た。 (a)α−アミラーゼの活性の測定:乾重量0.5gの
可溶性澱粉(ナカライテスク株式会社製)を約40ml
の蒸留水に懸濁し、沸騰浴中で溶解した後、50mlに
なるように蒸留水を用いて調整し、それを0.1M酢酸
緩衝液(pH4.5)50mlと混合して基質溶液とす
る。該基質溶液の0.3mlを試験管に採取して、40
℃で5分間インキュベートし、そこに同様に5分間プレ
インキュベートした試料酵素溶液(試料酵素1000m
gを蒸留水100mlに正確に溶解した酵素溶液)0.
2mlを添加して良く混和し40℃にてインキュベート
する。試料酵素溶液の添加から正確に20分経過後に、
ソモジ試薬[調製法は以下の(b)に示す]0.5ml

ト溶液中のマルトース濃度(Mo)(μmol/ml)
より、以下の数式により試料酵素液中の酵素活性を算

る還元糖を産生する酵素量を1unitとする。そし
て、下記の数式によって算出される試料酵素液の酵素
活性から、試料酵素液を調製する際の試料酵素の濃度に
