/第2リガンド複合体の 解離が実質的に阻止される条件下で且つ十分制限された時間で行ない、 (c)検出可能な標識を、該標識部分が該第1リガンド/第2リガンド複合体 の形成に際して該固相上に保持されるように、工程(a)又は(b)の前又は後 の何れかにおいて、該第2リガンドに結合し、 (d)該標識部分を検出して該試料中の該分析物の存在又はレベルを測定する ことを含む、上記の方法。 2.接触の工程を、第1リガンド/第2リガンド複合体形成の時間に対するプロ ットが同じパラメーターの理想化されたプロットから実質的に分かれない条件下 で十分制限した時間で行ない、該理想化されたプロットは、工程(a)で形成さ れた該分析物/第2リガンド複合体の 解離が起きない反応を表す、請求項1に記載の方法。 3.接触の工程を、前記の混合物の任意の部分についての前記の固相との平均接 触時間が約10秒より短い条件下で且つ十分に制限された時間で行なう、請求項 1に記載の方法。 4.接触の工程を、前記の混合物の任意の部分についての前記の固相との平均接 触時間が約1秒より短い条件下で且つ十分に制限された時間で行なう、請求項3 に記載の方法。 5.前記の第1リガンドと前記の分析物とが同一である、請求項1に記載の方法 。 6.前記の第1リガンドと前記の分析物とが異なる、請求項1に記載の方法。 7.前記のリガンドが前記の分析物のアナログを含む、請求項6に記載の方法。 8.前記の分析物及び前記の第2リガンドが工程(a)中類似の濃度で存在する 、請求項1に記載の方法。 9.接触の工程が工程(a)で生成した前記の混合物を実質的に過剰の前記の第 1リガンドを結合して有する固相と接触させることを含む、請求項1に記載の方 法。 10.分析物/第2結合リガンド複合体の形成が第1結合リガンド/第2結合リ ガンド複合体の形成を阻止する、請求項1に記載の方法。 11.試料中の分析物の存在又はレベルを検出する方法であって、下記の工程: (a)該試料を: (i)該分析物と結合し得る第1リガンドを結合して有する固相、及び (ii)第1リガンド/分析物/第2リガンド複合体が該固相上に形成される ように該分析物又は第1リガンド/分析物複合体と結合し得る第2リガンド と接触させ、該接触は、該第2リガンドと該固相との間の任意の非特異的結合が 実質的に阻止される条件下で且つ十分制限された時間で行なわれ、 (b)検出可能な標識を、該第2リガンドに、該第1リガンド/分析物/第2 リガンド複合体の形成の際に該標識部分が該固相上に保持されるように、該第1 リガンド/分析物/第2リガンド複合体の形成の前又は後に結合させ、 (c)該標識を検出して、該試料中の該分析物の存在又はレベルを測定する ことを含む、上記の方法。 12.接触の工程が下記の工程: (a')前記の試料を前記の固相と、第1リガンド/分析物複合体が該固相上 に形成されるように接触させ、そして (a")該第1リガンド/分析物複合体を前記の第2リガンドと、前記の第1 リガンド/分析物/第2リガンド複合体が該固相上に形成されるように接触させ ることを含む、請求項11に記載の方法。 13.接触の工程が下記の工程: (a')前記の試料を第2リガンドと、分析物/第2リガンド複合体が形成さ れるように接触させ、そして (a")前記の固相を該分析物/第2リガンド複合体と、前記の第1リガンド /分析物/第2リガンド複合体が該固相上に形成されるように接触させる ことを含む、請求項11に記載の方法。 14.接触の工程が、前記の試料を前記の固相及び前記の第2リガンドと、前記 の第1リガンド/分析物/第2リガンド複合体が該固相上に形成されるように同 時に接触させることを含む、請求項11に記載の方法。 15.接触の工程が、前記の非特異的結合が平衡に到達せず且つ第1結合曲線が 安定に達しない条件下で十分に制限された時間で接触させることを含み、該結合 曲線において、非特異的な第2リガンド/固相複合体の形成が時間に対してプロ ットされる、請求項11に記載の方法。 16.接触の工程が、前記の第2リガンドの前記の分析物又は前記の第1リガン ド/分析物複合体への結合が平衡に到達せず且つ前記の第1リガンド/分析物/ 第2リガンド複合体の形成が時間に対してプロットされた第2結合曲線が安定に 達しない条件下で十分制限された時間で接触させることを含む、請求項15に記 載の方法。 17.接触の工程が、前記の第1結合曲線及び前記の第2結合曲線の両者がほぼ 直線的である条件下で十分制限 された時間で接触させることを含む、請求項16に記載の方法。 18.接触の工程が、前記の試料を、前記の分析物に比べて実質的に過剰の前記 の第1リガンドを結合して有する固相と接触させることを含む、請求項11に記 載の方法。 19.接触の工程が、前記の試料を、前記の第1リガンド/分析物複合体に比べ て実質的に過剰の前記の第2リガンドと接触させることを含む、請求項11に記 載の方法。 20.第1リガンド/第2リガンド複合体の固相上での形成を検出する方法であ って、下記の工程: (a)該第2リガンドと結合し得る分析物を、分析物/第2リガンド複合体が 形成されるように該第2リガンドと接触させ、 (b)該分析物/第2リガンド複合体を、該第2リガンドと結合し得る第1リ ガンドを結合して有する固相と、第1リガンド/第2リガンド複合体が形成され るように接触させ、該接触は、工程(a)で形成された該分析物/第2リガンド 複合体の解離が実質的に阻止される条件下で十分制限された時間で行ない、 (c)検出可能な標識を、該第2リガンドに、該標識部分が該第1リガンド/ 第2リガンド複合体の形成の際に該固相上に保持されるように、工程(a)又は (b)の前又は後に結合させ、 (d)該標識部分を検出し、それにより、該固相上の該第1リガンド/第2リ ガンド複合体の形成を検出することを含む、上記の方法。 21.前記の固相上の前記の第1リガンド/第2リガンド複合体のレベルを定量 する工程を更に含む、請求項20に記載の方法。 22.前記のレベルを、第2リガンドを固相との接触の前に前記の分析物と接触 させない場合に該固相上に形成される第1リガンド/第2リガンド複合体のレベ ルと比較することを更に含む、請求項21に記載の方法。
