Abstract
PURPOSE: To determine the quantity of microbial cells, by culturing the microorganism in a medium containing a saccharide at a specific concentration range under aerobic conditions, and measuring the concentration of ethanol, or by collecting the above cultivation medium, adding said medium to a saccahride- containing medium charged in a separate cultivator, and measuring the produced ethanol.
CONSTITUTION: Microorganism capable of producing ethanol from saccharide is cultured in a cultivator 3, and a saccharide such as glucose, fructose, etc. is added to the cultivation liquid 8 at a concentration of 0.1W10g/l. Ethanol produced by the microorganism is sent to the detector with a carrier gas 6 through the "Teflon " tube 7, and its concentration is measured to determine the quantity of the microbial cells. As an alternative method, a proper amount of the above cultivation medium is added to the medium charged beforehand in a separate cultivator, and the amount of produced ethanol is measured. In this case, the concentration of saccharide in the medium prepared beforehand is adjusted to 0.1W50g/l.
