Abstract
PURPOSE: A sample for measuring enzyme activity is made act on L-leucylamide, then an immobilized oxidase is made to act on the reaction product to measure the amount of consumed oxygen or formed hydrogen peroxide, thus conducting the measurement of enzyme activity.
CONSTITUTION: An L-leucylamide of the formula (R is substituted methylamino group or residue of D-aminoacid) or its soluble salt is used as a substrate, on which a sample for measuring enzyme activity, which liberates methylamine or D-aminoacid by the reaction, is made to act. The L-leucylamide is, e.g., L-leucyl-benzylamide, L-leucylthylamine, L-leucyl-n-butylamide, L-leucyl-D-methionine or the like. An oxidase is made to act on the reaction product, i.e., substituted methylamine or D-aminoacid, to measure the amount of the consumed oxygen or formed hydrogen peroside, thus determining the activity.
