Claims (1)
- 【特許請求の範囲】 1.中枢神経系腫瘍細胞を選択的に死滅させる方法であって、 (a)該腫瘍細胞をシトクロムP450遺伝子を含むウイルスベクターに感染さ せ、その際、その遺伝子産物の発現は該腫瘍細胞を該腫瘍細胞の細胞周期に依存 することなく化学療法剤に感受性にさせ、 (b)該腫瘍細胞を該化学療法剤と接触させ、ついで (c)該腫瘍細胞を選択的に死滅させる ことを特徴とする方法。 2.該シトクロム遺伝子が、P450 2B1、P450 2B6、P450 2A6、P450 2C6、P450 2C8、P450 2C9、P450 2C 11、またはP450 3A4である請求項1に記載の方法。 3.該シトクロム遺伝子が、P450 2B1である請求項2に記載の方法。 4.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項1 に記載の方法。 5.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項2 に記載の方法。 6.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項3 に記載の方法。 7.該中枢神経系腫瘍細胞が、星状細胞腫、寡突起膠腫、髄膜腫、神経線維腫 、神経膠芽腫、上衣腫、神経鞘腫、および神経線維肉腫である請求項1に記載の 方法。 8.該中枢神経系腫瘍細胞が、星状細胞腫、寡突起膠腫、髄膜腫、神経線維腫 、神経膠芽腫、上衣腫、神経鞘腫、および神経線維肉腫である請求項5に記載の 方法。 9.該ウイルスベクターがレトロウイルスである請求項2に記載の方法。 10.該ウイルスベクターがレトロウイルスである請求項5に記載の方法。 11.悪性腫瘍の治療における化学療法剤の効能を増大させる方法であって、 (a)該悪性腫瘍をシトクロムP450遺伝子を含むウイルスベクターに感染さ せ、 (b)該腫瘍を、シトクロムP450遺伝子産物にさらされると細胞毒性の代謝 産物に活性化される化学療法剤と接触させ、ついで (c)該化学療法剤の治療学的有効性を改善にするに充分高いレベルの細胞毒性 代謝産物を該腫瘍自体の中に生成させる ことを特徴とする方法。 12.該シトクロム遺伝子が、P450 2B1、P450 2B6、P450 2A6、P450 2C6、P450 2C8、P450 2C9、P450 2 C11、またはP450 3A4である請求項11に記載の方法。 13.該シトクロム遺伝子が、P450 2B1である請求項12に記載の方 法。 14.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 11に記載の方法。 15.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 12に記載の方法。 16.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 13に記載の方法。 17.該悪性腫瘍が中枢神経系に位置する請求項11に記載の方法。 18.該悪性腫瘍が中枢神経系に位置する請求項16に記載の方法。 19.該ウイルスベクターがレトロウイルスである請求項11に記載の方法。 20.中枢神経系腫瘍細胞を選択的に死滅させる方法であって、 (a)該腫瘍細胞をシトクロムP450 2B1遺伝子を含むウイルスベクター に感染させ、その際、その遺伝子産物の発現は該腫瘍細胞を該腫瘍細胞の細胞周 期に依存することなくシクロホスファミドに感受性にさせ、 (b)該腫瘍細胞を該シクロホスファミドと接触させ、ついで (c)該腫瘍細胞を選択的に死滅させる ことを特徴とする方法。 21.悪性腫瘍の治療における化学療法剤の効能を増大させる方法であって、 (a)該悪性腫瘍をシトクロムP450 2B1遺伝子を含むウイルスベクター に感染させ、 (b)該腫瘍を、シトクロムP450 2B1遺伝子産物にさらされると細胞毒 性の代謝産物に活性化されるシクロホスファミドと接触させ、ついで (c)該シクロホスファミドの治療学的有効性を改善にするに充分高いレベルの 細胞毒性代謝産物を該腫瘍自体の中に生成させる ことを特徴とする方法。 22.末梢腫瘍細胞を選択的に死滅させる方法であって、 (a)該腫瘍細胞をシトクロムP450遺伝子を含むウイルスベクターに感染さ せ、その際、その遺伝子産物の発現は該腫瘍細胞を該腫瘍細胞の細胞周期に依存 することなく化学療法剤に感受性にさせ、 (b)該腫瘍細胞を該化学療法剤と接触させ、ついで (c)該腫瘍細胞を選択的に死滅させる ことを特徴とする方法。 23.該シトクロム遺伝子が、P450 2B1、P450 2B6、P450 2A6、P450 2C6、P450 2C8、P450 2C9、P450 2 C11、またはP450 3A4である請求項22に記載の方法。 24.該シトクロム遺伝子が、P450 2B1である請求項23に記載の方 法。 25.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 22に記載の方法。 26.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 23に記載の方法。 27.該化学療法剤がシクロホスファミドまたはイホスファミドである請求項 24に記載の方法。 28.該化学療法剤がシクロホスファミドである請求項27に記載の方法。 29.該ウイルスベクターがレトロウイルスである請求項23に記載の方法。 30.該ウイルスベクターがレトロウイルスである請求項26に記載の方法。 31.該末梢腫瘍細胞が乳腫瘍細胞である請求項22に記載の方法。 32.該末梢腫瘍細胞が乳腫瘍細胞である請求項23に記載の方法。 33.該末梢腫瘍細胞が乳腫瘍細胞である請求項24に記載の方法。 34.該末梢腫瘍細胞が乳腫瘍細胞である請求項25に記載の方法。 35.該末梢腫瘍細胞が乳腫瘍細胞である請求項26に記載の方法。 36.該末梢腫瘍細胞が乳腫瘍細胞である請求項27に記載の方法。 37.該末梢腫瘍細胞が乳腫瘍細胞である請求項28に記載の方法。
