Claims (1)
- 【特許請求の範囲】 1.オンコスタチンM受容体のβ鎖(OSM−Rβ)のポリペプチドに共有結 合したgp130ポリペプチドを含む、オンコスタチンMと結合可能な受容体で あって、 上述のgp130ポリペプチドが以下の式a)−c): a) SEQ ID NO:1に提示されるヌクレオチド配列のコード領域を含むDNA; b) (a)のDNAに高強度条件下でハイブリダイズすることのできるDNA;およ び c) SEQ ID NO:2に提示されるアミノ酸配列をコードするDNA; からなるグループから選択されるDNAによってコードされる生物学的に活性な ポリペプチドであり、そして上述のOSM−Rβポリペプチドが以下のa')− c'): a') SEQ ID NO:5に提示されるヌクレオチド配列のコード領域を含むDNA; b') (a')のDNAに高強度条件下でハイブリダイズすることのできるDNA;お よび c') SEQ ID NO:6に提示するアミノ酸配列をコードするDNA; からなるグループから選択されるDNAによってコードされる生物学的に活性なポ リペプチドである、上述の受容体。 2.上述の受容体が可溶性のOSM-Rβポリペプチドに共有結合した可溶性のgp1 30ポリペプチドを含む、請求項1に記載の受容体。 3.上述の受容体がペプチドリンカーを介してOSM-Rβに共有結合したgp130を 含む、請求項1に記載の受容体。 4.上述の受容体が以下の式: R1-L-R2またはR2-L-R1 [式中、R1は可溶性のgp130を表しており;R2は可溶性のOSM-Rβを表しており ;そして、Lはペプチドリンカーを表している。] の組換え体融合タンパク質である、請求項3に記載の受容体。 5.上述した可溶性のgp130がSEQ ID NO:2のアミノ酸-22からy、または1からy [ここでyは308から597までのうちの整数を表す]を含み;上述した可溶性のOSM -RβがSEQ ID NO:6のアミノ酸-27からx、または1からx[ここでxは432から714ま でのうちの整数を表す]を含む、請求項4に記載の受容体。 6.請求項4の受容体をコードする単離されたDNA配列。 7.請求項6のDNA配列を含む組換え体発現ベクター。 8.上述のタンパク質の発現を促進する条件下で上述の融合タンパク質をコー ドするDNA配列を含む発現ベクターで形質転換した宿主細胞の培養し、そして上 述の融合タンパク質を回収することを含む、請求項4に記載の受容体を調製する 方法。 9.可溶性のgp130のC末端に結合した抗体のFc領域のポリペプチドを含む第一 の融合ポリペプチド、および可溶性のOSM-RβのC末端に結合した抗体のFc領域の ポリペプチドを含む第二の融合ポリペプチドからなり、上述の第一の融合タンパ ク質はFc領域のポリペプチドの間のジスルフィド結合を介して第二の融合ポリペ プチドに結合している、請求項2に記載の受容体。 10.上述した第一および第二の融合ポリペプチドの発現を促進する条件下で、 上述の第一の融合ポリペプチドをコードしている第一の発現ベクター、および第 二の融合ポリペプチドをコードしている第二の発現ベクターを共にトランスフェ クションした宿主細胞を培養し、そして上述の受容体を回収することを含む、請 求項9に記載の受容体を調製する方法。 11.上述の可溶性のgp130がSEQ ID NO:2のアミノ酸-22からy、または1からy [ここでyは308から597までのうちの整数を表す]を含み;可溶性のOSM-RβがSE Q ID NO:6のアミノ酸-27からx、または1からx[ここでxは432から714までのうち の整数を表す]を含む、請求項9に記載の受容体。 12.請求項2の受容体、および適当な希釈液または担体を含む組成物。 13.上述のDNAが以下のもの: a) SEQ ID NO:5に提示されるヌクレオチド配列のコード領域を含むDNA; b) 高強度条件下で(a)のDNAにハイブリダイズすることができ、生物学的に 活性なOSM-RβポリペプチドをコードするDNA;および c) SEQ ID NO:6に提示されるアミノ酸配列をコードするDNA からなるグループから選択される、OSM-Rβポリペプチドをコードしている単離 されたDNA。 14.上述のOSM-RβがSEQ ID NO:6のアミノ酸-27から952、およびアミノ酸1か ら952からなるグループから選択されるアミノ酸配列を含む、請求項13に記載の 単離されたDNA。 15.上述のDNAが可溶性のOSM-Rβポリペプチドをコードする、請求項13に記載 の単離されたDNA。 16.上述の可溶性のOSM-RβポリペプチドがSEQ ID NO:6のアミノ酸-27からx、 または1からx[ここでxは432から714までのうちの整数である]を含む、請求項1 5に記載のDNA。 17.OSM-RβポリペプチドがN末端のアミノ酸配列 Glu Arg Leu Pro Leu Thr P ro Val Ser Leu Lys Valによって特徴付けられる、OSM-Rβポリペプチドをコー ドする単離されたDNA。 18.請求項13に記載のDNAを含む発現ベクター。 19.請求項15に記載のDNAを含む発現ベクター。 20.OSM-Rβの発現を促進する条件下において請求項18に記載のベクターで形 質転換した宿主細胞を培養し、そしてその培養液からOSM-Rβポリペプチドを回 収することを含む、OSM-Rβポリペプチドを調製する方法。 21.OSM-Rβの発現を促進する条件下において請求項19に記載のベクターで形 質転換した宿主細胞を培養し、そしてその培養液からOSM-Rβポリペプチドを回 収することを含む、OSM-Rβポリペプチドを調製する方法。 22.請求項13に記載のDNAによってコードされる精製されたOSM-Rβポリペプチ ド。 23.上述のOSM-Rβが可溶性のOSM-Rβポリペプチドである、請求項22に記載の OSM-Rβポリペプチド。 24.SEQ ID NO:6のアミノ酸-27からx、または1からx[ここでxは432から714ま でのうちの整数である]を含む、請求項23に記載の可溶性のOSM-Rβ。 25.上述のOSM-RβポリペプチドがN末端のアミノ酸配列 Glu Arg Leu Pro Leu Thr Pro Val Ser Leu Lys Valによって特徴付けられる、精製されたOSM-Rβポ リペプチド。 26.SEQ ID NO:6のアミノ酸1から952を含む、請求項25に記載のOSM-Rβポリペ プチド。 27.上述のOSM-Rβが細胞株ATCC 69675に含まれる組換え体ベクター内のOSM- Rβ cDNAによってコードされる、請求項22に記載のOSM-Rβポリペプチド。 28.上述のポリペプチドが上述のOSM-RβポリペプチドのC末端と融合させたFc ポリペプチドをさらに含む、請求項15に記載の可溶性のOSM-Rβポリペプチド。 29.請求項13に記載のOSM-Rβポリペプチドと免疫反応する抗体。 30.上述の抗体がモノクローナル抗体である、請求項29に記載の抗体。 31.SEQ ID NO:5に存在するDNA配列のコード領域の少なくとも約14ヌクレオチ ドの配列を含む単離された核酸分子、またはその相補的なDNAまたはRNA。
