Claims (1)
- 【特許請求の範囲】 1.テトラサイクリントランスアクチベーター融合タンパク質をコードするポ リヌクレオチド分子を含んでなる組成物であって、前記タンパク質が、原核性te tリプレッサーおよび真核性転写アクチベータータンパク質を含み、前記ポリヌ クレオチド分子が、誘導性最小プロモーターに作動可能的に結合しており、前記 プロモーターが、少なくとも1つのtetオペレーター配列を含有するものである 組成物。 2.前記テトラサイクリントランスアクチベーター融合タンパク質をコードす るポリヌクレオチド分子のオープンリーディングフレームが、その5'末端で修 飾され、翻訳開始のための最適な環境を提供するものである請求項1に記載の組 成物。 3.前記テトラサイクリントランスアクチベーター融合タンパク質をコードす るポリヌクレオチド分子のオープンリーディングフレームの5'末端が、さらに 修飾され、唯一の制限部位を提供するものである請求項2に記載の組成物。 4.前記唯一の制限部位がHindIIIである請求項3に記載の組成物。 5.前記テトラサイクリントランスアクチベーター融合タンパク質をコードす るポリヌクレオチド分子のオープンリーディングフレームが、その5'末端で修 飾され、配列番号で識別されるオリゴヌクレオチドをコードするものである請求 項4に記載の組成物。 6.DNAである請求項1に記載の組成物。 7.請求項1に記載のポリヌクレオチド分子を含有するクローニングベクター を含んでなる組成物。 8.請求項1に記載のポリヌクレオチド分子によりトランスフェクションされ た真核細胞を含んでなる組成物。 9.前記真核細胞がさらに、少なくとも1つのtetオペレーター配列を含有す る誘導性最小プロモーターに作動可能的に結合した異種タンパク質をコードする ポリヌクレオチド分子によりトランスフェクションされているものである請求項 8に記載の組成物。 10.前記ポリヌクレオチド分子の少なくとも1つが、最小プロモーターおよび 7個のtetオペレーター配列に作動可能的に結合しているものである請求項9に 記載の組成物。 11.異種タンパク質の発現を減少または停止させる方法であって、 (a) 真核細胞を、 (i) テトラサイクリントランスアクチベーター融合タンパク質をコードする 第1のポリヌクレオチド分子であって、前記タンパク質はが原核性tetリプレッ サーおよび真核性転写アクチベータータンパク質を含み、前記ポリヌクレオチド 分子が誘導性最小プロモーターに作動可能的に結合しており、前記プロモーター は、少なくとも1つのtetオペレーター配列を含有するものである分子、 (ii) 前記異種タンパク質をコードする第2のポリヌクレオチド分子であっ て、前記タンパク質は、誘導性最小プロモーターに作動可能的に結合しており、 前記プロモーターは、少なくとも1つのtetオペレーター配列を含有するもので ある分子 により形質転換し、並びに (b) テトラサイクリンまたはテトラサイクリン類似体を含む培地中で前記真核 細胞を培養することを含んでなる方法。 12.前記第2のポリヌクレオチド分子が、最小プロモーターおよび7個のtet オペレーター配列に作動可能的に結合するものである請求項11に記載の方法。 13.異種タンパク質の発現を活性化または高める方法であって、 (a) 真核細胞を、 (i) テトラサイクリントランスアクチベーター融合タンパク質をコードする 第1のポリヌクレオチド分子であって、前記タンパク質は、原核性tetリプレッ サーおよび真核性転写アクチベータータンパク質を含み、前記ポリヌクレオチド 分子が誘導性プロモーターに作動可能的に結合しており、前記プロモーターは、 少なくとも1つのtetオペレーター配列を含有するものである分子、 (ii) 異種タンパク質をコードする第2のポリヌクレオチド分子であって、 前記タンパク質は、誘導性最小プロモーターに作動可能的に結合しており、前記 プロモーターは、少なくとも1つのtetオペレーター配列を含有するものである 分子 により形質転換し、並びに (b) テトラサイクリンまたはテトラサイクリン類似体を欠く培地中で前記真核 細胞を培養することを含んでなる方法。 14.前記誘導性最小プロモーターに対するテトラサイクリントランスアクチベ ーター融合タンパク質の結合を抑制するために十分な量のテトラサイクリンを含 有する請求項8に記載の組成物。 15.前記テトラサイクリントランスアクチベーター融合タンパク質をコードす るポリヌクレオチド分子が、テトラサイクリンの不存在下に前記異種タンパク質 をコードするポリヌクレオチド分子の発現を駆動するために十分な量に発現され る請求項9に記載の組成物。 16.テトラサイクリントランスアクチベーター融合タンパク質が、前記異種タ ンパク質の発現を駆動するために十分な量に存在する請求項9に記載の組成物。 17.プラスミドpTet-Spliceより実質的になる組成物。 18.プラスミドpTet-tTAKより実質的になる組成物。 19.少なくとも2つの収容手段を内部に密閉的に有する担体手段を含んでなる キットであって、第1の収容手段が、テトラサイクリントランスアクチベーター 融合タンパク質をコードする第1のポリヌクレオチド分子であって、前記タンパ ク質が原核性tetリプレッサーおよび真核性転写アクチベータータンパク質を含 み、前記ポリヌクレオチド分子が誘導性最小プロモーターに作動可能的に結合し ており、前記プロモーターが少なくとも1つのtetオペレーター配列を含有する ものである分子を含有し、第2の収容手段が、前記誘導性最小プロモーターをコ ードする第2のポリヌクレオチド分子であって、前記プロモーターが少なくとも 1つのtetオペレーター配列を含有し、前記tetオペレーター配列がポリペプチド をコードする異種ポリヌクレオチド配列に作動可能的に結合するために効果的に 配置されるものである分子を含有することを特徴とするキット。 20.少なくとも2つの収容手段を内部に密閉的に有する担体手段を含んでなる キットであって、第1の収容手段が、テトラサイクリントランスアクチベーター 融合タンパク質をコードする第1のポリヌクレオチド分子であって、前記タン パク質が原核性tetリプレッサーおよび真核性転写アクチベータータンパク質を 含み、前記ポリヌクレオチド分子が誘導性最小プロモーターに作動可能的に結合 しており、前記プロモーターは、少なくとも1つのtetオペレーター配列を含有 するものである分子によりトランスフェクションされた真核細胞を含有し、第2 の収容手段が、誘導性最小プロモーターを含む第2のポリヌクレオチド分子であ って、前記プロモーターが少なくとも1つのtetオペレーター配列を含有し、前 記tetオペレーター配列が異種ポリペプチドをコードする異種ポリヌクレオチド 配列に作動可能的に結合するために効果的に配置されるものである分子を含有す ることを特徴とするキット。
