Abstract
PURPOSE: To separate hyaluronic acid and chondroitin sulfate from animal tissue, easily, in high yield, by heating the animal tissue in a solution of a quaternary ammonium salt, and treating with protease to separate hyaluronic acid, and then treating the residue with a salt solution to obtain chondroitin sulfate.
CONSTITUTION: Animal tissue such as cockscombs, naval cords, skin, etc. is heated in an aqueous solution of a quaternary ammonium slat, cut to pieces, added with water, and treated with protease. The treated solution is filtered to concentrate hyaluronic acid in the filtrate (solution A). The residue is treated with a solution of a salt such as NaCl or of an alkali such as NaOH, to collect chondroitin sulfate in the filtrate (solution B). The hyaluronic acid and the chondroitin sulfate are precipitated by adding an alcohol to the solutions A and B.
