レチノイン酸受容体エプシロン
本発明は、新たに同定されたポリヌクレオチド、そのようなポリヌクレオチド
によりコードされるポリペプチド、そのようなポリヌクレオチドおよびポリペプ
チドの使用、さらにはまた、そのようなポリヌクレオチドおよびポリペプチドの
製造に関する。とりわけ、本発明のポリペプチドは、レチノイン酸受容体エプシ
ロン(RARε)である。本発明はまた、そのようなポリペプチドの作用を阻害す
ることにも関する。
レチノイド(ビタミンA誘導体)は、多数の組織の正常な成長、外観(vision)、
維持、生殖(reproduction)および全体の生存に重要である(Wolbach,S.B.、J .Exp.Med.
、42:753−777(1925))。加えて、ビタミンA欠乏(
VAD)雌親の子孫は、多くの発生欠陥を示し、胚形成の間、レチノイドもまた
重要であることを示す(Wilson,J.G.ら、Am.J.Anat.、92:189−21
7(1953))。レチノイン酸(RA)投与により、胎児並びに若年および成年
の動物におけるビタミンA欠乏の作用を阻止する、および/または逆転させるこ
とができる(Wilson,J.G.ら、Am.J.Anat.、92:189−217(19
53))。哺乳動物の胚に対する母性RA投与の劇的な催奇作用並びに脊椎動物
の胚発生および両生動物におけるリム再生に対するレチノイドの局所投与の目覚
ましい作用は、RAが実際はモルフォゲン(発生の間、位置情報を与える)であり
得、また器官形成の間、重要な役割もまた果し得るという考えに著しく寄与して
いる(Tabin,C.J.、Cell、66:199−217(1991))。
レチノイドはまた、多数の組織の正常な成長、外観、維持、生殖および全体の
生存にも重要である(Wolbach,S.B.およびHowe,P.R.、J.Exp.Med.、4
2:753−777(1925))。
RAシグナルの作用は、リガンド誘導性転写調節因子のスーパーファミリーに
属する受容体の2つのファミリーによって媒介されると考えられ、これらには、
ステロイド甲状腺ホルモンおよびビタミンD3受容体が含まれる(Evans,R.M.
、Science、240:889−895(1988))。
レチノイン酸受容体遺伝子は、ステロイドホルモン受容体ファミリーとして知
られている遺伝子のスーパーファミリーに属する。このファミリーにおける遺伝
子は全て、別々の領域またはドメインに分けることができ、これらは時に、領域
A/B、C、D、E、およびFと呼ばれる。C領域はDNA結合ドメインをコー
ドし、E領域はリガンド結合ドメインをコードし、またF領域はカルボキシ末端
ドメインをコードする。D領域は「ヒンジ」として機能すると考えられる。A/
B(またはN末端)領域の機能は完全に明らかとなっているわけではないが、受容
体転写活性の向上および抑制と関係し得る(Hollenbergら、Cell、55:89
9−906(1988))。
RA受容体(RAR)ファミリー(RARα、βおよびγ並びにそれらのアイソ
フォーム)は、オールトランス(all-trans)およびノンシス(non-cis)の両方のR
Aにより活性化される。RARδもまたクローン化されている(Mech,Dev.、4
0:99−112(1993))。ある一定の種内では、3つのRAR型のDN
A結合領域およびリガンド結合領域ドメインは非常に似ているが、C末端領域お
よび中間領域は、類似性を全く、またはほとんど示さない。3つのRAR型のア
ミノ酸配列もまた、それらのB領域において顕著に異なっており、またそれらの
主要なアイソフォーム(α1およびα2、β1〜β4、γ1およびγ2、並びに
δ1およびδ2)はさらに、それらのN末端A領域において異なる(Leid,M.ら
、Trends Biochem.Sci.、17:427−433(1992))。
RARγ変異体マウスは、精嚢および前立腺の扁平上皮化生が原因となって、
成長障害、初期の致死、および雄の生殖不能(sterility)を示す(Lohnes,D.ら
、Cell、73:643−658(1993))。
RARγ転写物は、それらの胚由来に関係なく、前軟骨性圧縮物中に見い出さ
れ、軟骨への制限、および扁平角質化上皮の分化が続いて起こる(Dolle,P.ら
、Nature、342:702−705(1989))。これらの観察結果は、形態
形成および軟骨形成におけるRARγの役割を示唆する(Dolleら、Developmen t
、
100:1133−1151(1990))。
本発明のDNA配列およびそのようなDNA配列によりコードされるポリペプ
チドは、レチノイン酸受容体ファミリーに属し、またレチノイン酸受容体ガンマ
に対して最も相同性である。モジュレーターおよびDNA結合ドメインは両方と
も、ショウジョウバエのエクジソン受容体の対応する部分で最も高い相同性を共
有する。発生の間、エクジソン受容体が重要な役割を果す。
本発明の一態様により、RARεである新規成熟ポリペプチド、さらにはまた
、そのフラグメント、アナログおよび誘導体を提供する。本発明のポリペプチド
は、ヒト起源である。
本発明の別の態様により、そのようなポリペプチドをコードするポリヌクレオ
チド(DNAまたはRNA)を提供する。
本発明のまたさらなる態様により、そのようなポリペプチドを組換え技術によ
り製造する方法を提供する。
本発明のまたさらなる態様により、そのようなポリペプチド、またはそのよう
なポリペプチドをコードするポリヌクレオチドを治療目的、例えば、その新規ホ
ルモン、DNA結合部位、遺伝子標的および組織特異性を同定するのに利用する
方法を提供し、これは、発生、分化、組織再生、生殖、並びに免疫および造血系
の刺激を促進するための治療標的を提供する。
本発明のまたさらなる態様により、そのようなポリペプチドに対する抗体を提
供する。
本発明のこれらの態様および他の態様は、本明細書中の教示から当業者に明ら
かであろう。

本発明の一態様により、第1図の推定アミノ酸配列を有する成熟ポリペプチド
、または1994年3月18日にATCC寄託番号第75754号として寄託さ
れたクローンのcDNAによりコードされる成熟ポリペプチドをコードする、単
離された核酸(ポリヌクレオチド)を提供する。
本発明のポリペプチドをコードするポリヌクレオチドは、精巣、脾臓および胸
腺から得ることができる。本発明のポリヌクレオチドは、成人の肺から得られた
cDNAライブラリー中で発見された。それは、構造的には、レチノイン酸受容
体ファミリーに関係する。それは、約433アミノ酸残基のタンパク質をコード
する開放読み枠(オープンリーディングフレーム)を含む。そのタンパク質は、レ
チノイン酸受容体ガンマに対し、モジュレーター領域の70アミノ酸長さ以上で
48%の同一性および58%の類似性をもって、最も高い相同性の度合を示す。
同様に、DNA結合領域およびリガンド結合領域もまた、レチノイン酸受容体ガ
ンマに対して非常に相同性である(第5図)。
本発明のポリヌクレオチドは、RNAの形で、またはDNAの形であり得、こ
のDNAには、cDNA、ゲノムDNA、および合成DNAが含まれる。該DN
Aは、二本鎖または一本鎖であり得、また一本鎖であるなら、コード鎖または非
コード(アンチ−センス)鎖であり得る。成熟ポリペプチドをコードするコード配
列は、第1図に示すコード配列または寄託されたクローンのコード配列と同じで
あってよく、あるいは遺伝コードの重複または縮重の結果として、第1図のDN
Aまたは寄託されたcDNAと同じ成熟ポリペプチドをコードする異なったコー
ド配列であってもよい。
第1図の成熟ポリペプチドまたは寄託されたcDNAによりコードされる成熟
ポリペプチドをコードするポリヌクレオチドには、以下のものが含まれ得る:成
熟ポリペプチドのコード配列のみ;成熟ポリペプチドのコード配列、およびリー
ダーもしくは分泌配列といったような付加的コード配列;成熟ポリペプチドのコ
ード配列(また場合により、付加的コード配列)、および成熟ポリペプチドのコー
ド配列のイントロンまたは非コード配列5'および/または3'といったような非
コード配列。
従って、「ポリペプチドをコードするポリヌクレオチド」という用語は、ポリ
ペプチドのコード配列のみが含まれるポリヌクレオチド、さらにはまた、付加的
コードおよび/または非コード配列が含まれるポリヌクレオチドを包含する。
本発明はさらに、第1図の推定アミノ酸配列を有するポリペプチドまたは寄託
されたクローンのcDNAによりコードされるポリペプチドのフラグメント、ア
ナログおよび誘導体をコードする、上記のポリヌクレオチドの変異体に関する。
ポリヌクレオチドの変異体は、ポリヌクレオチドの天然に存在するアレル変異体
またはポリヌクレオチドの天然には存在しない変異体であり得る。
従って、本発明には、第1図に示すのと同じ成熟ポリペプチドまたは寄託され
たクローンのcDNAによりコードされる同じ成熟ポリペプチドをコードするポ
リヌクレオチド、さらにはまた、そのようなポリヌクレオチドの変異体が含まれ
、これらの変異体は、第1図のポリペプチドまたは寄託されたクローンのcDN
Aによりコードされるポリペプチドのフラグメント、誘導体またはアナログをコ
ードする。そのようなヌクレオチド変異体には、欠失変異体、置換変異体並びに
付加および挿入変異体が含まれる
先に示したように、該ポリヌクレオチドは、第1図に示すコード配列の天然に
存在するアレル変異体または寄託されたクローンのコード配列の天然に存在する
アレル変異体であるコード配列を有し得る。当業界で知られているように、アレ
ル変異体は、1つまたはそれ以上のヌクレオチドの置換、欠失または付加を有し
得る別の形のポリヌクレオチド配列であり、これは、コードされるポリペプチド
の機能を実質的には変えない。
本発明のポリヌクレオチドはまた、本発明のポリペプチドの精製を可能とする
マーカー配列に枠内で融合したコード配列も有し得る。細胞宿主の場合には、そ
のマーカー配列は、マーカーに融合した成熟ポリペプチドの精製を提供するため
の、pQE−9(Qiagen)ベクターにより与えられるヘキサ−ヒスチジンタグ(tag
)であってよく、または、例えば、哺乳動物宿主、例えば、COS−7細胞を使
用する場合には、そのマーカー配列は、赤血球凝集素(HA)タグであってよい。
HAタグは、インフルエンザ赤血球凝集素タンパク質から得られるエピトープに
対応する(Wilson,I.ら、Cell、37:767(1984))。
本発明はさらに、配列間に少なくとも50%、また好ましくは70%の同一性
がある場合に、上記の配列にハイブリダイズするポリヌクレオチドに関する。本
発明は特に、ストリンジェント条件下、上記のポリヌクレオチドにハイブリダイ
ズするポリヌクレオチドに関する。本明細書中で使用する場合、「ストリンジェ
ント条件」という用語は、配列間に少なくとも95%、また好ましくは少なくと
も97%の同一性がある場合にのみ、ハイブリダイゼーションが起こるであろう
ことを意味する。好ましい態様では、上記のポリヌクレオチドにハイブリダイズ
するポリヌクレオチドは、第1図のcDNAまたは寄託されたcDNAによりコー
ドされる成熟ポリペプチドと同じ生物学的機能または活性を実質的には保有する
ポリペプチドをコードする。
本明細書中で言う寄託は、特許手続上の微生物の寄託の国際承認に関するブダ
ペスト条約の条件の下に保持されるであろう。これらの寄託は、単に当業者への
便宜として与えられるものであって、寄託が35 U.S.C.§112下に要求
されることを承認するものではない。寄託された物質中に含まれるポリヌクレオ
チドの配列、さらにはまた、それによりコードされるポリペプチドのアミノ酸配
列は、本明細書の一部を構成して、本明細書中の配列のいずれかの記載と矛盾す
る際はいつでも照合している。寄託された物質を製造し、使用し、または販売す
るには、実施許諾が要求され得、またそのような実施許諾は、ここでは付与され
ない。
本発明はさらに、第1図の推定アミノ酸配列を有する、または寄託されたcD
NAによりコードされるアミノ酸配列を有するRARεポリペプチド、さらには
また、そのようなポリペプチドのフラグメント、アナログおよび誘導体に関する
。
第1図のポリペプチドまたは寄託されたcDNAによりコードされるポリペプ
チドを示す場合、「フラグメント」、「誘導体」および「アナログ」という用語
は、そのようなポリペプチドと実質的に同じ生物学的機能または活性を保有する
ポリペプチドを意味する。従って、アナログには、プロプロテイン部分を切断す
ることにより活性化して、活性な成熟ポリペプチドを製造することができるプロ
プロテインが含まれる。
本発明のポリペプチドは、組換えポリペプチド、天然ポリペプチドまたは合成
ポリペプチド、好ましくは組換ポリペプチドであってよい。
第1図のポリペプチドまたは寄託されたcDNAによりコードされるポリペプ
チドのフラグメント、誘導体またはアナログは、(i)1つまたはそれ以上のア
ミノ酸残基が同型または非同型アミノ酸残基(好ましくは、同型アミノ酸残基)で
置換されており、またそのような置換アミノ酸残基が遺伝コードによりコードさ
れるものであってもよく、またはコードされたものでなくてもよいもの、(ii)
1つまたはそれ以上のアミノ酸残基に置換基が含まれるもの、または(iii)成
熟ポリペプチドが、該ポリペプチドの半減期を増加させる化合物(例えば、ポリ
エチレングリコール)のような、他の化合物と融合しているもの、または(iv)
リーダーもしくは分泌配列、または成熟ポリペプチドの精製に使用される配列、
またはプロプロテイン配列といったような、付加的アミノ酸が成熟ポリペプチド
に融合しているものであり得る。そのようなフラグメント、誘導体およびアナロ
グは、本明細書中の教示から当業者の範囲内であると思われる。
本発明のポリペプチドおよびポリヌクレオチドは、単離された形で提供される
のが好ましく、好ましくは、均一となるまで精製される。
「単離された」という用語は、物質がその元の環境(例えば、それが天然に存
在するなら、天然の環境)から除去されていることを意味する。例えば、生きて
いる動物にある天然に存在するポリヌクレオチドまたはポリペプチドは単離され
ていないが、天然の系における共存物質のいくつかまたは全てから分離された同
じポリヌクレオチドまたはポリペプチドは単離されている。そのようなポリヌク
レオチドはベクターの部分となり得、および/またはそのようなポリヌクレオチ
ドまたはポリペプチドは組成物の部分となり得、またそのようなベクターまたは
組成物はその天然の環境の部分ではないという点で、なお単離されている。
本発明はまた、本発明のポリヌクレオチドが含まれるベクター、本発明のベク
ターで遺伝的に操作された宿主細胞、および本発明のポリペプチドの組換え技術
による製造にも関する。
宿主細胞は、例えば、クローニングベクターまたは発現ベクターであり得る本
発明のベクターで遺伝的に操作される(トランスデュースされ、またはトランス
フォームされ、またはトランスフェクトされる)。該ベクターは、例えば、プラ
スミド、ウイルス粒子、ファージ等の形であり得る。操作された宿主細胞は、プ
ロモーターを活性化し、トランスフォーマントを選択し、またはRARε遺伝子
を増幅するのに適するよう改変された従来の栄養培地で培養することができる。
温度、pH等といったような培養条件は、発現用に選択された宿主細胞で先に使
用した培養条件であって、当業者に明らかであろう。
本発明のポリヌクレオチドは、ペプチドを組換え技術により製造するのに使用
することができる。従って、例えば、該ポリヌクレオチドを、ポリペプチドを発
現するための様々な発現ベクターのいずれか1つに含ませることができる。その
ようなベクターには、染色体、非染色体および合成DNA配列、例えば、SV4
0の誘導体;細菌プラスミド;ファージDNA;バキュロウイルス;酵母プラス
ミド;プラスミドとファージDNAとの組合せから得られるベクター;ワクシニ
ア、アデノウイルス、鶏痘ウイルス、仮性狂犬病といったようなウイルスDNA
が含まれる。しかし、他のいずれのベクターも、それが宿主中で複製可能であっ
て、生存可能である限り、使用することができる。
適当なDNA配列を様々な方法によりベクターに挿入することができる。一般
には、DNA配列を当業界で既知の方法により適当な制限エンドヌクレアーゼ部
位に挿入する。そのような方法および他の方法は、当業者の範囲内であると思わ
れる。
発現ベクターのDNA配列を、適当な発現制御配列(プロモーター)に作動可能
に結合し、mRNA合成を行わせる。そのようなプロモーターの代表例として、
以下のものが挙げられる:LTRまたはSV40プロモーター、E.coli. lacま
たはtrp、ファージラムダPLプロモーター、および原核もしくは真核細胞または
それらのウイルスでの遺伝子の発現を制御することが知られている他のプロモー
ター。発現ベクターはまた、翻訳開始のためのリボソーム結合部位および転写終
結区も含む。該ベクターはまた、発現を増幅するのに適当な配列も含み得る。
さらに、発現ベクターは、真核細胞培養の場合にはジヒドロフォレートレダク
ターゼまたはネオマイシン耐性といったような、またはE.coliではテトラサイ
クリンまたはアンピシリン耐性といったような、トランスフォームされた宿主細
胞の選択のための表現型特性を与えるために、1つまたはそれ以上の選択可能な
マーカー遺伝子を含むのが好ましい。
上記のような適当なDNA配列、さらにはまた、適当なプロモーターまたは制
御配列を含むベクターを、適当な宿主をトランスフォームするために使用して、
その宿主がタンパク質を発現するのを可能にすることができる。
適当な宿主の代表例として、以下のものが挙げられる:E.coli、Streptomyc es
、Salmonella typhimuriumといったような細菌細胞;酵母のような真菌細胞
;DrosophilaおよびSf9といったような昆虫細胞;CHO、COSまたはBow
esメラノーマといったような動物細胞;植物細胞等。適当な宿主の選択は、本明
細書中の教示から当業者の範囲内であると思われる。
とりわけ、本発明にはまた、先に広く記載した配列を1つまたはそれ以上含ん
でなる組換え構築物も含まれる。その構築物は、本発明の配列が順または逆方向
で挿入されている、プラスミドまたはウイルスベクターといったようなベクター
を含んでなる。この実施態様の好ましい態様では、該構築物はさらに、例えば、
該配列に作動可能に結合したプロモーターを含め、制御配列を含んでなる。適当
なベクターおよびプロモーターが多数、当業者に知られていて、市販されている
。以下のベクターを例として挙ける。細菌用:pQE70、pQE60、pQE−
9(Qiagen)、pbs、pD10、ファージスクリプト(phagescript)、psiX174
、pbluescript SK、pbsks、pNH8A、pNH16a、pNH18A、pNH46

ratagene);ptrc99a、pKK223−3、pKK233−3、pDR54、pRI
T5(Pharmacia)、pRSおよびpGEM。真核生物用:pWLNEO、pSV2C
AT、pOG44、pXT1、pSG(Stratagene)、pSVK3、pBPV、pMS
G、pSVL(Pharmacia)。しかし、他のいずれのプラスミドまたはベクターも
、それらが宿主中で複製可能であって、生存可能である限り、使用することがで
きる。
プロモーター領域は、CAT(クロラムフェニコールトランスフェラーゼ)ベク
ターまたは選択マーカーを有する他のベクターを用いて、いずれかの所望の遺伝
子から選択することができる。2つの適当なベクターは、PKK232−8およ
びPCM7である。個々に名付けられた細菌プロモーターには、lac I、lacZ、
T3、T7、gpt、ラムダPR、PLおよびrpが含まれる。真核プロモーターには
、CMV即時初期(immediate early)、HSVチミジンキナーゼ、初期および後
期SV40、レトロウイルス由来のLTR、およびマウスのメタロチオネイン−
Iが含まれる。適当なベクターおよびプロモーターの選択は、十分、当業者のレ
ベルの範囲内である。
さらなる態様では、本発明は、上記の構築物を含む宿主細胞に関する。その宿
主細胞は、哺乳動物細胞のような高等真核細胞、酵母細胞のような低等真核細胞
であり得、または該宿主細胞は、細菌細胞のような原核細胞であり得る。構築物
の宿主細胞への導入は、リン酸カルシウムトランスフェクション、DEAE−デ
キストラン媒介トランスフェクションまたはエレクトロポレーションにより行う
ことができる。(Davis,L.、Dibner,M.、Battey,L.、Basic Methods in
Molecular Biology(1986))。
宿主細胞中の構築物を通常の方法で使用して、組換え配列によりコードされる
遺伝子産物を製造することができる。あるいはまた、本発明のポリペプチドは、
従来のペプチド合成装置により合成的に製造することができる。
成熟タンパク質は、適当なプロモーターの制御下、哺乳動物細胞、酵母、細菌
、または他の細胞中で発現させることができる。そのようなタンパク質を、本発
明のDNA構築物から得られるRNAを用いて製造するために、無細胞翻訳系も
ま
た使用することができる。原核および真核宿主で使用するのに適当なクローニン
グおよび発現ベクターは、Sambrookら、Molecular Cloning:A Laboratory
Manual,第2版、Cold Spring Harbor、ニューヨーク、(1989)により
記載されており、この開示は、本明細書の一部を構成する。
本発明のポリペプチドをコードするDNAの高等真核生物による転写は、エン
ハンサー配列をベクターに挿入することにより増加する。エンハンサーは、プロ
モーターに作用してその転写を増加させる、通常、約10〜300bpの、DNA
のシス作用性要素である。例には、bp100〜270の、複製開始点の後期側に
あるSV40エンハンサー、サイトメガロウイルス初期プロモーターエンハンサ
ー、複製開始点の後期側にあるポリオーマエンハンサー、およびアデノウイルス
エンハンサーが含まれる。
通例、組換え発現ベクターには、複製開始点、および宿主細胞のトランスフォ
ーメーションを可能にする選択可能なマーカー、例えば、E.coliのアンピシリ
ン耐性遺伝子およびS.cerevisiae TRP1遺伝子、並びに下流の構造配列の転
写を行わせるために高度に発現される遺伝子から得られるプロモーターが含まれ
るであろう。そのようなプロモーターは、とりわけ、3−ホスホグリセリン酸キ
ナーゼ(PGK)のような解糖系酵素、α因子、酸性ホスファターゼ、または熱シ
ョックタンパク質をコードするオペロンから得ることができる。ヘテロロガス構
造配列は、翻訳開始および終結配列、また好ましくは、翻訳されたタンパク質の
細胞周辺腔または細胞外媒体への分泌を行わせることができるリーダー配列と共
に、適当な相(phase)で構築される。場合により、そのヘテロロガス配列は、所
望の特性、例えば、発現された組換え生成物の安定化または精製の簡易化を与え
るN−末端同定ペプチドが含まれる融合タンパク質をコードすることができる。
細菌で使用するのに有用な発現ベクターは、所望のタンパク質をコードする構
造DNA配列を、適当な翻訳開始および終結シグナルと共に、機能的なプロモー
ターを有する作動可能なリーディング相に挿入することにより構築される。その
ベクターは、ベクターの維持を確実なものとするために、また所望により、宿主
内での増幅を与えるために、1つまたはそれ以上の表現型の選択可能なマーカー
および複製開始点を含んでなるであろう。トランスフォーメーションに適当な原
核宿主には、E.coli、Bacillus subtilis、Salmonella typhimurium、並びに
Pseudomonas属、Streptomyces属、およびStaphylococcus属の範囲内の様々な
種が含まれるが、他のものもまた、選択物質として使用することができる。
代表的であるが、非限定的な例として、細菌で使用するのに有用なベクターは
、周知のクローニングベクター pBR322(ATCC 37017)の遺伝要素
を含んでなる市販のプラスミドから得られる、選択可能なマーカーおよび細菌の
複製開始点を含んでなり得る。そのような市販のベクターには、例えば、pKK
223−3(Pharmacia Fine Chemicals、Uppsala、スウェーデン)およびG
EM1(Promega Biotec、Madison、WI、米国)が含まれる。これらのpBR
322「骨核」部分を適当なプロモーターおよび発現されるべき構造配列と組み合
わせる。
適当な宿主株をトランスフォーメーションして、その宿主株を適当な細胞密度
までの増殖させた後、選択されたプロモーターを適当な方法(例えば、温度シフ

組換えタンパク質を発現させるために、様々な哺乳動物細胞培養系もまた使用
することができる。哺乳動物発現系の例には、Gluzman、Cell、23:175
(1981)により記載されている、サルの腎臓線維芽細胞のCOS−7系、お
よび適合可能なベクターを発現させることができる他の細胞系、例えば、C12
7、3T3、CHO、HeLaおよびBHK細胞系が含まれる。哺乳動物発現ベク
ターは、複製開始点、適当なプロモーターおよびエンハンサー、またいずれかの
必要なリボソーム結合部位、ポリアデニル化部位、スプライスドナーおよびアク

RARεポリペプチドは、硫酸アンモニウムまたはエタノール沈降、酸抽出、

とができる。組換え製造方法で使用する宿主により、本発明のポリペプチドは、
グリコシル化され得るか、またはグルコシル化され得ない。本発明のポリペプチ
ドにはまた、最初のメチオニンアミノ酸残基が含まれ得る。
RARεホルモン受容体は、その新規ホルモン、DNA結合部位、遺伝子標的

できる;しかし、2,000bpより大きいクローンは、簡単な検出に十分なシグ

2,000bpが良好であり、4,000はより良好であって、4,000以上は、

冒された個体と冒されていない個体との比較は、通例、染色体スプレッドから
見ることができるか、またはそのcDNA配列に基づいたPCRを利用して検出
することができる、欠失またはトランスロケーションといったような、染色体に
おける構造変化を先ず探す。最後には、変異の存在を確認するために、また変異
を多型性から区別するために、幾つかの個体由来の遺伝子の完全な配列決定が必
要とされる。
ポリペプチド、それらのフラグメントもしくは他の誘導体、もしくはそれらの
アナログ、またはそれらを発現する細胞を免疫源として使用して、それらに対す

以下の実施例の理解を容易にするために、幾つかの頻繁に出てくる方法および

「プラスミド」は、前置きする小文字のpおよび/または続けて大文字および
/または数字により示す。本明細書中の出発プラスミドは、市販されていて、限

DNAの「消化」は、DNAのある配列にのみ作用する制限酵素でDNAを触

「オリゴヌクレオチド」は、化学的に合成することができる、一本鎖ポリデオ
キシヌクレオチド、または2つの相補的ポリヌクレオチド鎖を示す。そのような
合成オリゴヌクレオチドは5'ホスフェートを有さないことから、キナーゼの存
在下、ATPでホスフェートを加えることなしには、別のオリゴヌクレオチドに
ライゲートしないであろう。合成オリゴヌクレオチドは、脱リン酸化されていな

実施例 1
インビトロにおける転写および翻訳によるRARεの発現
TNT Coupled Reticulocyte Lysate System(Promega、2800 Wood
s Hollow Road、Madison、WI 53771−5399)を用いて、RARε
のインビトロにおける転写および翻訳を行った。RARεをコードするcDNA
、ATCC第75754号をEcoRI〜XhoI方向にクローン化した。EcoRI
部位は遺伝子の5'末端を規定し、またXhoI部位は遺伝子の3'末端を規定する
。遺伝子をT3の方向で挿入した。T3は、特異的プロモーターを認識して、D
NAをmRNAに転写する、バクテリオファージRNAポリメラーゼを定義する
。ウサギの網状赤血球ライゼートにT3 RNAポリメラーゼを補い、メッセー
ジを転写するためにT3 RNAポリメラーゼを、また新生RNAを翻訳するた
めに網状赤血球ライゼートを利用して、T3プロモーターによりタンパク質の発
現を行わせる。RARε DNAを含む環状(または直鎖状)プラスミド1μgをT
NTTMライゼートに直接加えて、反応容積50μl中、T3 RNAポリメラーゼ
および[35S]−メチオニンと共に、30℃で1時間インキュベートした。インキ
ュベーション後、翻訳生成物を10% SDS−PAGEゲル電気泳動により分
離した。そのゲルを10% 酢酸、15% メタノール中に30分間固定した後、
Bio−Radゲル乾燥装置で1時間乾燥した。Kodak XARフィルムを用いて、
オートラジオグラフィーを行った。そのフィルムを増感紙に−80℃でさらした
。主要な翻訳生成物は、49Kdに見ることができる(第4図)。
実施例 2
バキュロウイルス−昆虫細胞システムにおけるRARεの発現および精製
10% FBSを補ったグレイスの昆虫培地(Grace's Insect medium)(Gibc
o)を用いての培養において、Sf9昆虫細胞を27℃で維持する。バキュロウイ
ルス伝達プラスミドpA2はGentzらにより構築された(未公開結果)(第5図)。
PCRを利用して、BamHI部位が5'末端に、またAsp 718部位が3'末端
に含まれる完全なヒトRARε cDNAを増幅した。ヒトRARεの増幅に使用
したPCRオリゴヌクレオチドプライマーは:
である。ヒトRARε cDNAを含む増幅されたフラグメント、およびプラスミ
ドpA2をBamHIおよびAsp 718により消化して、Genecleanキットにより
精製した。ヒトRARε cDNAを含む増幅されたフラグメントをベクターpA
2中に挿入して、組換え伝達ベクターpA2RARεを生成した。制限エンドヌ
クレアーゼ、PCRおよびDNAシークエンシングを利用して、その構築物の正

Laboratory Manual(1992)、W.H.Freeman and Company]により記載
された。プラークは、青色−白色選択を利用することにより、組換えバキュロウ
イルスに関してスクリーニングされるであろう。目視によりスクリーニングされ
たプラークは、PCRスクリーニングにより分析して確認しなくてはならない。
組換えウイルスが得られたら、Sf9細胞(2×106細胞/ml)を、組換えRAR
ウイルス(108プラーク形成単位/ml)を用いて、室温で2時間感染させること
ができる。トランスフェクション後、そのウイルス播種材料(inoculum)を新たな
培地で置き換える。上清および細胞を60−72時間後に収集する。次いで、S
DS−PAGEにより、細胞外および細胞内画分をRARεの発現に関して分析
する。受容体は、連続陰イオン交換、ゲル濾過およびDNAアフィニティークロ
マトグラフィーにより、部分的に精製することができる。
実施例 3
ヒト組織におけるRARεの発現パターン
ノーザンブロット分析を行って、ヒト組織におけるRARεの発現レベルを調
べた。全体の細胞RNA試料をRNAzolTM B システム(Biotecx Laboratori
es,Inc.6023 South Loop East、Houston、TX 77033)で単離し
た。指定された各々のヒト組織から単離された総RNA約10μgを1% アガロ
ースゲルで分離して、ナイロンフィルター上にブロットした(Sambrook、Frits
ch、およびManiatis、Molecular Cloning、Cold Spring Harbor Press、
(1989))。Stratagene Prime−It キットにより、標識化反応をDNA

ime−3 Prime,Inc.5603 Arapahoe Road、Boulder、CO 80303

This technique can be used with cDNAs as short as 500 or 600 bases; however, clones larger than 2,000 bp are more likely to bind to unique chromosomal locations with sufficient signal intensity for easy detection. FISH requires the use of clones from which ESTs are obtained, and the longer the better. For example, 2,000 bp is better, 4,000 is better, and 4,000 or more is probably not necessary to get good results at a reasonable time rate.
