- 【特許請求の範囲】 1.実質的に、(a)グリセリン: (b)細胞生存度を高めるのに十分な量のアルカリ金属塩化物; (c)環境条件下で細胞維持を支持するのに有効な量の生理学的に代謝し得る単 糖類; 及び (d)プラズマ膜(plasma membrane)構造完全性を維持するた めに有効な量の血漿アルブミン からなる濃厚凍結保存溶液。 2.更に希釈剤を含有している請求項1記載の凍結保存溶液。 3.溶液のpHが約7と7.5の間である請求項1記載の凍結保存溶液。 4.濃厚液が実質的に(a)約60ないし80重量%のグリセリン、(b)約5ない し約10重量%のアルカリ金属塩化物、(c)約0.2ないし約1重量%の単糖類 ;及び(d)約20ないし30重量%の血漿アルブミンからなる請求項1記載の凍 結保存溶液。 5.溶液が実質的に(a)約20ないし40重量%のグリセリン、(b)約1.5な いし約5重量%のアルカリ金属塩化物、(c)約0.1ないし約0.5重量%の単 糖類;(d)約6ないし15重量%の血漿アルブミン及び(e)残部希釈剤(bala nce diluent)からなる請求項2記載の凍結保存溶液。 6.アルカリ金属塩化物が塩化ナトリウムである請求項1記載の濃厚凍結保存 溶液。 7.アルカリ金属塩化物が塩化ナトリウムである請求項2記載の凍結保存溶液 。 8.単糖類がグルコースである請求項1記載の濃厚凍結保存溶液。 9.単糖類がグルコースである請求項2記載の凍結保存溶液。 10.血漿アルブミンがヒト血漿アルブミンである請求項1記載の濃厚凍結保 存溶液。 11.血漿アルブミンがヒト血漿アルブミンである請求項2記載の凍結保存溶 液。 12.実質的に、 (a)約60ないし80重量%のグリセリン; (b)約5ないし約10重量%のアルカリ金属塩化物; (c)約0.2ないし約1重量%のグルコース;及び (d)約20ないし30重量%の血漿アルブミン からなる濃厚凍結保存溶液。 13.実質的に、 (a)約20ないし40重量%のグリセリン; (b)約1.5ないし約5重量%のアルカリ金属塩化物; (c)約0.1ないし約0.5重量%のグルコース; (d)約6ないし15重量%の血漿アルブミン;及び (e)残部の希釈剤(balance of diluent) からなる凍結保存溶液。 14.(i)請求項2に記載の凍結保存溶液に標的細胞を導入し、そして (ii)保護された標的細胞を凍結保存することからなる細胞を凍結保存する方 法。 15.標的細胞を含む溶液の、凍結保存溶液に対する体積比が約1:1ないし 約1:10でありそして標的細胞の、凍結保存溶液に対する比率が約1×105 細胞/mlないし約1×109細胞/mlである請求項14記載の方法。 16.標的細胞が造血幹細胞又は前駆細胞である請求項14記載の方法。 17.標的細胞が血液の血小板である請求項14記載の方法。 18.標的細胞がCD34+ 細胞であると同定されている細胞である請求項14 記載の方法。 19.標的細胞が臍帯の血液から採取される請求項14記載の方法。 20.(i)請求項13に記載の凍結保存溶液に標的細胞を導入し、そして (ii)保護された標的細胞を凍結保存することからなる細胞を凍結保存する方 法。 21.標的細胞を含む溶液の、凍結保存溶液に対する体積比が約1:1ないし 約1:10でありそして標的細胞の、凍結保存溶液に対する比率が約1×105 細胞/ mlないし約1×109細胞/mlである請求項20記載の方法。 22.保護された標的細胞が分当り1−2℃の速度で約−70と約−196℃ の間の温度まで冷却される請求項20記載の方法。 23.(i)請求項2記載の凍結保存溶液に標的細胞を導入し、(ii)保護さ れた標的細胞を凍結保存し、そして(iii)保護された標的細胞を解凍すること からなる生存細胞を凍結保存しそして回収するための方法。 24.標的細胞を含む溶液の、凍結保存溶液に対する体積比が約1:1ないし 1:10の間であり標的細胞に対する凍結保存溶液の比が約1×105細胞/m lないし約1×109細胞/mlの間にある請求項23記載の方法。 25.標的細胞が造血幹細胞又は前駆細胞である請求項23記載の方法。 26.(i)標的細胞を請求項2記載の凍結保存溶液に導入し、(ii)保護さ れた標的細胞を凍結保存し、 (iii)保護された標的細胞を解凍し、そして(iv)解凍した保護された標的細 胞を、標的細胞と凍結保存溶液の分離をしないで、その標的細胞を必要としてい る対象に導入することからなる細胞の凍結保存、回収と医療的使用のための方法 。 27.標的細胞を含む溶液の、凍結保存溶液に対する体積比が約1:1ないし 1:10の間であり標的細胞に 対する凍結保存溶液の比が約1×105細胞/mlないし約1×109細胞/ml の間にある請求項26記載の方法。 28.標的細胞が造血幹細胞又は前駆細胞である請求項26記載の方法。 29.標的細胞が血液の血小板である請求項26記載の方法。 30.標的細胞がCD34+ 細胞であると同定されている細胞である請求項26 記載の方法。 31.標的細胞が臍帯の血液から得られる請求項26記載の方法。 32.対象がヒトである請求項26記載の方法。
