Abstract

PURPOSE: To obtain the title compound useful as an intermediate for nucleoside, by treating a 5-halogeno-S2,5'-cyclo-2-thiouridine with an alkoxide, and by hydrolyzing the reaction product.
CONSTITUTION: A novel 5-halogeno-S2,5'-cyclo-2-thiouridine of formula I (X is halogen; X is protecting group), e.g. 5-bromo-S2,5'-cyclo-2',3'-O-isopropylidene-2- thiouridine, is treated with an alkoxide, e.g. sodium methoxide, to give an S6,5'- cyclo-O2-alkyluridine of formula II (R is alkyl group), which is hydrolyzed to form an S6,5'-cyclouridine, e.g. S6,5'-cyclo-2',3'-O-isopropylideneuridine, of formula III (Z' is H or protecting group).
USE: Reagents for biochemical studies, e.g. synthetic substrates for studying the specificity of nucleic acid metabolic enzymes.
