Abstract
PURPOSE: To conduct the liquid chromatography of the constituent of low molecular weight without deteriorating the performance of a separating column by a method wherein the constituent of high molecular weight is removed beforehand by using a water-size separating column and the constituent of low molecular weight alone is introduced into the main column for separation.
CONSTITUTION: First, a sample for measuring the flow-out velocity is injected in the eluent and is introduced into the water-size separating column, and then the time from injection of the sample to the point of time when the flow-out of the constituent of high molecular weight in the sample from the column is completed and the constituent of low molecular weight starts to flow out is measured beforehand. Next, the sample to be measured is injected into the eluent and then passed through the water-size separating column together with the eluent, and there the constituent which flows out by the time measured beforehand is removed. And, the constituent which flows out after the lapse of a prescribed time alone is introduced into the main column for sepatation and thus the analysis of the constituent of low molecular weight is performed with a liquid chromatography.
