Abstract

PURPOSE: To separate superoxide.dismutase from the supernatant liquid obtained by disintegrating the tissue of human placenta with an aqeous solution of potassium chloride or sodium chloride.
CONSTITUTION: The sufficiently blooded fresh human placenta obtained through normal delivery, or its frozen product is added with 5 times weigth of 1.15% potassium chloride solution or 0.9% sodium chloride solution, and maintained at 0W4°C to effect complete disintegration of the tissue. The precipitate is removed therfrom by centrifugal separation, and the supernatant liquid is heated at about 60W75°C for about 5min. The produced precipitate is removed by filtration, and the filtrate is mixed with ammonium solfate solution of 50W90% saturation. The precipitate is dissolved and dialyzed, and the dialyzate is subjected to ion exchange Sephadex chromatography or treated with a carrier bonded with superoxide.dismutase antibody, to obtain mitochondrian.superoxide.dismutase and soluble fraction of superoxide.dismutase.
