Abstract
PURPOSE: To determine the vitamin B12 in food, feed, blood, etc., in high reproducibility and sensitivity, by measuring the degree of proliferation of vitamin B12- requiring marine methanol-assimilable bacteria.
CONSTITUTION: Vitamin B12 is determined using vitamin B12-requiring, marine methanol-assimilable bacteria, i.e. Pseudomonas marinomethanolica YK4007 (FERM-P No.3621) or Methylomonas methylomarina YK2004 (FERM-P No.3622). The procedures are as follows. Add a series of standard vitamin B12 solutions having different concentrations to test tubes containing methanol. Add the sample solution to be determined to a test tube containing methanol. Inoculate and culture the above bacteria in the test tubes. Determine the number of the proliferated bacterial cells in each test tube by measuring the optical density (OD value) of the solution. Compare the OD value of the sample solution with that of the standard solutions, and determine the B12 concentration of the sample solution.
USE: The process enables the determination of vitamin B12-active substances in sea water and vitamin B12 in various various marine products which is difficult by the conventional microbiological process.
