Abstract

PURPOSE: To obtain the titled compound in high yield without preparing undesirable sulfides as by-products, by converting an N-halogen-substituted acylamino acid or its salt into a thiol with an inexpensive alkali metal hydrosulfide in a polar nonproton solven.
CONSTITUTION: An N-halogen-substituted acylaminoacid shown by the formula I [X is halogen; R is H or CH3; Y is O, S, SO, SO2(CH2)m(m=1 or 2); (n) is 1W3] or its salt is reacted with an alkali metal hydrosulfide or alkaline earth metal hydrosulfide in a polar nonproton solvent (e.g., N,N-dimethylformamide, etc.) to give the desired compound [e.g., 1-(3-mercapto-2-D-methylpropanoyl)-L-proline, etc.] shown by the formula II.
EFFECT: Racemization will not occur at all under heating at 40W60°C.
USE: A hypotensor, etc.
