Abstract

PURPOSE: To determine acyl coenzyme A useful for the clinical measurement of free fatty acids in blood, easily, accurately and rapidly, by measuring the H2O2 produced by the oxidative activity of acyl coenzyme A-oxidase.
CONSTITUTION: A liquid specimen containing acyl coenzyme A is adjusted to pH 7.0W8.5, and added with an acyl coenzyme A-oxidase, an SH reagent, a peroxidase or catalase, and a color developing reagent, separately or at a time. The mixture is heated at 37°C for 5W60min in an open atmosphere, and the optical density is measured using a light corresponding to the color of the liquid, e.g. a light having a wave length of 400W600nm. The amount of the acyl coenzyme A is determined from the calibration curve prepared by the process similar to the above. The addition of the SH reagent (a maleimide derivative, iodoacetamide, 4,4-bisdimethylamino-benzhydrol, etc.) improves the sensitivity of the measurement.
