BACKGROUND OF THE INVENTION Various devices are known for progressively delivering anthelmintic agents or trace elements over an extended period of time into the ruminal pouch of a ruminant animal. An example of such a device is described in British patent specification no.
6CD No. 907. In the device of this patent, a brecast cylindrical plug containing a therapeutic mixture of anthelmintics is housed within a tubular body and urged by a spring toward a diaphragm υ opening at one end. The end of the plug that is in contact with the diaphragm opening disintegrates when it comes into contact with the fluid in the regastric region. One disadvantage of this arrangement is that the opening may be obstructed by grass or other objects within the rumen, preventing the mixture from being delivered at a consistent rate. Other problems with this device include a poor seal between the plug and the tubular body, which allows fluid to enter, resulting in irregular disassembly of the plug and a constant supply of dewormer. It lies in not doing it.

The device contains the therapeutic mixture within a weighted tube, the ends of which are closed in sections by porous members impregnated with Idrodel material. When the device is placed in the regastric region, the therapeutic mixture is progressively administered through the porous end piece. Because the dosing of such mixtures is by osmosis, the rate of dosing gradually decreases as the amount of mixture in the device is reduced in use. This reduction in dosing rate is a disadvantage when it is necessary to administer the mixture to the animal in a fixed ratio.

Also, even in the case of the collapse of the known device mentioned above, administration continues more or less continuously in the trenches where the therapeutic mixture is consumed. However, when administering anthelmintic drugs in this manner, if for some reason the amount of anthelmintic administered is insufficient to kill all the wheezes, subsequent outbreaks of worms may Tolerance may develop.

(Another Means to Solve the Problems) The medicinal substance developed by the present inventors is particularly suitable for use as an anticoagulant internal administration device, although it is not exclusive. Each of the various embodiments of this medicament overcomes one or more of the drawbacks noted in both previously mentioned devices. For intragastric use, such devices can be configured to provide continuous or periodic dosing (or both simultaneously) as desired. This drug can also be used for other purposes. Applications include, for example, the administration of therapeutic agents, nutrients and other drugs to humans and animals. It can also be used to administer drugs such as insecticides, aquatic plants, herbicides, fish nutrients, vaccines, etc. to rivers, reservoirs, lakes, ponds, pools (including swimming pools), tanks, etc.

If a plurality of inner segments are provided, these inner segments can be made of the same or different materials. However, as mentioned earlier, each of the kidnapping fees is expendable. Two or more inner segments may also be spaced apart from the inert flange member extending inwardly from the inert outer segment. The inner segment may also serve to hold the drug together, for example by perching on a central rod as described in more detail below. In some embodiments, more types of inner segments spanning two ties 7° may be provided, each made of a different consumable material. Each of the consumable materials contains at least one biologically active ingredient, although it need not be the same biologically active ingredient as each other.

Alternatively, one material may contain biologically active components, for example, and the other material may be a consumable material containing substantially biologically inactive components. You can also do that. In general, if two types of inner segments are provided, the two types of segments can also be alternated within the core. for example,
If one type of inner segment contains a substantially biologically inert component, such segment is compatible with another type of inner segment, i.e., an inner segment containing a biologically active component. It functions as a spacer between. This configuration is particularly useful when periodic drug administration is desired. The inner segments made of the same material can also be connected to each other by circumferential ridges. This circumferential ridge extends longitudinally inside the casing formed by the outer segment and is preferably integral with the inner segment.

In embodiments where a single or integral inner segment is provided, this inner segment naturally constitutes the entire core tl. In embodiments where the liquid-tight casing does not cover either end of the drug body, the interior is designed to provide alternating delivery of one or more biologically active ingredients from each end during use. You can also place segments. For example, if the first ingredient is (hypothetically) 21
The second component can be fed from one end after a period of 21 days, and then the second component can be fed from the opposite end after a further 21 days, or this can be done alternately. Of course, the same or one or more different biologically active components can also be fed simultaneously from both ends in this configuration.

In one preferred form, the or each inner segment is generally cylindrical or
It is disc-shaped. If there is more than one inner segment, the inner segments may be held together by a fit between an annular projection on each disc and a recess in an adjacent disc. Alternatively, the disc can also be held within an annular body forming the outer segment. The drug bodies are held together by securing the outer segments together as described in more detail below. In some embodiments, the or each inner segment includes a central hole or opening through which the central rod passes. A fit may also be provided between the segment and the rod to hold at least a portion of the drug substance together. The lock has a suitably tapered or stepped diameter as required to facilitate engagement with the segment. Adhesives or other securing compounds may also be used to help hold the inner segment on the rod. Alternatively, the flange members can project inwardly from the outer segments as described above. These flange members are located within the circumferential groove of the rod and are capable of holding the drug substances together.

The outer segment can be associated with the inner segment, or can be attached to an individual or pair of inner segments. Accordingly, the outer segments may be configured to abut each other to form a seal between the outer segments. This seal can be formed by the fabric and elasticity of the material. The outer segment is made from such materials. For example, they are made of elastic plastic materials such as polyvinyl chloride, or natural or synthetic rubber. Examples of synthetic rubber include silicone, imprene, styrene, butadiene, and EPM.
There are phthyl, chloroprene, nitrile, polysulfide, polyurethane, chlorosulfonyl, polyethylene, fluoro, acrylic, shrimp chlorohydrin, polypropylene rubber, or norbornene rubber.

For example, the outer segments overlap each other to hold the drug together.

In another form, particularly but not exclusively intended for administration to preanimated animals, the present invention provides a drug body comprised of a plurality of axially spaced material parts. The parts do not corrode within the pharyngeal pouch, forming voids or depressions between them. The biologically active substance in the cavity or recess of the tube and the means made of a material that does not corrode in the ruminant pouch are arranged in such a way that one end of the means is exposed. is supported. As a result, in use, as the means corrodes away, the parts are successively disengaged from the assembly of the means, each releasing a dose of biologically active substance. .

When used as a sea bream internal administration device, spitting out of the drug according to the present invention can be prevented by adding an appropriate geometric structure or by adding an appropriate weight. Said geometrical structure may be described, for example, in British Patent Specification No. 1606970.
It is equipped with wings as described in the issue. In general, the K to ensure that the drug remains in the stomach is 2.25 to 3.
.. 5 & /d, preferably 2.5.9/II
It must have an overall density of L/. Preferably,
This density should increase as the drug body is progressively depleted. Depending on the materials that make up the drug body, it is possible to easily obtain the required weight, but in this case,
When a certain amount of the drug is consumed in the stomach,
The density of the drug decreases below the minimum density necessary to retain it, and the device becomes expelled. Therefore, it is desirable to provide a separate weight to the drug body.

The weight may be constructed from a suitable material, yet still provide the required overall density of drug. For example, iron, steel, copper, tin, lead, tungsten, zinc, chromium, cobalt, nickel, magnesium, or alloys consisting of one or more of these metals, or one or more of the aforementioned metals. Heavy metals can also be made from alloys consisting of one or more other metals. Particularly preferred are steel or die-cast alloys of iron or zinc. These active metal weights often remain in the reciprocal sac even after the rest of the drug body has been exhausted. Apart from this, substantially consumable materials;
For example, a consumable zinc alloy can also be used. The heavy metal may, if desired, consist of a matrix material, for example with metallic spheres of iron or other metals mixed within an active primary material (i.e., a polymer) or a consumable primary material (i.e., a magnesium alloy). can. If the main material is active, the sea bream will generally remain in the ruminal sea bream gastric pouch while the main material is exhausted. After the weight is exhausted, the metal ball is small enough to be excreted. The weight can also include particulate material such as iron balls of suitable density. The particulate material is contained within a retaining sleeve made of plastic, for example, or a consumable shell made of magnesium alloy, for example. The sleeve itself is
Surrounded by a plastic or other active sleeve. Particulate materials can also be mixed into the core of the device. In general, the weight (in any form) can be placed at one end of the drug body, at any position on the drug body, and/or as a central rod as described above. In the latter case, the rod is preferably constructed of active material.

At least a portion of the rod or core is made of a different metal or alloy of metals than that of the weight. Therefore, if both the reed and the rod or core are exposed to a liquid environment, and the liquid environment can act as an electrolyte, a battery effect occurs. This phenomenon commonly occurs when the drug substance is used in the presacral sac of an animal or in contaminated water such as a river or lake. Battery effects determine the corrosion rate of the rod or core. If you want to strengthen the battery effect, you can attach a suitable metal different from the reed to the reed. The weight can also be provided with inserts of different suitable metals or metal alloys.

To be used orally in the rumen, the drug according to the present invention includes:
It has a weight and dimensions suitable for the particular ruminant to be used. Although the weights and dimensions of the parts required for a given application will be obvious to those skilled in the art (e.g. with knowledge of the density requirements noted above) - by way of example, a drug to be administered to cattle: 60 to 200111 m, for example 50 to 15011 m, typically about 90 IiL. It still has a diameter of 15 to 40 m, for example 20 to 30 m, typically about 25 m. The number of doses used is 2 to 10, typically 5.
A dose of biologically active ingredient can be administered over a period of 1 to 12 months, such as 6 to 6 months, typically about 3 and a half months. Each dose is 10■ to 10y1 e.g. 100
m9 to 1y1 typically consists of 750~. J. L. Riner et al. showed that the influence of the liquid concentration in the ruminal chain pouch and the position of ingestion on the consumption of the pre-administered drug was determined by J. L. Riner et al.

It is made of consumable metals such as aluminum, zinc, titanium, and alloys thereof, or of alloys of magnesium with one or more other metals, among others. Other metals are selected, for example, from the following (all percentages are by weight): up to 1.5% silicon, up to 1% nickel, up to 15% zinc (e.g. about 12%) , up to 2% manganese, up to 4% copper (e.g. about 2% or 6%),
There is up to 15% aluminum (eg about 12%) and up to 0.8% zorconium. It may also contain traces of cobalt and/or selenium. It is obvious to those skilled in the art that not all of the aforementioned metals are interchangeable. For example, it is not normally possible to include both aluminum and zorconium in an alloy. Also, manganese is incompatible with copper and aluminum. Thus, for example, if aluminum is added in the alloy, it cannot contain more than 0.6% manganese.

As previously mentioned, the amount of biologically active components as trace elements to be administered to animals can remain stable and can be used alone or in combination with other metals if selected appropriately. It can be consumed in the fluid of the rumen as an alloy with metal. Additionally, the metal formulation can be varied to allow for the incorporation of one or more biologically active agents. However, most other biologically active mixtures used alone or in combination require them to be manufactured as powders that are compressed. If necessary, it is shaped into a tablet using a binder, for example.

In particular, the therapeutic agent can also be formed into separate units such as capsules, cashews or tablets. Each unit contains a predetermined amount of powder or granular drug. Such formulations may be obtained by dispensing or may involve a step of holding the drug together by one or more additional ingredients binder and/or carrier. Such methods are generally carried out by uniformly kneading the drug using a liquid or finely divided solid carrier, or both, and then, if necessary, shaping the finished product to suit the desired use. be done. For example, tablets can be manufactured by compressing or molding a powder or lees powder of the drug, adding one or more additional ingredients as necessary. Compressed tablets are made by compressing the drug in a free-flowing state, such as powder or granules, in a suitable machine, with the addition of binders, lubricants, inert diluents, surfactants or dispersants as required. It can also be obtained by Molded tablets may be made by molding the powdered medicament moistened with an inert liquid diluent in a suitable machine. Alternatively, the formulation of such tablets may be carried out by incorporating one or more conventional solid carriers, such as cocoa butter, into the medicament and shaping the resulting mixture.

5 is formed. In the drug bodies according to FIGS. 1 to 4, unless otherwise stated, the magnesium alloy in the examples described below is a magnesium alloy containing 12% aluminum and 2% steel in weight percent. A tablet 17 is accommodated in each recess 15, respectively. This tablet has anthelmintic medicinal properties with biologically active substances, preferably oxfendazole (
oxfendazole) or levamisole (le
vamisole). These tablets should have a second type of inner segment IF#*. Tablet 17 and magnesium alloy segment 3.5.
7.9 and 11 constitute the core of drug substance 1. To ensure that the drug remains in the small stomach during use,
A steel end weight 19 is provided. This end weight is made of magnesium alloy segments 3, 5, 7, 9
.. It is attached to the drug substance 1 by an annular protrusion 21 similar to the protrusion 13 at 11. The inner segments of the magnesium alloy housing are each surrounded by an outer segment 23 of silicone rubber with a U-shaped cross section. The dimensions of these outer segments 23 are such that when the drug substance 1 is assembled as shown, the outer segments 23 are compressed;
The joints 25 between the segments are traps so that they are effectively sealed. The outer segments 23 thus form a liquid barrier for the drug substance.

R7 in segment 3 of the magnesium alloy does not contain a deworming tablet, but if necessary, place such a tablet in recess 1γ and immediately add the first dose of dewormer when administering drug substance 1. It can also be done. This first batch of tablets is made by crimping the annular protrusion of the first segment 3, indicated by reference numeral 28.
) 'L, or bend the edge of the protrusion 28 appropriately,
Alternatively, the recess 27 can be held by covering it with a suitable magnesium alloy cap (not shown), for example. Apart from these, such first batch tablets can also be fixed using a suitable adhesive. However, if there is no tablet in the first recess 27 as shown, the first tablet 17 will be exposed when the wear progresses sufficiently. As a result, the animal receives its first dose of dewormer. Once the disks of almost all segments 3 have been worn out, there is no longer any support for the outermost segment, indicated by reference numeral 29. Therefore, this segment may become dislodged and eventually be excreted or
This segment is expelled due to extremely low pressure.

Alternatively, in this embodiment, the components of the drug body 1 are fixed to the central rod 103 equivalent to the bar using an interlocking screw structure, for example if said central rod is in the form of a stud. You can also do that. However, it is not necessary to provide threads along the entire length of this rod. In this example, opposing threads are provided in holes 105 and 115. However, if the stud is provided with a head and is configured to protrude from the end weight 11γ at the end of the end weight 11γ spaced from the segment 107, i.e. if the stud is a bolt, then the end weight 117
There is no need to provide a hole 115 for screwing. Similarly,
In the embodiment of FIGS. 8 and 9, if the shank 65 or 85 is provided with a head projecting from one end of the drug substance, the magnesium alloy inner segment 62.8
The hole in the center of 9 does not need to be threaded. In the latter two examples (FIGS. 8 and 9), instead of having a head on the shank 65 or 85, if the shank is a bolt, a magnesium alloy inner segment 62 at the tip is used.
Alternatively, screws could be attached to holes 63 or 9 that pass through 89.

FIG. 14 shows drug body 149 with a single inner segment 151 core. The inner segment comprises a matrix of material such as wax, corrodible or biologically degradable polymer, bulk of compacted metal particles, or paris paste. A therapeutic agent, such as an anthelmintic, is incorporated within this matrix. This matrix is surrounded by a plurality of outer segments 153 having the same structure and equipment as the embodiment shown in FIGS. 6 and 7. This special drug allows the anthelmintic to be administered continuously rather than at intervals. In the illustrated device, no end weights are provided so that the outer segment is expelled or evacuated from the rumen after most of the device has been consumed. However, an additional weight (not shown), such as a central red, or at the end or length of the drug body, may be used to maintain intragastric pressure until all of the anthelmintic has been administered. It is also possible to attach weights or the like consisting of suitable structures arranged along the line.

The technique for securing the magnesium alloy inner segment 157 and outer segment 161 to each other can be improved as shown in FIG. Here, a circumferential ridge 167 on the inner surface of the outer segment 161 is located in an opposing circumferential groove 169 in the magnesium alloy inner segment 15γhc. Relative movements are thus prevented. As previously mentioned, the dewormer tablet segment 159 is sandwiched between 1!HC of the magnesium alloy inner segment.
A seal between adjacent outer segments of the sleeve is obtained by compressing these outer segments. These seals also take advantage of the properties of the material that makes up the outer segment. However, other inert materials that do not need to form a suitable seal upon compression may also be used.
