- 【特許請求の範囲】 1.植物における可逆的な雄性不稔を作出するための方法であり、 (a)(i)植物において発現した時に花粉の形成または機能を抑制する遺伝子産 物をコードするDNA配列と、 (ii)該DNA配列の発現を制御するオペレーターと、 (iii)花粉の形成または機能に不可欠な細胞に特異的であり、かつ遺伝子 産物をコードするDNA配列と機能的に連結しているプロモーターとを含む第1の組 換えDNA分子による遺伝的な形質転換が可能である該花粉産生性の植物ゲノムへ の導入、 (b)該DNA配列の発現の結果として雄性不稔が実現される条件下における花粉 産生性の植物の育成、ならびに (c)雄性捻性である雑種植物を作成するため、DNA結合タンパク質をコードす るDNA配列、および該タンパク質が雄性不稔植物の組換えDNAのオペレーターと結 合することができるDNA配列の発現を制御するプロモーターを含む第2の組換えDN A配列分子がゲノムに組み込まれた雄性捻性の系統に由来する花粉と該雄性不稔 の植物との交雑、を含む方法。 2.第1の組換え分子の遺伝子産物がサイトトキシンである、請求項1記載の方 法。 3.第1の組換え分子のプロモーターが葯特異的なプロモーターである、請求 項1記載の方法。 4.葯特異的なプロモーターがプロモーター5126のヌクレオチド配列を含む、 請求項3記載の方法。 5.オペレーターがlexAオペレーターである、請求項1記載の方法。 6.第1の組換え分子または第2の組換えDNA分子がさらに選択マーカー遺伝子 を含む、請求項1記載の方法。 7.DNA結合タンパク質がlexAタンパク質である、請求項1記載の方法。 8.第2の組換えDNA分子のプロモーターが花粉の形成または機能に不可欠な細 胞に特異的なプロモーターである、請求項1記載の方法。 9.第2の組換えDNA分子のプロモーターが葯特異的なプロモーターである、請 求項8記載の方法。 10.葯特異的なプロモーターがプロモーター5126のヌクレオチド配列を含む、 請求項9記載の方法。 11.第2の組換えDNA分子のプロモーターが誘導性のプロモーターである、請求 項1記載の方法。 12.第2の組換えDNA分子のプロモーターが構成的プロモーターである、請求項 1記載の方法。 13.構成的プロモーターがトウモロコシ・ユビキチンプロモーターである、請 求項12記載の方法。 14.構築物がPHP6522である、請求項13記載の方法。 15.構築物がPHP6555である、請求項10記載の方法。 16.プロモーターがDP5055のSca-NcoI断片のヌクレオチド配列を含む、請求項 4記載の方法。 17.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に少なくとも503塩基にわたるヌクレオチド配列を含む、請求項4記載の 方法。 18.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-503位から-1位までのヌクレオチド配列を含む、請求項4記載の方法 。 19.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-587位から-1位までのヌクレオチド配列を含む、請求項4記載の方法 。 20.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-890位から-1位までのヌクレオチド配列を含む、請求項4記載の方法 。 21.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-503位から-134位までのヌクレオチド配列を含む、請求項4記載の方 法。 22.プロモーターがDP5055のSca-NcoI断片のヌクレオチド配列を含む、請求項 10記載の方法。 23.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に少なくとも503塩基にわたるヌクレオチド配列を含む、請求項10記載 の方 法。 24.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-503位から-1位までのヌクレオチド配列を含む、請求項10記載の方法 。 25.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-587位から-1位までのヌクレオチド配列を含む、請求項10記載の方法 。 26.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-890位から-1位までのヌクレオチド配列を含む、請求項10記載の方法 。 27.プロモーターが図1の第1488ヌクレオチドにある開始コドンから相対的に みて上流に-503位から-134位までのヌクレオチド配列を含む、請求項10記載の方 法。 28.サイトトキシンがDAMメチラーゼである、請求項2記載の方法。 29.(i)花粉の形成または機能を抑制する遺伝子産物をコードするDNA配列、 および(ii)花粉の形成または機能に不可欠な細胞に特異的で、該遺伝子産物を コードするDNA配列と機能的に連結しているプロモーター、を含む組換えDNA分子 を含む形質転換された植物の産生、ならびに 該DNA配列の発現の結果としての雄性不稔が実現される条件における植物の育 成、の段階を含む雄性不稔植物の作出方法。 30.遺伝子産物がサイトトキシンである、請求項29記載の方法。 31.プロモーターが葯特異的なプロモーターである、請求項29記載の方法。 32.葯特異的なプロモーターが5126プロモーターである、請求項31記載の方法 。 33.DNA配列が外因性メチラーゼ遺伝子をコードする、請求項29記載の方法。 34.外因性メチラーゼ遺伝子がDAMメチラーゼ遺伝子である、請求項33記載の 方法。 35.プロモーターが葯特異的なプロモーターである、請求項34記載の方法。 36.葯特異的なプロモーターが5126プロモーターである、請求項35記載の方法 。 37.請求項29記載の方法により作出される植物。 38.請求項1記載の方法により作出される植物。
