Abstract

PURPOSE: To provide an enzyme activating plasminogen with stabilized activity (such as resistance to heat treatment, stability during transportation, or during freeze drying process) by combining specific substances.
CONSTITUTION: Urokinase and dextran sulfate of 1,000W20,000 molecular weight (instrinsic viscosity of 0.030W0.040, sulfur content of 3W6%) are combined at a ratio of 1:10W10:1 to form covalent bonds, thus giving said enzyme. The enzyme is obtained by effecting reaction between dextran sulfate and cyan halide under alkaline conditions (0.1W5 times amount, based on the dextran sulfate, of cyanogen halide is used at a pH of 8W13.0 and 0°C W room temperature), followed by acting urokinase on the activated dextran sulfate at a ratio of 1:10W10:1 urokinase to dextran sulfate at 0°C W room temperature under weak alkali conditions for 1W30hr.
