Claims (1)
- 【特許請求の範囲】 1.BMP−10蛋白をコードしている単離DNA配列。 2.該DNAが、 (a)配列番号:1のヌクレオチド779または797から1102まで; (b)配列番号:10のヌクレオチド1108または1126から1431ま で;および (c)緊縮条件下で(a)または(b)にハイブリダイズし、軟骨および/ま たは骨を形成する能力を示す蛋白をコードしている配列 からなる群より選択される請求項1記載のDNA配列。 3.該DNAが、 (a)配列番号:2のアミノ酸1から108までをコードしているヌクレオチ ド; (b)配列番号:11のアミノ酸1から108までをコードしているヌクレオ チド;および (c)緊縮条件下で(a)または(b)にハイブリダイズし、軟骨および/ま たは骨を形成する能力を示す蛋白をコードしている配列 からなる群より選択される請求項1記載のDNA配列。 4.請求項1のDNA配列で形質転換された宿主細胞。 5.請求項2のDNA配列で形質転換された宿主細胞。 6.請求項3のDNA配列で形質転換された宿主細胞。 7.軟骨および/または骨の形成を誘導する能力により特徴づけられる蛋白を コードしている配列を有する単離DNA分子であって、 (a)配列番号:1のヌクレオチド779から1105まで; (b)配列番号:10のヌクレオチト1108から1431まで;および (c)(a)または(b)に対する天然に存在する対立遺伝子配列および等価 な縮重コドン配列 からなる群より選択されるDNA配列からなるDNA分子。 8.請求項7記載のDNA分子で形質転換された宿主細胞。 9.作動するように発現調節配列に連結された請求項7記載のDNA分子から なるベクター。 10.請求項9記載のベクターで形質転換された宿主細胞。 11.BMP−10蛋白をコードしている単離DNA分子であって、配列番号 :10のヌクレオチド160から1431までからなるDNA分子。 12.BMP−10蛋白をコードしているDNA暗号配列からなる単離DNA 分子であって、配列番号:10のヌクレオチド1108から1431までからな り、さらに、適当な5'末端側のプロペプチドをコードしておりDNA暗号配列 の読み取り枠に連結しているヌクレオチド配列からなるDNA分子。 13.作動するように発現調節配列に連結された請求項11記載のDNA分子 からなるベクター。 14.請求項12のベクターで形質転換された宿主細胞。 15.以下の工程(a)、(b): (a)BMP−10蛋白をコードしているヌクレオチド配列からなる請求項2 記載のDNA配列で形質転換された宿主細胞を培養し;次いで (b)培地からBMP−10蛋白を回収し、精製する からなる精製BMP−10蛋白の製造方法。 16.以下の工程(a)、(b): (a)BMP−10蛋白をコードしているヌクレオチド配列からなる請求項3 記載のDNA配列で形質転換された宿主細胞を培養し;次いで、 (b)培地からBMP−10蛋白を回収し、精製する からなる精製BMP−10蛋白の製造方法。 17.以下の工程(a)、(b): (a)BMP−10蛋白をコードしているヌクレオチド配列からなる請求項7 記載のDNA配列で形質転換された宿主細胞を培養し;次いで、 (b)培地からBMP−10蛋白を回収し、精製する からなる精製BMP−10蛋白の製造方法。 18.配列番号:11に示すアミノ酸1から108までのアミノ酸配列からな る精製BMP−10ポリペプチド 19.ポリペプチドが二量体であって、各サブユニットが少なくとも配列番号 :11のアミノ酸1から108までのアミノ酸配列からなるものである請求項1 8記載の精製BMP−10ポリペプチド。 20.1のサブユニットが少なくとも配列番号:11のアミノ酸1から108 までのアミノ酸配列からなり、もう1つのサブユニットがBMP−1、BMP− 2、BMP−3、BMP−4、BMP−5、BMP−6、BMP−7、BMP− 8およびBMP−9からなる群より選択される請求項18記載の精製BMP−1 0ポリペプチド。 21.以下の工程(a)、(b): (a)配列番号:10に示すヌクレオチド1108から1431までのヌクレ オチド配列からなるDNA分子で形質転換された細胞を培養し;次いで、 (b)配列番号:11に示すアミノ酸1から108までのアミノ酸配列からな る蛋白を該培地から回収し精製する により製造される精製BMP−10蛋白。 22.細胞が哺乳動物細胞で、DNAがさらに配列番号:10のヌクレオチド 160から1107までからなる、請求項21記載の精製BMP−10蛋白。 23.軟骨および/または骨の形成を誘導する能力により特徴づけられる精製 BMP−10蛋白。 24.医薬上許容される担体と混合された有効量の請求項23記載のBMP− 10蛋白からなる医薬組成物。 25.さらに、該組成物を支持し、骨および/または軟骨の成長のための表面 を提供するマトリックスからなる請求項24記載の組成物。 26.該マトリックスが、ヒドロキシアパタイト、コラーゲン、ポリ乳酸およ びリン酸三石灰からなる群より選択される材料からなる請求項25記載の組成物 。 27.骨および/または軟骨の形成を必要とする患者に有効量の請求項25記 載の組成物を投与することを特徴とする、患者における骨および/または軟骨の 形成を誘導する方法。 28.蛋白のTGF−βスーパーファミリー(superfamily)のメンバー由来 のプロペプチドをコードしているDNA配列からなり、BMP−10ポリペプチ ドをコードしているDNA配列の正しい読み取り枠に連結しているキメラDNA 分子。
