Description
本発明は単純ヘルペスウィルス1壓および2型(一般的
にH8V−1、H8V−2と略記される)に対する予防
および治療のためのワクチンに関するものである。更に
詳しくは、H8V−1およびH8V−2の糖タンパン質
Bの中に見い出されるアミノ酸鎖と一致する配列を全要
素として構成されているアミノ酸鎖を発現するために必
要な遺伝子以外を除外したヌクレオチドを含むプラスミ
ドの生成のだめの、組、換えDNA技術の応用に関する
ものであシ、さらに、そのDNAによって発現されるア
ミノ酸鎖、H8V−1またはH8V−2に対して効果的
免疫応答を生ずるそれらの誘導体、そのDNAによって
生成したアミノ酸鎖生成物に基ずくワクチンにそれぞれ
関するものである。
本発明は生存原核細胞、生存真核細胞(これらは組換え
DNAを挿入されたり、取シ出される能力を持ち、且つ
そのDNA分子を発現することができる。)において、
糖タンパク質B中の、タンパク質部分の合成を誘導する
のに用いられるDNA分子を、与える。本発明組換えD
NA技術を使用することは、コスト的に効率がよく、生
成されたアミノ酸鎖は、生ウィルスによる汚染の危険の
ない、合成、予防、治療薬である。
単純ヘルペスウィルス1型および2型のDNAは、15
0 kb を含有し、10〇−200のタンパク質を暗
号化するために十分な情報を持つ。約30のタンパク質
は、ウィルス粒子中に存在し、その残存暗号化タンパク
質は、宿主細胞中のウィルスの複製や集合に関係してい
る。タンパク質膜に囲まれたDNA核に加えて、ウィル
スは、外膜構造を持つ。その膜は、脂質およびウィルス
暗号化糖タンパク質から構成されている。これらの糖タ
ンパク質は、特有なアミノ酸残基で始する1つまたはそ
れよシ多くの糖鎖を持つタンパク質である。4つのH8
V 糖タンパク質gA/gB(gAはgBの先駆体)、
gC,gD、およびgE が存在する。
H8Vは、おそらくウィルス膜および細胞膜の融合によ
シ宿主細胞中に侵入し、この溶解にはgB が介在する
。ウィルスは、核において複製する。複製の間に、糖タ
ンパク質は合成され、感染細胞の原形質膜外表面まで達
する糖タンパク質の多くと共に、細胞表面に移送される
。ウィルス糖タンパク質はウィルス表面および細胞表面
から外部に出るので、H8V特異抗体と結合する最初の
ウィルス分子である。
一般的にワクチンは、総ての要素を持つウィルス粒子(
生ウィルス、弱毒ウィルス、化ウィルス〕または、ウィ
ルスの構造タンパク質(サブユニットワクチン〕から成
シ立っている。生H8Vワクチンについて、有効性を認
める報告があるけれども、多くの専門家は、その方法を
勧めない。なぜ欧らば、H8Vは、発癌活性および発癌
補助活性を持つ可能性があり、さらに一般的に言って、
人の生存を脅かすタイプのウィルスでないからである。
これに対して、サブユニットワクチンは、1つまたはそ
れよシ多くの糖タンパク質あるいはそれらの非グリコジ
ル化i non −glycosylated)アミノ
酸鎖で構成されておシ、どちらの免疫活性部位をとって
もよシ望ましい。生ワクチンは、生成が安価であるため
魅力的であフ、予防接種の後、増殖するので大変少量で
すむ。
しかし、危険性を秘めている。サブユニットワクチンは
、タンパク質の多量生成が必要とされるし、コストも高
額となるのであるが、安全性がある。
本発明による組換えDNA技術による大腸菌中で合成さ
れたウィルス糖タンパク質およびタンパク質成分は、ウ
ィルスDNAに完全に制約されない。総ての要素を持つ
DNAでなく、唯一単独のウィルス糖タンパク質遺伝子
が存在する。組換えDNA技術は、ワクチン生成にかか
る生ウイルス汚染に関する危−艙を除くものである。
主なウィルスタンパク質の内の唯一のgBは、すでに生
物学的機能が知られておシ、ウィルス生長のだめの基本
要素であることが明らかにされている。以下の例に続く
表に、H8V−1およびH8V−2のそれぞれgB の
D’NA暗号化配列を示す。本発明は、糖タンパク質B
のアミノ酸鎖を発現する能力を持つヌクレオチド配列を
示す。微生物へ遺伝子を挿入することにより、H8V糖
タンパク質に対応するタンパク質を合成することができ
る。
gB の糖部分でなく、タンパク質部分のみが、細菌中
で合成されるが、そのタンパク質は、免疫決定基の主要
な根源物質である。発現のため、微生物、つまシ大腸菌
の様な典型的な細菌は、要求されるアミノ酸鎖を発現す
る能力を持つプラスミドを含む普通増殖培地で増殖され
る。本発明において、細菌の膜断片から抽出された、本
発明の精製H8Vタンパク質は、タンパク質に対する抗
体を作るために、動物(例えば、ウサギ)へ接種されう
る。細菌中で作られたgB タンパク質に対する応答で
、動物体内で生成されたこれらの抗体は、ウィルスを不
活化(中和)する能力について試験されうる。
細菌中で、H8V−gB 発現が、極限まで増加した後
、およびgB に対して生成された抗体による適切なウ
ィルス中和の実証の後、gBタンパク質ワクチンは、動
物で試験され、最後に臨床試験において評価されうる。
ウサギに対する約3μg タンパク質/に9の一回接種
で、抗体応答を生じさせる。
gB の完全に暗号化された配列が、細菌において発現
され得たが、免疫学的に活性を持つgB の一部のみを
発現することが、なぜ望ましいかには、実践的な理由が
ある。もし大腸菌の細胞質において発現されたgB の
3次元構造を変えうるならば、2つの広範囲におよぶ疎
水性部位、つま)信号部位と、膜への橋渡し部位が存在
する。(後に記す、配列に関する要約参照。)完成した
タンパク質が、感染細胞膜中に存在する時、信号配列は
、酵素的に除却され、膜への橋渡し残基は、膜中に存在
する。膜の細胞質側における残存C末端残基は、抗体に
近すきえない。従って、903全体の内の75ON末端
アミノ酸残基までが、ワクチンに関して、抗原的に活性
を持つと思われる。
親水性残基は、タンパク質表面上に存在すると思われる
。β−螺旋の領域中のこれらの残基は、抗原決定因子の
候補者である。しかし、1次構造と、3次構造が、1次
元的に関連しないゆえに、抗原部位を形成する親水残基
は、必しも近接していると言えない。それにもかかわら
ず、長い鎖の内の適切に選択された、長さが10−20
のアミノ酸の近接残基は、たいてい抗原性を持ち、その
残基を持つ本来のタンパク質と結合する抗体の生成を導
きえる。従って、最も単純なワクチンは、高抗原活性を
持つ、ショートペプチドであシ、そのペプチドの配列は
、gB 遺伝子のヌクレオチド配列の知識から導びかれ
る。
H8V−1プラスミド
pKBXXと呼ばれるクローンにより例示される、本発
明における単純ヘルペスウィルス1型の代表的な組換え
DNAクローンは、以下に示す方法によシ作成される。
KO8系統から精製されたDNAは、制限酵素の供給者
によって決められた反応条件を用い、制限エンドヌクレ
アーゼBam HI を使って切断される。同時に、ベ
クターDNApBR322(参照Boljvar et
al、e Gene 2.95−113.1977]
はgam HIを使って切断される。仁の酵素は、有機
溶媒による抽出によシ除去させることができ、不活化さ
れうる。つまfi、DNAを低温でエタノールを使って
沈降させ、吸引による、乾燥によシ、残存溶媒は、取シ
除かれる。
BamHI 切断H8V−IDNAは、Bam HI
切断pBR322ベクター(このベクターは、テトラサ
イクリン耐性遺伝子 (rerlにおいて独特なJLL
IIIHI部位を持つ)の、類似した粘性末端と結紮さ
れる。結紮は、メーカーから提供された条件によシ、約
4℃で、T 4 DNAリガーゼを使って便宜に行なわ
れる。切断反応も結合反応も、どちらも密閉されたプラ
スチック容器中で行なわれる。
反応の同様な継続は、再びKO8系統を用いて同酵素単
位数の制限エンドヌクレアーゼEcoRIを用いての切
断および旦c oRIを用いて、ベクターDNApBR
325(Bolivar 、 Gene 4 。
121−136 :1978)を切断することで行なわ
れる。酵素は再び除かれ、EeORI切断H8V−1は
、共通した結紮条件を使って、pBR325(クロラム
フェニコール耐性fcmrl遺伝子において独特なEc
oRI部位を持つJの類似した末端と結紮された。
プラスミドpBR325は、アンピシリン(Apr)、
テトラサイクリン、クロラムフェニコールに対して薬剤
耐性を明確に示す遺伝子を含み、pBR322は、アン
ピシリン耐性およびテトラサイクリン耐性遺伝子を持つ
。
大腸菌中への結紮混合物の導入の結果、H8V−1断片
を含むことが推測されるベクターは、pBR325につ
いては、クロラムフェニコール、pBR322について
はテトラサイクリン(これらの物質の存在において生長
能力を失なう)に対する感受性としてでなく、アンピシ
リンに対する抵抗性によシ同定される。薬剤耐性遺伝子
および他の遺伝子は、異種断片または無関係DNAによ
シ、はとんどいつも不活化される。
旦eoRI−F断片またはμtmHI−G断片を含む組
換えプラスミドは、これらの酵素について、H8V−1
の既知の制限地図と、H8V−1挿入物の電気泳動的移
動性を比較することによシ同定される。制限断片は、ヌ
クレオチドの長さの減少と一致している、アガロースゲ
ル上の移動性の増加の順に、アルファベットA−Zの内
の大文字で示される。H8V−1の同系統1すなわちK
O8は、どれかひとつの制限エンドヌクレアーゼについ
て、唯一ひとつの制限地図を現わすであろう。しかし、
異なる系統のH8V−1は、KO8と全く無関係に増殖
していくので、必ずしも全てではないが、同じ制限部位
の大部分を共有する。

断片は、前述の結紮法によりまず作られ、それから5
alIを使っての部分消化を受け、Sal I断片を除
くために再び結紮される。これは0345シ児H1部位
から0.3878al1部位までの座標を持つフラグメ
ントpKBG −BS3を生ずる。これは2つのXho
llo、368およびQ、372)部位を含む。
pKBXXにおいて、要求された配列を得るために、p
KBG−BS3はとHIを使った完全消化およびXho
lを使った部分消化を受けさせる。アガロース電気泳動
の結果、0.345から0.372までの13amHI
−Xho I断片と対応する4、Okb結合結合層認
され、ゲルから電気溶離され、gam旧、互配I切断p

RNAおよびタンパク質合成の方向は、左から右へであ
る。多重クローニング部位は、大腸菌β−ガラクトシダ

H8V−1クローン化配列(但しH8V−1配列におい

ターは、H8V−1クローンを同じ制限部位ヲ持つ他の
いくつかのベクター中で再クローン化させ得る。pKB
XXを作る目的で、pUC9DNAは、BamHIおよ
び5alIを処理され、H8V−1配列(BamHIか
らXhoIまでf 0.345から0.372まで)は
、これら処理された部位と結紮される。その適当なりロ
ーン(形質転換分析の後の)は制限地図によシ同定され
た。
pKBXX内に含まれるH8V−1配列は、上述の制限
地図を得るための制限部位の記載中のそれであることが

状態によって異なるゆえに、それらは異なる配列部位で
DNAを切断するが、粘性の強い末端部位+TCGA1

長したコロニーは、プラスミドDNAを調製するために
使われる。
実施例2
プラスミドpKBG−BS3
先の実施例で得られたB a m Hニー Gクローン
は、先の実施例の方法によシ制限酵素処理を受ける。下
に示す5atIを使った代表的な反応において、使用さ
れた制限緩衝液は、以下を含むRB−8である。
トリス緩衝液 6□MpH7,4

こうして、10μPBamHI−Gは、RB制限緩衝t
i、25μを中において、室温で15分間、約1単位の
5atI酵素で部分消化される。実施例2の中での成果
は、0.3458amH1部位から0.387SatI
部位までの座標で、断片pKBG−BS3を生産する。
実施例3
pKBXXを得るために、10μノのpKBG= B
S −3は、2時間、8単位のBamHIを使って完全
に消化した後、RB−1中で1単位X h o Iを用
いて部分消化される。
どちらの場合も、制限緩衝液1が使われる。
ここに用いられた様な部分消化において、1単位の酵素
は、10μ7 のDNAについて使用され、反応時間は
、15分に減する。
アガロース電気泳動において、0.345から0.37
2のBamHI−Xh o I断片に対応する4、 O
K b結合物が、得られる。この断片は、ゲルから電気
溶離され、24°lのクロロホルム対3メチルブタノー
ルの混合液の等量を使って抽出され、エタノール沈殿さ
れる。この断片は、それから、BamHI−8aAI切
断pUC9と結紮される。かくして、pUC9は、初め
に前述の方法により、BamHIおよび5atIの作用
を受け、それから、対応する部位に対して、H8V−1
配列(上記電気溶離された断片、o345BamH1か
ら0.372 X h oI)と、実施例IBの全力法
によって結紮される。
実施例4
pBR325のEcoRI部位への、H8,V−2(H
G52系統)のEcoR1断片のクローニング
A、制限エンドヌクレアーゼ処理
エツペンドルフバイアル(Eppendorf via
l )(プラスチック製で、スナップキャップを持つ)
内で、pBR325[ベセスタ調査研究所〕2μt (
04μ51)、HG52 36μt(4μ9)、Eco
R)5μt(40単位)は制限緩衝液2の110μを中
で消化される。
制限緩衝7& 2は、pH7,4の100 m M ト
リス緩衝液、50 nt M塩化ナトリウム、10mM
塩化マグネシウム、10mMメルカプトエタノール10
0μy/mtヌクレアーゼフリー牛血清アルブミンを含
む。消化は、37℃で3時間行なわれる。
消化の後、反応混合物は、TE(1mMEDTA1p1
18の10 m M トリス)を使って400μtに希
釈される。この溶液は、制′限エンドヌクレアーゼを不
活化するために、15分間65℃へ加熱し、それから、
室温に冷却される。この溶液は、それから、いくらかの
残存酵素を不活化し、取シ除くために、24:1のクロ
ロホルム対3メチルフタノールとの混合液の等量を使っ
て2回抽出される。
他のプラスチックバイアルへ、残余溶液240μt を
移した後、ここに、5M塩化ナトリウム60μtおよび
、冷却エタノール500 ttl(約2倍量)が加えら
れる。混合物は、30分間−75℃で保持され、後、上
清は、除かれ、DNAペレットは、エタノールを除去す
るために吸引乾燥される。
B、 ’)ロー:/ p 52 KG作tL−のための
結紮乾燥ペレット(pellet ) は、リカーゼ緩
衝液25μを中に再浮遊される。(リカーゼ緩衝液は、
pl+ 7.8の50 m M トリス緩衝液。

最初のバイアルにおいて、p52EG2011A −(
約5μ2)は、37℃で90分間2X制限緩衝液122
μを中で、J(amHI、Hi n d、ut、 Ec

RIの1μlを使って消化される。それからi量の1M
トリスが加えられ、培養は更に90分継続する。
第二のバイアルにおいて、1oμtのpUC9は、制限
緩衝液] 45μlと共に、B a mHI 、H3n
dIllの1μtf使って、37℃で3時間培養される
。
制限緩衝液]、 (Re5tiction buffe
r 1 )は、pl+7.4の]0mMトリスを含み、
その緩衝液は、他の点では制限緩衝液2と同様である。
消化後、切断DNAは、0.75%アガロースゲルにお
ける電気泳動により分離される。目的結合物は、アガロ
ースから電気溶離され、それから実施例4のように抽出
きれ、結紮され、大腸菌を形質転換するために便われる
。
実施例6
p52EG−BHからのp52BXXの単離最初のバイ
アルにおいて、p52gG−BHIOμt(約5μり
)は、制限緩衝液1 40μ、/、+:IQこ、B a
m J(]、 +Hi nd用の1μlと共に加えら
れ、37℃で2時間30分培養される。それから、、X
ho11単位が加えられ、培養は、p52EG−BHの
多くの分子における2つのXh。
■部位の内の1つのみを切断するために15分間継続さ
れる。
第2のバイアルにおいて、10μtのpUC9は、制限
緩衝液RB−84011を中で、37℃で2時間30分
、f3amHIの1μtおよび 5ajIの2μtを使
って消化される。
使用された制限緩衝液はRB−8であり、以下を含む。

実施例7
細菌における、pKBXXからの1B断片の発現
A、rB断片の生成
10μtのp K B XX D N A (約10μ
g)および、10μtのnazI制限エンドヌクレアー
ゼは、等量の2XB a tI緩衝液(Naαを含まな
い制限緩衝液RB −]、 )へ加えられ、DNAは、
エツペンドルフ・スナップキャップ・バイアル中で、3
7℃で16時間消化される。0.7%アカロースゲルを
用いた′電気泳動による、2つのDNA断片(1,48
Kb、5.20Kb)の分離の後、要求される1、48
Kbは、ゲルから電気溶離される。
DNAは、残存BatI酵素を不活化し除却するために
、フェノール、クロロホルム、3メチルブタノールのそ
れぞれ24:24:1の混合物の等量を使って2回抽出
される。
DNAを含む水層は、酢酸ナトリウム(3M保存溶液)
へ、入れ、−75℃で、30分の保管期間冷却エタノー
ルの2倍量を加えることにより沈殿される。エタノール
をゆるやかに注ぎ、DNAを吸引乾燥し、これを500
μy/μtの牛血清アルブミン25μtに、等量の2
X B a L 31緩衝液(pIIs、 oの40m
Mトリス、24 mMcaC1!2. 24 mMM9

)および2 lit のBat31(2,5単位)を
加えた溶液に再浮遊される。酵素およびDNAのこれら
の濃縮は、DNAの各末端から、約100ヌクレオチド
/ minの加水分解を起こさせる。
Bat31調製物は、5′ から3′ も3′から5′
もどちらについても、エキソヌクレアーゼ分解活性(
exonucleolytic activity )
を含み、ゆえに、DNAのどちらの鎖も、加水分解され
る。5μlのサンプルがOから3分の間に20秒間隔で
取シ出され、50μtのフェノール−クロロホルム−3
メチルブタノールヲ含有するエツペンドルフスナップキ
ャツプバイアルへ加えられる。(前の段落参照)
ためられたDNA断片は、前に示したように抽出され沈
殿され、50μtのTE中に再浮遊される。さらに、サ
ンプル量中の小変化部を持つ残存モノヌクレオチドを除
却するために、回転透析によってでファデックス小型カ
ラム中を通過される。
これに、HinclIを使った培養によりプラント末端
化されたpUC9ベクターDNA1μ(約0.1μ9)
が加えられる。通常、10μMATPを共なう5μtの
IOXリカーゼ゛緩衝液[]、Xリガーゼ緩衝液につい
てのセクション4(実施例4)を参照〕は、3μtのT
4DNAリカーゼ(4x10sユニット/mt ) と
共に加えられる。得られた溶液は、室温で16時間培養
され実施例ICにおけるように大腸菌を形質転換するた
めに、ただちに使われる。
B、抗原的に活性なIB断片
上記の形質転換実験から得られた、独特なコロニーから
の細菌は、10m1の培養液に入れられることに使用さ
れ、05の光学密度(OD)に生長される。おのおのの
培地からのjT−[1111は、ペレット化され、10
%トリクロロ酢酸(TCAj中に再浮遊され、沈殿物は
、同量の10%TCAを使って、エラペンドルススナツ
プキャップバイアル中で洗浄される。
最後にペレットは、■00μtのTE中で洗浄され、p
H8,02Mトリス−ベース(Tris−base )
2μtを加えることにより中和される。
10μlのサンプルは、ニトロセルロースフィルターへ
庄かれ、37℃で30分間、風乾される。細菌のタンパ
ク質やDNAは、フィルターへ拘束され残るであろう。
ニトロセルロースの細片(145X190喘)は、緩衝
液A(1)117.5の10mMトリス。
17(1mMNaα、0.1mMフェニルメチルスルホ
ニルフルオリド、さらに加えて、0.01%SDSおよ
び10mM塩化マグネシウムも含む) 5 mA 中に
浸される。DNAは、2 ttq/ mA のデオキシ
リボヌクレアーゼl(DNa5el)溶液■0μtを加
えることにより、10分間消化される。ニトロセルロー
スのすべての処理は、]、8X150+mnスクリュー
キャップ(screw−cap )型試験管中において
行なわれる。ニトロセルロース自体は、試験管の内側に
巻きつけられる。細片は、約10rpmで回転するドラ
ム上で試験管を回転させることにより、すべて連続培養
基中に浸すことにより保持される。
DNa s e ■およびヌクレオチドは、OD】%S
DSを含む緩衝液A 5 ml で2回、5分間づつ洗
浄することによシ除かれる。H8V−1抗体の非特異的
結合は、0,01%SDSおよび10%馬血清を含む緩
衝o、A 3 ml を使って、ニトロセルロースを処
理することによりブロックされる。処理は3時間行なわ
れる。細片は、001%SDSを含む緩衝液A5mtで
2同各10分づつ洗浄される。
H8V−1抗血清(精密な科学原理に基すいて典型的に
得られた)は、細菌溶解物(Lysite ) 中に存
在する、抗原的活性を持ついくつかのfIB断片へ、血
清中のfB抗体を結合させるために加えられる。10μ
tの抗血清は、500μtの溶液B(1?MMEDTA
、0.1%SDS、0.1%トリトンX−100,およ
び4%馬血清を含む緩衝MA)へ加えられ、室温で16
時間結合させられる。
溶液83 ml で2回、10分間づつ洗浄の後、2μ
Ciの1251タンパク質A〔ニューイングランド核(
New England Nuclear ) :]を
含む溶iB 500μlが加えられる。タンパク質Aは
、黄色ブドウ球菌からのものであり、■2G抗体の一定
領域と結合する。タンパク質Aは、室温で3時間で、こ
れらの部位と結合させられる。最後に、フィルターは、
馬血清をitない溶液85 mA で5回、15分づつ
洗浄され、乾燥され、24から72時間X−OmatX
線フィルムにさらされる。
細菌において合成されたfB生成物を試験するために、
Bat31清化DNAから合成された1Bペプチド(実
施例7)および、後述されている最後のクローンの調製
物(実施例8参照)は、SDS電気電気的動的離された
ペプチドのラジオイムノ反応により試験され得る。1B
生成物が、taCプロモーターの制御のもとに存在する
ゆえに、その生産は、tacタンパク質(IPTGのよ
うな)の合成を誘導する化学物質によって誘導されなけ
ればならない。
細菌の10 mt 培養は、)PTGの存在のあるなし
において、0D05まで生長され、細胞はペレット(p
ellet ) 化され、10%の冷却されたT CA
l ml 中に再浮遊され、30分間冷却される。2
回目の遠心の後、細胞は、10%TCAで洗浄きれ、最
後のペレット(pellet ) は、pH8,Oの2
Mトリス−ベース(base ) 2μtで中性にされ
、電気泳動サンプル溶液(pH7,0の5mMトリス、
2%SDS、5%βメルカプトエタノール。
0.005%ブロムフェノールブルー)の100μを中
に再浮遊される。10μtのサンプルは、9.5%ゲル
へ与えられ、ペプチドは、100ボルトで6時間、分離
される。
このペプチドは、ニトロセルロースシート上に電気的フ
ロント(electroblott ) 化され、その
シートは、風乾される。2Bペプチドは、ラジオイムノ
アッセイによp、1i’Bの存在について検定するため
に、実施例7に示しだようにニトロセルロースシートを
処理することに視覚化される。予想された犬きさのペプ
チドが視敵化されるはずである。
ウサキヘ接棟されるだめの抗原として使用するyB断片
を調料するために、誘導細菌培養物は、TCAで処理さ
れ、第1段落で述べたように、電気泳動サンプル溶液中
に再浮遊される。ただし、総容量は、約100μ2 の
2Bペプチドを得るために10倍に増加される。ゲルの
適当な領域からのfBは、フロイントのアジュバント中
で、つぶされどろどろにされ、抗体生成のためにウサギ
に接種される。25μ2 のペプチドは、最初および引
き続く2回目の注射において1更用され得る。次に、ウ
サギからの抗血清は、1B沈降能および、ウィルス中和
能について試験され、それによって、望ましい分画が確
認される。
実施例8
細餉中のp 52 BXXからの2B断片の発現A、1
B分画の生成
エツペンドルフスナップキャツプバイアル(Eppen
dorf 5nap cap vial )中のp52
BXX DNA(25μ2m)はエタノール沈殿され、
ペレット(pellet ) は、70%エタノールで
洗浄され、吸引乾燥されそうして45μtのRB 6
(100m M Naα、pH7,4の6mMトリスH
α、6 m M MFα2.5mM2メルカプトエタノ
ール、100μ’ / mt−BSA)中に再浮遊され
る。制限エンドヌクレアーゼNrul(4unit/μ
tのものを4μt)が加えられ、その混合物は、37℃
で一晩、牛血清アルブミンの存在下で培養される。反応
停止溶液(5μt)が加えられ、その反応混合物は、反
応停止溶液中の、不活化酵素、洗浄物、染色物を除去す
るだめに、TE緩衝液中で、バイオゲル(Bio Ge
l ) P −100を使用する回転透析を2回サイク
ル受ける。DNAはエタノール沈殿され、洗浄され、乾
燥される。DNAは、25 /It(DB S A(5

アーゼ的活性を含んでいるので、DNAのどちらの鎖・
も加水分解される。7μtサンプルがそれぞれ、1分3
0秒から4分30秒まで、30秒間隔で取シ出され、T
E43μtおよび反応停止溶液5μt6含むバイアルへ
加えられる。Ba131切断の範囲は、アガロースゲル
(0,75%ゲル)でサンプルを10μを処理すること
によシ決定される。
Ba131切断使った消化によシ約IKbだけ短かくさ
れたと思われるDNAサンプルは、回転透析を2サイク
ル受ける。l0XRB1の最終量の0およびX h o
l (15unit /1i7)1μtが加えられ、
その混合物は、37℃で8時間培養される。Xhoiは
、ヌクレオチド2672でDNAを切断し、2つの断片
を生ずる。反応停止溶液5μtが加えられ、おのおのの
時間点からDNAの1が、0.75%アガロースゲル上
の2つの近接するくほみのおのおのへ加えられる。電気
泳動け、54ボルトで一晩行なわれる。DNAは、臭化
エチジウム染色により視覚化され、1.7Kb連結物は
、ゲルから切り離され、電気溶離(0,IX電極緩衝液
中で、108ボルトで3時間)される。臭化エチジウム
は、TEで飽和されたnブタノールの等量(約3 ml
) で3回抽出することにより除去される。クロロホ
ルム:3メヂルブタノール(24:1)の等量を使って
2回、抽出が行なわれ、DNAは、エタノール沈殿され
、エタノールを使って洗浄され、乾燥される。
DNAは、TE50μを中に再浮遊され、4μ9 のp
trcsベクターDNA (予め、王maiと5ail
とを使った連続消化にさらされた)と同じように、回転
透析を2サイクル受ける。ベクターDNAd、エタノー
ル沈殿され、洗浄され、48μtのSmal緩衝液(ト

おのおのの時間点から得られるH8V−2DNA断片(
約2tty) は、0.2,1(7)plJC8,、D
NAと混ぜられる。10Xリカーゼ緩衝液の1ん量、1

および3μAのT4DNAリガーセ(4×1o5uni
ts / ml ) が加えられ、全反応液量が、50
μtになる結紮は、室温で一晩行なわれる。反応混合物
は、実施例1cの技術にょ9、E、coAi RDP2
11を形質転換するだめに、直ちに使用される。
fB−βガラクトシダーゼ融合タンパク質の同定および
免疫的活性
各個′))細菌コロニーは、適当なH8V−2挿入物を
伴なうプラスミド(p52ΔNXとと呼ばれる部類の)
を挿入することにより、およびl PTG (イソプロ
ピル−β−D−チオカラクトシト)の存在において生長
する含有゛′吻体を挿入すると々によシ得られる。これ
らコロニーからの培養物(10ml ) は、約1のO
Dへ、誘導状態(50%エタノール中に56 mf /
ml の1. P T G 4μt)または、非誘導
状態(20%(V/V )グルコース浴液150μt)
において、■メジューム中で生長される。細胞は、ペレ
ット化され、(60007×5分)、] Omy/ m
t のリソチーム、4μ9 / mtの膵臓デオキシリ
ボヌクレアーゼ■(DNasel)を含む溶解緩衝液(

に加えられる。最初のペレット中に存在する膜タンパク
質もまだ、電気泳動サンプル溶液100μを中に可溶化
される。
12%アクリルアミドゲル(分析用ゲル)は、ゲル緩衝
液(pl+ 8.8の0.375Mトリス。
HCl、0.1%5DS)の100 ml 中にアクリ
ルアミド(12ym ) とN、N’−ジアリル酒石酸
ジアミド(0,56℃m) を溶かすことによシ作られ
る。0.6 mA の過硫酸アンモニウム溶液(水9
mAに対して17m)および30μtのN、 N、 N
’N’−テトラメチルエチレンジアミンは、重合過程を
触媒するために、90m1 に対して加えられる。溶液
は、吸引のもとで、ガスを除かれ、4E、mt が、お
のおのの分離ゲルを形成するために使用される。2枚の
ガラス板(+ 5.9 X i 9cm)は、そのおの
おのの側面において1.2 mmスペーサーにより、お
よび、板の底部で節3のスペーサーにより間隔をあけら
れ、その2枚のガラス板の間に、溶液は注がれる。完全
な集合物は、クランプ(clamp )によりきさえら
れたプラスチックゲルスタンドへ同時に保持され、緑は
、アカロース(水100 mtに対して1.62m)で
密閉される。プラスチック・ブランク(blank )
(幅1Bcm、長さ’;!、、 5 cm +厚さ1
、2 in )は、ゲルの上表面でなめらかな表面を作
るために挿入される。ゲルが重合(]時間)された後、
ブランクは、取シ除かれ、15mL の堆積ゲル(5%

トリス、Hα、0.1%SDSにおける同じ方法で使ら
れた)は、分離ゲルの上表面に注がれる。10の歯を持
つコウム(comb ) (おのおの幅85胴、×長さ
15.2 ram )が、堆積ゲルへ添えられる。
堆積ゲルの重合(1時間)の後、電気泳動1ナンプル溶
液甲のタンパク質サンプル(5μLから10μt)は、
コウムの歯によって作られたおのおのくほみへ加えられ
る。ゲルは、トリス−グリシン電極緩衝液(0025M
トリス。
0.192Mグリシン、01%S D S 、 pHは
8.5)中で4時間30分、108ボルトで電気泳動さ
れる。電気泳動の後、ゲル板は分離され、ゲル中のタン
パク質は、染色されるが、収い収られる。
タンパク質を視覚化するために、コマイセブリリアント
ブルー(Coomarsse Br1lliantBl
ue ) 染色液(0,25%染色液1 (1,2+X
氷酢酸、45.4xytタノ−/L)u、30分間ゲル
へ加えられ、それから、メタノール−酢ば一水(5−7
,5−87,5)中で一晩脱色される。
抗体反応に使用されるタンパク質は、染色されず、12
タイポリアクリルアミド−8DSゲルにおける電気泳動
を使用し−で分離した後ニトロセルロース紙上に吸い取
られる。電気吸い取り器が使われ、吸い敗りは、メーカ
ーより提供された指示により行なわれる。収い取シは、
20X(V/V)メタノールを含む電極緩衝液(192
mMグリシン、25mMトリス−HC/!、I XS
D S 、 pH8,3KA製すれた)中で、6oボル
ト1時間30分行なわれる。
タンパク質がニトロセルロースへ移動すれた後、抗体結
合測定のだめに直径100mmのプラスチックペトリ皿
