Claims (1)
- 【特許請求の範囲】 1.テンプレートとして作用して、複数の異なる組織を含む機能的置換骨格連結 をインビボで形成する、哺乳動物における移植のためのデバイスであって、ここ で少なくとも1つの該複数の異なる組織が非無機質化組織であり、該デバイスが 以下を含む、デバイス: a) 浸潤細胞の付着を可能にする単一の構造を規定し、そして骨格連結の複数 の異なる組織(該組織の少なくとも1つが非無機質化組織である)に対して特異 性を有する残留物または骨格連結の複数の異なる組織(該組織の少なくとも1つ が非無機質化組織である)由来の残留物を含み;そして 置換しようとする骨格連結の大きさおよび形状を模倣する大きさおよび形状を 有し、そして該マトリックスの表面上に配置される、 生体適合性、生分解性マトリックス、 b) 置換しようとする骨格連結を模倣する大きさおよび形状を有する新たな該 複数の異なる組織(該新たな組織のうち少なくとも1つが非無機質化組織である )の形成を誘導し、それゆえ機能的置換骨格連結の再生を可能にするに十分な量 の骨形成タンパク質。 2.前記マトリックスが哺乳動物ドナー由来の失活化組織を含む、請求項1に記 載のデバイス。 3.前記マトリックスが関節軟骨の残留物および骨の残留物を含む、請求項1に 記載のデバイス。 4.前記マトリックスが脱水した哺乳動物組織を含む、請求項1に記載のデバイ ス。 5.前記置換非無機質化組織が、関節軟骨、靭帯、腱、滑液包、および滑液膜組 織からなる群より選択される、請求項1に記載のデバイス。 6.前記骨格連結が滑膜性連結または関節連結を規定する、請求項1に記載のデ バイス。 7.前記デバイスが失活化した無傷の骨格連結構造を規定する、請求項1に記載 のデバイス。 8.骨格連結内に関節軟骨置換組織をインビボで形成する、哺乳動物における移 植のためのデバイスであって、該デバイスが以下を含む、デバイス: 生体適合性、生体吸収性マトリックスの表面上に配置される骨形成タンパク質で あって、該マトリックスが、浸潤細胞の付着を可能にし、そして関節軟骨組織に 特異的な残留物または関節軟骨組織に由来する残留物を含む構造を規定する、骨 形成タンパク質。 9.骨格連結内に置換非無機質化組織をインビボで形成する、哺乳動物における 移植のためのデバイスであって、該デバイスが以下を含む、デバイス: 生体適合性、生体吸収性マトリックスの表面上に配置される骨形成タンパク質で あって、該マトリックスが浸潤細胞の付着を可能にし、そして非無機質化骨格連 結組織(置換しようとする該組織に本質的に対応する)に特異的な残留物または 非無機質化骨格連結組織(置換しようとする該組織に本質的に対応する)由来の 残留物を含む構造を規定する、骨形成タンパク質。 10.前記非無機質化組織が無血管組織である、請求項9に記載のデバイス。 11.前記非無機質化組織が関節軟骨、靭帯、滑液膜、および滑液包組織からな る群より選択される、請求項8に記載のデバイス。 12.前記マトリックスが失活化同種組織または失活化異種組織を含む、請求項 8または9に記載のデバイス。 13.前記マトリックスが、コラーゲンと、乳酸、グリコール酸、酪酸およびこ れらの組み合わせのモノマーを含むポリマーと、ヒドロキシアパタイトと、リン 酸三カルシウムと、それらの混合物とからなる群より選択される材料を含む、請 求項8または9に記載のデバイス。 14.微粒子物質に結合して成型可能な固体を形成するに適した材料をさらに含 む、請求項8または9に記載のデバイス。 15.前記骨形成タンパク質がOP-1、OP-2、BMP2、BMP3、BMP4、BMP5、BMP6、OP X、またはそれらの機能的等価物のホモダイマーまたはヘテロダイマーを含む、 請求項1、8、または9に記載のデバイス。 16.哺乳動物において、複数の異なる組織(ここで、該複数の異なる組織の少 なくとも1つが非無機質化組織である)を含む自家移植置換骨格連結の形成を誘 導する方法であって、該方法が以下の工程を包含する、方法: a) 生体吸収性、生体適合性マトリックスの表面上に配置される骨形成タンパ ク質を含むデバイスを提供する工程であって、該マトリックスが 浸潤細胞の付着を可能にし、そして骨格連結の該複数の異なる組織(該組織の 少なくとも1つが非無機質化組織である)に対して特異性を有する残留物または 、骨格連結の該複数の異なる組織(該組織の少なくとも1つが非無機質化組織で ある)に由来する残留物を含む単一構造を規定し、そして 置換しようとする骨格連結の大きさおよび形状を模倣する大きさおよび形状を 有する(該新しい組織の少なくとも1つが非無機質化組織である)、工程;およ び b) 哺乳動物の部位に該デバイスを移植し、それにより機能性骨格連結の形成 を誘導する工程。 17.前記哺乳動物の前記部位が置換しようとする内因性身体部分を規定する、 請求項16に記載の方法。 18.前記マトリックスが、形状および構造的関係において、置換しようとする 前記複数の異なる組織に対応するような大きさである残留物をさらに含む、請求 項16に記載の方法。 19.前記複数の異なる組織が骨および軟骨を含む、請求項16に記載の方法。 20.前記マトリックスが失活化同種組織または失活化異種組織を含む、請求項 16に記載の方法。 21.前記複数の異なる組織の1つが無血管組織である、請求項16に記載の方 法。 22.骨表面上の関節軟骨をインビボで修復する方法であって、該方法が以下の 工程を包含する、方法: 哺乳動物の部位で該骨表面上に、生体適合性、生体吸収性マトリックスの表面 上に配置される骨形成タンパク質を含むデバイスを提供する工程であって、該マ トリックスが、軟骨組織に特異的な残留物または軟骨組織由来の残留物を含み、 そして浸潤細胞の付着を可能にする構造を規定する、工程。 23.前記部位が滑膜腔に生じる、請求項22に記載の方法。 24.哺乳動物において骨格連結内の非無機質化組織を回復する方法であって、 該方法が以下の工程を包含する、方法: 哺乳動物の該骨格連結に、生体適合性、生体吸収性マトリックスの表面上に配 置される骨形成タンパク質を含むデバイスを提供する工程であって、該マトリッ クスが、組織(置換しようとする該非無機質化組織に本質的に対応する)に特異 的な残留物または組織(置換しようとする該非無機質化組織に本質的に対応する )由来の残留物を含み、そして浸潤細胞の付着を可能にする構造を規定する、工 程。 25.回復させようとする前記非無機質化組織が無血管組織を含む、請求項24 に記載の方法。 26.回復させようとする前記非無機質化組織が、関節軟骨、腱、靭帯、滑液包 、および滑液膜組織からなる群より選択される、請求項24に記載の方法。 27.前記マトリックスが同種関節軟骨または異種関節軟骨由来である、請求項 22または24に記載の方法。 28.前記デバイスが成型可能な固体を含む、請求項22または24に記載の方 法。 29.前記デバイスが可撓性シートを含む、請求項22または24に記載の方法 。 30.前記デバイスが、コラーゲンと、乳酸、酪酸、グリコール酸およびそれら の混合物を含むポリマーと、ヒドロキシアパタイトと、それらの組み合わせとを 含む、請求項22または24に記載の方法。 31.前記骨形成タンパク質がOP-1、OP-2、BMP2、BMP3、BMP4、BMP5、BMP6、OP Xのホモダイマーまたはヘテロダイマー、あるいはそれらの機能的等価物を含む 、請求項16、22、または24に記載の方法。
