Claims (1)
- 【特許請求の範囲】 1.外来遺伝子を発現するための組換アデノウィルスベクター系であって: (a)リコンビナーゼにより認識される標的部位がウィルスのパッケージング シグナルの両側に隣接する改質早期領域1(E1)を有する第一アデノウィルス ベクター、並びに (b)前記1.(a)のウィルスの複製を変換でき、且つlox P部位の間での 部位特異的組換を触媒できる組換Creを更に発現する細胞系、並びに (c)(i)ウィルスゲノムの約35,000bpまでの大型領域の欠失を有するが、 ウィルスDNAの複製のため及びウィルス核酸のビリオンに至るパッケージングの ためにin cisである必要のあるAdウィルス配列、即ち、左方及び右方ITR並びに パッケージング配列は保持し、且つ (ii)サイズ約35,000bpの外来核酸の1又は複数のフラグメントを有す る、 第二ベクター、 を含んで成る組換アデノウィルスベクター系。 2.前記リコンビナーゼがCreであり、そして前記標的部位がlox Pである、 請求項1記載の組換アデノウィルスベクター系。 3.請求項1の(a)に記載のベクター(ヘルパーウィルス)を作製するため に利用するプラスミドであって、改質環状アデノウィルスゲノムを含んで成り、 ここで前記ウィルスゲノムの領域E3由来の配列は(a)リコンビナーゼのため の標的部位により囲まれたウィルスゲノム由来のパッケージングシグナル、(b )細菌性複製起点、及び(c)抗生物質耐性遺伝子、により置換されている、前 記プラスミド。 4.前記リコンビナーゼがCreであり、前記標的部位がlox Pであり、そして 前記抗生物質がアンピシリンである、請求項3記載のプラスミド。 5.請求項1の(c)に記載のベクター(ヘルパー依存性ウィルス)を作製す るために利用するプラスミドであって、ウィルスゲノムの左方及び右方ITR並び にパッケージングシグナル、プラスミド複製起点及び抗生物質耐性遺伝子、並び に長さ35,000bpまでの外来核酸を含んで成る前記プラスミド。 6.前記抗生物質がアンピシリンである、請求項5記載のプラスミド。 7.その核酸をビリオンへとパッケージングすることができず、またウィルス 遺伝子の全て又はほとんどが欠失され、且つ外来核酸で置換されている第二ヘル パー依存性ベクターの複製を補完するようin trasで作用できるウィルス機能を 保持するようなヘルパーウィルスゲノムを製造する方法であって: アデノウィルスベクターの複製を支持する細胞系を、 (a)Creリコンビナーゼにより認識されるlox P部位がウィルスのパッケー ジングシグナルの両側に隣接するようにして改質早期領域1(E1)を有する第 一ベクター及び (b)(i)ウィルスゲノムの約35,000bpまでの大型領域の欠失を有するが、 ウィルスDNAの精製のため及びウィルス核酸のビリオンに至るパッケージングの ために又はcisである必要のあるAdウィルス配列、即ち、左方及び右方ITR並びに パッケージング配列は保持している第二ベクター、 により同時感染せしめ、前記細胞の中で発現されたCreリコンビナーゼが前記第 一ベクターのパッケージングシグナルに隣接するlox P部位に作用して前記パッケージングシグナルの切除を誘導せしめることを含ん で成る方法。
