Description
Claims (1)
- 【特許請求の範囲】 1.(a)被検体を含むと思われる試料を提供し; (b)該試料を、もし該被検体が存在するならばプローブ−被検体複合体を形成 する多機能性プローブと接触させ、該プローブは結合ドメインおよび成長促進活 性ドメインの少なくとも両方を有し、該成長促進活性ドメインは増殖性細胞の成 長を促進することができ; (c)該プローブ−被検体複合体がバックグラウンド上で検出できるように、十 分量の非結合多機能性プローブを除去し; (d)調節物質および成長が工程(b)の複合体の成長促進活性により増強され る増殖性細胞の両方を含む反応マトリックスと接触させることにより細菌連鎖反 応系を形成し; (e)反応マトリックス内で成長促進環境を形成し、そして該増殖性細胞の成長 を可能とするために十分な時間、工程(d)の反応系をインキューベーションし 、そして (f)バックグラウンド上に新たな該増殖性細胞の存在を観察する、工程を特徴 とする試料中の被検体の存在を測定する方法。 2.工程(c)の除去が、上記系から非結合の多機能性プローブを洗浄すること により行われることを特徴とする、請求の範囲第1項に記載の方法。 3.工程(c)の除去が、非結合の多機能性プローブを不活性化するが物理的に は存在するように残すことにより行われることを特徴とする、請求の範囲第1項 に記載の方法。 4.上記成長促進活性が酵素的であることを特徴とする、請求の範囲第1項に記 載の方法。 5.調節物質が抗生物質であることを特徴とする、請求の範囲第1項に記載の方 法。 6.増殖性細胞が、細菌、昆虫−由来細胞および藻類細胞から選択されることを 特徴とする、請求の範囲第1項に記載の方法。 7.上記結合ドメインがモノクローナル抗体であることを特徴とする、請求の範 囲第1項に記載の方法。 8.被検体が細胞表面抗原であることを特徴とする、請求の範囲第1項に記載の 方法。 9.上記被検体が哺乳動物腫瘍細胞のマーカーであることを特徴とする、請求の 範囲第1項に記載の方法。 10.上記試料が血液または骨髄であることを特徴とする、請求の範囲第1項に 記載の方法。 11.被検体が第二の透過細胞内に位置し、そして第1プローブが上記多機能性 プローブと組み合わせて使用されることを特徴とする、請求の範囲第1項に記載 の方法。 12.被検体が核酸であり、そして第1プローブがハプテン化ポリヌクレオチド であることを特徴とする、請求の範囲第11項に記載の方法。 13.上記増殖性細胞が発光しており、そして上記検出工程は感光フィルムを反 応マトリックスに、該感光フィルムを十分に露光するのに十分な時間適用し、そ して該フィルムを現像することを含むことを特徴とする、請求の範囲第1項に記 載の方法。 14.キットが (a)試料保持手段; (b)ヒドロゲル、増殖細胞および調節物質を含んで成る反応マトリッ クス、該細胞および該物質は該ヒドロゲル中に均一に分散しており、そして該マ トリックスは裏材材料に固定されている、ならびに (c)細胞成長促進ドメインおよび被検体結合ドメインの両方を有する多機能性 プローブ、 を含む、試料中の被検体を検出するための上記キット。 15.ブロット中に存在する被検体を検出するための、請求の範囲第14項に記 載のキットの使用。
